Claims
- 1. A unimolecular multi-arm block copolymer useful as a drug delivery vehicle, comprising:
a central core molecule providing at least three attachment sites available for covalent attachment to a copolymer arm; and a copolymer arm covalently attached to each of said at least three attachment sites of said central core molecule, each copolymer arm comprising an inner polypeptide segment covalently attached to said attachment site of said central core molecule and an outer hydrophilic polymer segment covalently attached to said polypeptide segment.
- 2. The unimolecular multi-arm block copolymer of claim 1, wherein said central core molecule is a residue of a polyamine having at least three termini bearing an amine group.
- 3. The unimolecular multi-arm block copolymer of claim 1, wherein said central core molecule comprises a residue of a polyol having at least three termini bearing a hydroxyl group.
- 4. The unimolecular multi-arm block copolymer of claim 3, wherein the polyol is selected from the group consisting of glycerol, sorbitol, pentaerythritol, and glycerol oligomers.
- 5. The unimolecular multi-arm block copolymer of claim 3, wherein the polyol is hexaglycerol.
- 6. The unimolecular multi-arm block copolymer of claim 3, wherein the polyol is hydroxypropyl-β-cyclodextrin.
- 7. The unimolecular multi-arm block copolymer of claim 3, wherein said central core molecule further comprises a hydrophilic oligomer chain attached to each termini of said residue of a polyol.
- 8. The unimolecular multi-arm block copolymer of claim 7, wherein said hydrophilic oligomer chain is an ethylene glycol oligomer chain having a molecular weight of about 100 Da to about 5,000 Da.
- 9. The unimolecular multi-arm block copolymer of claim 7, wherein each hydrophilic oligomer chain is covalently attached to said polypeptide segment through an amide linkage.
- 10. The unimolecular multi-arm block copolymer of claim 1, wherein said polypeptide segment comprises residues of amino acids selected from the group consisting of glycine, alanine, valine, leucine, isoleucine, methionine, proline, phenylalanine, tryptophan, serine, threonine, asparagine, glutamine, tyrosine, cysteine, lysine, arginine, histidine, aspartic acid, glutamic acid, and combinations thereof.
- 11. The unimolecular multi-arm block copolymer of claim 1, wherein said polypeptide segment comprises residues of charged amino acids.
- 12. The unimolecular multi-arm block copolymer of claim 11, wherein the charged amino acids are selected from the group consisting of arginine, lysine, histidine, glutamic acid, aspartic acid, and combinations thereof.
- 13. The unimolecular multi-arm block copolymer of claim 1, wherein said polypeptide segment comprises at least one amino acid residue bearing a functional group.
- 14. The unimolecular multi-arm block copolymer of claim 13, wherein the functional group is selected from the group consisting of hydroxyl, active ester, active carbonate, acetal, aldehyde, aldehyde hydrate, alkyl or aryl sulfonate, halide, disulfide, alkenyl, acrylate, methacrylate, acrylamide, active sulfone, amine, hydrazide, thiol, carboxylic acid, isocyanate, isothiocyanate, maleimide, vinylsulfone, dithiopyridine, vinylpyridine, iodoacetamide, epoxide, glyoxal, dione, mesylate, tosylate, and tresylate.
- 15. The unimolecular multi-arm block copolymer of claim 1, wherein the outer hydrophilic polymer segment comprises poly(ethylene glycol).
- 16. The unimolecular multi-arm block copolymer of claim 1, wherein each polypeptide segment has a molecular weight of about 100 Da to about 20,000 Da.
- 17. The unimolecular multi-arm block copolymer of claim 16, wherein each polypeptide segment has a molecular weight of about 500 Da to about 10,000 Da.
- 18. The unimolecular multi-arm block copolymer of claim 1, wherein the outer hydrophilic polymer segment has a molecular weight of about 100 Da to about 20,000 Da.
- 19. The unimolecular multi-arm block copolymer of claim 18, wherein the outer hydrophilic polymer segment has a molecular weight of about 500 Da to about 10,000 Da.
- 20. The unimolecular multi-arm block copolymer of claim 1, wherein the unimolecular multi-arm block copolymer has a total molecular weight of about 10,000 Da to about 200,000 Da.
- 21. The unimolecular multi-arm block copolymer of claim 20, wherein the unimolecular multi-arm block copolymer has a total molecular weight of about 20,000 Da to about 80,000 Da.
- 22. The unimolecular multi-arm block copolymer of claim 1, wherein said central core molecule has at least four attachment sites available for covalent attachment to a copolymer arm.
- 23. The unimolecular multi-arm block copolymer of claim 1, wherein said central core molecule has at least eight attachment sites available for covalent attachment to a copolymer arm.
- 24. The unimolecular multi-arm block copolymer of claim 1, wherein said central core molecule has 3 to about 25 attachment sites available for covalent attachment to a copolymer arm.
- 25. The unimolecular multi-arm block copolymer of claim 1, wherein at least one targeting moiety is covalently attached to at least one hydrophilic polymer segment.
- 26. The unimolecular multi-arm block copolymer of claim 25, wherein the targeting moiety is selected from the group consisting of a protein, an antibody, an antibody fragment, a peptide, a carbohydrate, a lipid, an oligonucleotide, DNA, RNA, and a small molecule having molecular weight less than 2,000 Da.
- 27. The unimolecular multi-arm block copolymer of claim 25, wherein the targeting moiety is mono-folic acid or anti-EGFr Fab.
- 28. The unimolecular multi-arm block copolymer of claim 1, wherein at least one capping group or functional group is covalently attached to at least one hydrophilic polymer segment.
- 29. The unimolecular multi-arm block copolymer of claim 28, wherein the capping group or functional group is selected from the group consisting of alkoxy, hydroxyl, active ester, active carbonate, acetal, aldehyde, aldehyde hydrate, alkyl or aryl sulfonate, halide, disulfide, alkenyl, acrylate, methacrylate, acrylamide, active sulfone, amine, hydrazide, thiol, carboxylic acid, isocyanate, isothiocyanate, maleimide, vinylsulfone, dithiopyridine, vinylpyridine, iodoacetamide, epoxide, glyoxal, dione, mesylate, tosylate, or tresylate.
- 30. The unimolecular multi-arm block copolymer of claim 1, having the structure:

- 32. The unimolecular multi-arm block copolymer of claim 30, wherein each E is hydroxy.

The unimolecular multi-arm block copolymer of claim 30, wherein each E is selected from the group consisting of alkoxy, hydroxyl, active ester, active carbonate, acetal, aldehyde, aldehyde hydrate, alkyl or aryl sulfonate, halide, disulfide, alkenyl, acrylate, methacrylate, acrylamide, active sulfone, amine, hydrazide, thiol, carboxylic acid, isocyanate, isothiocyanate, maleimide, vinylsulfone, dithiopyridine, vinylpyridine, iodoacetamide, epoxide, glyoxal, dione, mesylate, tosylate, or tresylate.
- 34. The unimolecular multi-arm block copolymer of claim 30, wherein A is a residue of a polyol selected from the group consisting of glycerol, sorbitol, pentaerythritol, hydroxypropyl-β-cyclodextrin, and glycerol oligomers.
- 35. The unimolecular multi-arm block copolymer of claim 30, wherein D comprises poly(ethylene glycol).
- 36. The unimolecular multi-arm block copolymer of claim 30, wherein C comprises residues of amino acids selected from the group consisting of glycine, alanine, valine, leucine, isoleucine, methionine, proline, phenylalanine, tryptophan, serine, threonine, asparagine, glutamine, tyrosine, cysteine, lysine, arginine, histidine, aspartic acid, glutamic acid, and combinations thereof.
- 37. The unimolecular multi-arm block copolymer of claim 30, wherein at least one amino acid residue of C bears a functional group.
- 38. The unimolecular multi-arm block copolymer of claim 30, wherein B is an ethylene glycol oligomer.
- 39. The unimolecular multi-arm block copolymer of claim 1, having the structure:
- 40. A pharmaceutical composition, comprising:
a unimolecular multi-arm block copolymer comprising a central core molecule providing at least three attachment sites available for covalent attachment to a copolymer arm, and a copolymer arm covalently attached to each of said at least three attachment sites of said central core molecule, each copolymer arm comprising an inner polypeptide segment covalently attached to said attachment site of said central core molecule and an outer hydrophilic polymer segment covalently attached to said polypeptide segment, wherein the central core molecule and the polypeptide segments define an inner core region and the polypeptide segments define a hydrophobic outer region; and a biologically active agent entrapped within the inner region of the unimolecular multi-arm block copolymer.
- 41. The pharmaceutical composition according to claim 40, wherein the polypeptide segment comprises one or more charged amino acid residues, and wherein the biologically active agent entrapped within the inner core region bears a charge opposite of the charge of the one or more charged amino acid residues.
- 42. The pharmaceutical composition according to claim 40, wherein the biologically active agent is covalently attached to an amino acid residue of the polypeptide segment.
- 43. The pharmaceutical composition according to claim 40, wherein both the inner core region and the biologically active agent are hydrophobic.
- 44. The pharmaceutical composition according to claim 40, further comprising a pharmaceutically acceptable carrier.
