Claims
- 1. A nucleic acid molecule comprising a heterologous nematode-resistance sequence operably linked to a promoter that drives expression of said heterologous nematode-resistance sequence in a plant cell, wherein said promoter comprises a nucleotide sequence selected from the group consisting of:
(a) a nucleotide sequence comprising the sequence set forth in SEQ ID NO:3; (b) a nucleotide sequence having at least 95% sequence identity to the sequence of SEQ ID NO:3, wherein said nucleotide sequence has nematode-regulated promoter activity; and (c) a nucleotide sequence deposited as Patent Deposit No. PTA-4028.
- 2. A vector comprising the nucleic acid molecule of claim 1.
- 3. A plant cell having stably incorporated into its genome the nucleic acid molecule of claim 1.
- 4. The plant cell of claim 3, wherein said plant cell is from a dicot plant.
- 5. The plant cell of claim 4, wherein said dicot plant is soybean.
- 6. A method of modulating the expression of a nucleotide sequence of interest, said method comprising stably incorporating in the genome of a plant cell said nucleotide sequence of interest operably linked to a promoter, wherein said promoter is selected from the group consisting of:
(a) a nucleotide sequence comprising the sequence set forth in SEQ ID NO:3; (b) a nucleotide sequence having at least 95% sequence identity to the sequence of SEQ ID NO:3, wherein said nucleotide sequence has nematode-regulated promoter activity; and (c) a nucleotide sequence deposited as Patent Deposit No. PTA-4028.
- 7. A nucleic acid molecule comprising a first nucleotide sequence comprising a heterologous nematode-resistance sequence operably linked to a first promoter capable of inducing transcription of said heterologous nematode-resistance sequence in a plant cell and a second nucleotide sequence comprising a sequence capable of inhibiting said heterologous nematode-resistance sequence operably linked to a second promoter capable of repressing transcription of said inhibitor of said heterologous nematode-resistance sequence in a plant cell, wherein said first promoter comprises a nucleotide sequence selected from the group consisting of:
(a) a nucleotide sequence comprising the sequence set forth in SEQ ID NO:3; (b) a nucleotide sequence having at least 95% sequence identity to the sequence of SEQ ID NO:3, wherein said nucleotide sequence has nematode-regulated promoter activity; and (c) a nucleotide sequence deposited as Patent Deposit No. PTA-4028.
- 8. An expression cassette comprising the nucleic acid molecule of claim 7.
- 9. A plant cell having stably incorporated in its genome the nucleic acid molecule of claim 7.
- 10. The plant cell of claim 9, wherein said plant cell is from a dicot plant.
- 11. The plant cell of claim 10, wherein said dicot plant is soybean.
- 12. A plant stably transformed with a nucleic acid molecule comprising a heterologous nematode-resistance sequence operably linked to a promoter that induces transcription of said nematode-resistance sequence in a plant cell in response to a nematode stimulus, wherein said promoter comprises a nucleotide sequence selected from the group consisting of:
(a) a nucleotide sequence comprising the sequence set forth in SEQ ID NO:3; (b) a nucleotide sequence having at least 95% sequence identity to the sequence of SEQ ID NO:3, wherein said nucleotide sequence has nematode-regulated promoter activity; and (c) a nucleotide sequence deposited as Patent Deposit No. PTA-4028.
- 13. The plant of claim 12, wherein said plant is a dicot.
- 14. The plant of claim 13, wherein said dicot is soybean.
- 15. Transformed seed of the plant of claim 12, wherein the seed comprise the nucleic acid molecule.
- 16. A method for conferring or improving nematode resistance in a plant, said method comprising transforming said plant with a nucleic acid molecule comprising a heterologous sequence operably linked to a promoter that induces transcription of said heterologous sequence in a plant cell in response to a nematode stimulus and regenerating stably transformed plants, wherein said promoter comprises a nucleotide sequence selected from the group consisting of:
(a) a nucleotide sequence comprising the sequence set forth in SEQ ID NO:3; (b) a nucleotide sequence having at least 95% sequence identity to the sequence of SEQ ID NO:3, wherein said nucleotide sequence has nematode-regulated promoter activity; and (c) a nucleotide sequence deposited as Patent Deposit No. PTA-4028.
- 17. The method of claim 16, wherein said plant is a dicot.
- 18. The method of claim 17, wherein said dicot is soybean.
- 19. An isolated polypeptide, the expression of which is induced in response to nematode infection, selected from the group consisting of:
(a) a polypeptide comprising the amino acid sequence set forth in SEQ ID NO:2; (b) a polypeptide encoded by a nucleotide sequence comprising the sequence set forth in SEQ ID NO:1; (c) a polypeptide encoded by a nucleotide sequence comprising the coding region of the sequence set forth in SEQ ID NO:5; (d) a polypeptide comprising an amino acid sequence encoded by a nucleotide sequence deposited as Patent Deposit No. PTA-4028; (e) a polypeptide encoded by a nucleotide sequence that has at least 80% sequence identity to the sequence set forth in SEQ ID NO:1, wherein expression of said polypeptide is induced in response to nematode infection; (f) a polypeptide encoded by a nucleotide sequence that has at least 80% sequence identity to the coding region of the nucleotide sequence set forth in SEQ ID NO:5, wherein expression of said polypeptide is induced in response to nematode infection; and (g) a polypeptide comprising an amino acid sequence having at least 80% sequence identity to the sequence set forth in SEQ ID NO:2, wherein expression of said polypeptide is induced in response to nematode infection.
- 20. An isolated nucleic acid molecule that encodes a polypeptide, the expression of which is induced in response to nematode infection, said nucleic acid molecule comprising a nucleotide sequence selected from the group consisting of:
(a) a nucleic acid molecule comprising the sequence set forth in SEQ ID NO:1; (b) a nucleic acid molecule comprising the coding region of the sequence set forth in SEQ ID NO:5; (c) a nucleic acid molecule comprising a sequence encoding the amino acid sequence set forth in SEQ ID NO:2; (d) a nucleic acid molecule comprising a sequence deposited as Patent Deposit No. PTA-4028; (e) a nucleic acid molecule comprising the full length complement of the sequence of (a), (b), (c), or (d); (f) a nucleic acid molecule comprising a sequence having at least 80% sequence identity to the nucleotide sequence set forth in SEQ ID NO:1, wherein said nucleic acid molecule encodes a polypeptide, the expression of which is induced in response to nematode infection; (g) a nucleic acid molecule comprising a sequence having at least 80% sequence identity to the coding region of the nucleotide sequence set forth in SEQ ID NO:5, wherein said nucleic acid molecule encodes a polypeptide, the expression of which is induced in response to nematode infection; and (h) A nucleic acid molecule encoding a polypeptide wherein said nucleic acid molecule encodes a polypeptide with at least 80% sequence identity to SEQ ID NO: 2.
- 21. A nucleotide construct comprising a nucleic acid molecule of claim 20, wherein said nucleic acid molecule is operably linked to a promoter that drives expression in a host cell.
- 22. A method for modulating the expression of a peptide of interest comprising transforming a host cell with the nucleic acid molecule of claim 20.
- 23. A cell having stably incorporated into its genome at least one nucleotide construct comprising a nucleic acid molecule operably linked to a heterologous promoter that drives expression in said cell, wherein said nucleic acid molecule encodes a polypeptide, the expression of which is induced in response to nematode infection; and wherein said nucleic acid molecule is selected from the group consisting of:
(a) a nucleic acid molecule comprising the sequence set forth in SEQ ID NO: 1; (b) a nucleic acid molecule comprising the coding sequence of the sequence set forth in SEQ ID NO:5; (c) a nucleic acid molecule comprising a sequence encoding the amino acid sequence set forth in SEQ ID NO:2; (d) a nucleic acid molecule comprising a sequence deposited as Patent Deposit No. PTA-4028; (e) a nucleic acid molecule comprising a full length complement of the sequence of (a), (b), (c), or (d); (f) a nucleic acid molecule comprising a sequence having at least 90% sequence identity to the nucleotide sequence set forth in SEQ ID NO:1, wherein said nucleic acid molecule encodes a polypeptide, the expression of which is induced in response to nematode infection; (g) a nucleic acid molecule comprising a sequence having at least 90% sequence identity to the coding region of the nucleotide sequence set forth in SEQ ID NO:5, wherein said nucleic acid molecule encodes a polypeptide, the expression of which is induced in response to nematode infection; and (h) A nucleic acid molecule encoding a polypeptide wherein said nucleic acid molecule encodes a polypeptide with at least 90% sequence identity to SEQ ID NO: 2.
- 24. The cell of claim 23, wherein said cell is a plant cell.
- 25. A transformed plant having stably incorporated into its genome at least one nucleotide construct comprising a nucleic acid molecule operably linked to a heterologous promoter that drives expression in a plant cell, wherein said nucleic acid molecule encodes a polypeptide, the expression of which is induced in response to nematode infection; and wherein said nucleic acid molecule is selected from the group consisting of:
(a) a nucleic acid molecule comprising the sequence set forth in SEQ ID NO: 1; (b) a nucleic acid molecule comprising the coding region of the sequence set forth in SEQ ID NO:5; (c) a nucleic acid molecule comprising a sequence encoding the amino acid sequence set forth in SEQ ID NO:2; (d) a nucleic acid molecule comprising a sequence deposited as Patent Deposit No. PTA-4028; (e) a nucleic acid molecule comprising a full length complement of the sequence of (a), (b), (c), or (d); (f) a nucleic acid molecule comprising a sequence having at least 90% sequence identity to the nucleotide sequence set forth in SEQ ID NO:1, wherein said nucleic acid molecule encodes a polypeptide, the expression of which is induced in response to nematode infection; (g) a nucleic acid molecule comprising a sequence having at least 90% sequence identity to the coding region of the nucleotide sequence set forth in SEQ ID NO:5, wherein said nucleic acid molecule encodes a polypeptide, the expression of which is induced in response to nematode infection; and (h) A nucleic acid molecule encoding a polypeptide wherein said nucleic acid molecule encodes a polypeptide with at least 90% sequence identity to SEQ ID NO: 2.
- 26. The plant of claim 25, wherein said promoter is selected from the group consisting of:
a) a constitutive promoter; b) a tissue-preferred promoter; and c) an inducible promoter.
- 27. The plant of claim 25, wherein said plant is a monocot.
- 28. The plant of claim 27, wherein said monocot is selected from the group consisting of maize, wheat, rice, barley, sorghum, and rye.
- 29. The plant of claim 25, wherein said plant is a dicot.
- 30. Transformed seed of the plant of claim 25, wherein the seed comprise the nucleotide construct.
- 31. A nucleotide construct comprising a promoter, wherein said promoter confers the ability to regulate transcription and comprises a nucleic acid molecule selected from the group consisting of:
(a) a nucleic acid molecule comprising the sequence set forth in SEQ ID NO:3; (b) a nucleic acid molecule comprising the sequence deposited as Patent Deposit No. PTA-4028; and (c) a nucleic acid molecule comprising a nucleotide sequence having at least 90% sequence identity to the sequence set forth in SEQ ID NO:3.
- 32. An expression vector comprising the nucleotide construct of claim 31.
- 33. A method for conferring or improving nematode resistance of a plant, said method comprising stably introducing into the genome of a plant at least one nucleotide construct comprising a nucleic acid molecule operably linked to a heterologous promoter that drives expression in a plant cell, wherein said nucleic acid molecule encodes a polypeptide, the expression of which is induced in response to nematode infection; and wherein said nucleic acid molecule is selected from the group consisting of:
(a) a nucleic acid molecule comprising the sequence set forth in SEQ ID NO: 1; (b) a nucleic acid molecule comprising the coding region of the sequence set forth in SEQ ID NO:5; (c) a nucleic acid molecule comprising a sequence encoding the amino acid sequence set forth in SEQ ID NO:2; (d) a nucleic acid molecule comprising a sequence deposited as Patent Deposit No. PTA-4028; (e) a nucleic acid molecule comprising an antisense sequence corresponding to a sequence of (a), (b), (c), or (d); (f) a nucleic acid molecule comprising a sequence having at least 90% sequence identity to the nucleotide sequence set forth in SEQ ID NO:1, wherein said nucleic acid molecule encodes a polypeptide, the expression of which is induced in response to nematode infection; (g) a nucleic acid molecule comprising a sequence having at least 90% sequence identity to the coding region of the nucleotide sequence set forth in SEQ ID NO:5, wherein said nucleic acid molecule encodes a polypeptide, the expression of which is induced in response to nematode infection; and (h) A nucleic acid molecule encoding a polypeptide wherein said nucleic acid molecule encodes a polypeptide with at least 90% sequence identity to SEQ ID NO: 2.
- 34. A nucleic acid molecule comprising a heterologous nematode-resistance sequence operably linked to a promoter that drives expression of said heterologous nematode-resistance sequence in a plant cell, wherein said promoter comprises the nucleotide sequence set forth in SEQ ID NO:3.
