Claims
- 1. An isolated polynucleotide comprising a polynucleotide selected from:
(a) a polynucleotide encoding a B2 or B4 beta expansin polypeptide of the sequences shown in SEQ ID NOS: land 3; (b) a polynucleotide which encodes the polypeptide sequence shown in SEQ ID NOS: 2 and 4, (c) a polynucleotide which selectively hybridizes to a polynucleotide of (a) or (b); (d) a polynucleotide having at least 50% sequence identity with polynucleotides of (a) or (b); (e) complementary sequences of polynucleotides of (a), (b), or (c); and (f) a polynucleotide comprising at least 25 contiguous nucleotides from a polynucleotide of (a), (b), (c), or (d).
- 2. A recombinant expression cassette comprising a polynucleotide selected from the group consisting of:
(a) a polynucleotide encoding a B2 or B4 beta expansin polypeptide of the sequences shown in SEQ ID NOS: land 3; (b) a polynucleotide which encodes the polypeptide sequence shown in SEQ ID NOS: 2 and 4, (c) a polynucleotide which selectively hybridizes to a polynucleotide of (a) or (b); (d) a polynucleotide having at least 50% sequence identity with polynucleotides of (a) or (b); (e) complementary sequences of polynucleotides of (a), (b), or (c); and (f) a polynucleotide comprising at least 25 contiguous nucleotides from a polynucleotide of (a), (b), (c), or (d).
- 3. A vector comprising a recombinant expression cassette comprising a polynucleotide selected from the group consisting of:
(a) a polynucleotide encoding a B2 or B4 beta expansin polypeptide of the sequences shown in SEQ ID NOS: land 3; (b) a polynucleotide which encodes the polypeptide sequence shown in SEQ ID NOS: 2 and 4, (c) a polynucleotide which selectively hybridizes to a polynucleotide of (a) or (b); (d) a polynucleotide having at least 50% sequence identity with polynucleotides of (a) or (b); (e) complementary sequences of polynucleotides of (a), (b), or (c); and (f) a polynucleotide comprising at least 25 contiguous nucleotides from a polynucleotide of (a), (b), (c), or (d).
- 4. A host cell comprising a recombinant expression cassette comprising a polynucleotide selected from the group consisting of:.
(a) a polynucleotide encoding a B2 or B4 beta expansin polypeptide of the sequences shown in SEQ ID NOS: land 3; (b) a polynucleotide which encodes the polypeptide sequence shown in SEQ ID NOS: 2 and 4, (c) a polynucleotide which selectively hybridizes to a polynucleotide of (a) or (b); (d) a polynucleotide having at least 50% sequence identity with polynucleotides of (a) or (b); (e) complementary sequences of polynucleotides of (a), (b), or (c); and (f) a polynucleotide comprising at least 25 contiguous nucleotides from a polynucleotide of (a), (b), (c), or (d).
- 5. The host cell of claim 4 wherein the cell is a plant cell.
- 6. The host cell of claim 5 wherein the cell is selected from the group consisting of maize, sorghum, wheat, tomato, soybean, alfalfa, sunflower, canola, cotton, and rice.
- 7. A transformed plant comprising a polynucleotide selected from the group consisting of:
(a) a polynucleotide encoding a B2 or B4 beta expansin polypeptide of the sequences shown in SEQ ID NOS: 1 and 3; (b) a polynucleotide which encodes the polypeptide sequence shown in SEQ ID NOS: 2 and 4, (c) a polynucleotide which selectively hybridizes to a polynucleotide of (a) or (b); (d) a polynucleotide having at least 50% sequence identity with polynucleotides of (a) or (b); (e) complementary sequences of polynucleotides of (a), (b), or (c); and (f) a polynucleotide comprising at least 25 contiguous nucleotides from a polynucleotide of (a), (b), (c), or (d).
- 8. A plant seed comprising a polynucleotide selected from the group consisting of:
(a) a polynucleotide encoding a B2 or B4 beta expansin polypeptide of the sequences shown in SEQ ID NOS: land 3; (b) a polynucleotide which encodes the polypeptide sequence shown in SEQ ID NOS: 2 and 4, (c) a polynucleotide which selectively hybridizes to a polynucleotide of (a) or (b); (d) a polynucleotide having at least 50% sequence identity with polynucleotides of (a) or (b); (e) complementary sequences of polynucleotides of (a), (b), or (c); and (f) a polynucleotide comprising at least 25 contiguous nucleotides from a polynucleotide of (a), (b), (c), or (d).
- 9. A method of altering cell wall characteristics in a plant comprising:
A) transforming a plant cell with a vector comprising a polynucleotide selected from the group consisting of:
(a) a polynucleotide encoding a B2 or B4 beta expansin polypeptide of the sequences shown in SEQ ID NOS: land 3; (b) a polynucleotide which encodes the polypeptide sequence shown in SEQ ID NOS: 2 and 4, (c) a polynucleotide which selectively hybridizes to a polynucleotide of (a) or (b); (d) a polynucleotide having at least 50% sequence identity with polynucleotides of (a) or (b); (e) complementary sequences of polynucleotides of (a), (b), or (c); and i. (f) a polynucleotide comprising at least 25 contiguous nucleotides from a polynucleotide of (a), (b), (c), or (d); B) growing the plant cell under plant growing conditions; and C) providing for the expression of said polynucleotide.
- 10. A method of making a beta expansin protein comprising the steps of:
A) expressing a polynucleotide in a recombinantly engineered cell, wherein the polynucleotide is selected from the group consisting of:
(a) a polynucleotide encoding a B2 or B4 beta expansin polypeptide of the sequences shown in SEQ ID NOS: land 3; (b) a polynucleotide which encodes the polypeptide sequence shown in SEQ ID NOS: 2 and 4, (c) a polynucleotide which selectively hybridizes to a polynucleotide of (a) or (b); (d) a polynucleotide having at least 50% sequence identity with polynucleotides of (a) or (b); (e) a polynucleotide comprising at least 25 contiguous nucleotides from a polynucleotide of (a), (b), (c), or (d); (f) a polynucleotide complementary to (a-e); said polynucleotide operably linked to a promoter; and B) purifying the enzyme.
- 11. A method of making an beta expansin comprising the steps of:
A) expressing a polynucleotide in a plant, wherein said polynucleotide is selected from the group consisting of:
(a) a polynucleotide encoding a B2 or B4 beta expansin polypeptide of the sequences shown in SEQ ID NOS: 1 and 3; (b) a polynucleotide which encodes the polypeptide sequence shown in SEQ ID NOS: 2 and 4, (c) a polynucleotide which selectively hybridizes to a polynucleotide of (a) or (b); (d) a polynucleotide having at least 50% sequence identity with polynucleotides of (a) or (b); (e) complementary sequences of polynucleotides of (a), (b), or (c); and i. (f) a polynucleotide comprising at least 25 contiguous nucleotides from a polynucleotide of (a), (b), (c), or (d).of i. through iii. operably linked to a promoter; and B) purifying the enzyme from the plant seed or other plant parts.
- 12. An isolated polynucleotide comprising a polynucleotide selected from:
a) a polynucleotide which hybridizes under high stringency conditions to a polynucleotide selected from SEQ ID NOS: 1 or 3; b) a polynucleotide comprising at least 90% identity to a polynucleotide selected from SEQ ID NOS: 1 or 3; and c) a polynucleotide complementary to a polynucleotide of (a) through (b).
- 13. A recombinant expression cassette comprising a polynucleotide selected from the group consisting of:
a) a polynucleotide which hybridizes under high stringency conditions to a polynucleotide selected from land 3; b) a polynucleotide comprising at least 90% identity to a polynucleotide selected from SEQ ID NOS: land 3; and c) a polynucleotide complementary to a polynucleotide of (a) through (b).
- 14. A vector comprising a recombinant expression cassette comprising a polynucleotide selected from the group consisting of:
a) a polynucleotide which hybridizes under high stringency conditions to a polynucleotide selected from SEQ ID NOS: 1 and 3; b) a polynucleotide comprising at least 90% identity to a polynucleotide selected from SEQ ID NOS: 1 and 3; and c) a polynucleotide complementary to a polynucleotide of (a) through (b).
- 15. A host cell comprising a recombinant expression cassette comprising a polynucleotide selected from the group consisting of:
a) a polynucleotide which hybridizes under high stringency conditions to a polynucleotide selected from 1 and 3; b) a polynucleotide comprising at least 90% identity to a polynucleotide selected from SEQ ID NOS: 1 and 3; and c) a polynucleotide complementary to a polynucleotide of (a) through (b).
- 16. The host cell of claim 15 wherein the cell is a plant cell.
- 17. The host cell of claim 16 wherein the cell is selected from the group consisting of maize, sorghum, wheat, tomato, soybean, alfalfa, sunflower, canola, cotton, and rice.
- 18. A transformed plant comprising a polynucleotide selected from the group consisting of:
a) a polynucleotide which hybridizes under high stringency conditions to a polynucleotide selected from SEQ ID NOS: 1 and 3; b) a polynucleotide comprising at least 90% identity to a polynucleotide selected from SEQ ID NOS: 1 and 3; and c) a polynucleotide complementary to a polynucleotide of (a) through (b).
- 19. A plant seed comprising a polynucleotide selected from the group consisting of:
a) a polynucleotide which hybridizes under high stringency conditions to a polynucleotide selected from SEQ ID NOS: land 3; b) a polynucleotide comprising at least 90% identity to a polynucleotide selected from SEQ ID NOS: 1 and 3; and c) a polynucleotide complementary to a polynucleotide of (a) through (b).
- 20. An isolated polypeptide having expansin-like activity and selected from the group consisting of
(a) a polypeptide comprising the amino acid sequence set forth in SEQ ID NO: 2 or 4; (b) a polypeptide encoded by a nucleotide sequence comprising the sequence set forth in SEQ ID NO: 1 or 3; (d) a polypeptide encoded by a nucleotide sequence that has at least 70% sequence identity to the sequence set forth in SEQ ID NO: 1 or 3; (e) a polypeptide comprising an amino acid sequence having at least 50% sequence identity to the sequence set forth in SEQ ID NO: 2, or 4; and (f) a polypeptide comprising an amino acid sequence of at least 30 consecutive amino acids of any of (a) through (e).
- 21. A polynucleotide encoding the polypeptide of claim 20.
