Claims
- 1. A nucleic acid labeling compound of the following structure:
- 2. A nucleic acid labeling compound according to claim 1 wherein A is selected from the group consisting of H, α-thiotriphosphate and H4O9P3—; X is O; Y is H or OH; Z is H or OR10, wherein R10 is H, alkyl or aryl.
- 3. A nucleic acid labeling compound according to claim 2 wherein Q is selected from the group consisting of colloidal gold, a fluorescent group and biotin.
- 4. A nucleic acid labeling compound according to claim 3 wherein Q is a biotin.
- 5. A nucleic acid labeling compound according to claim 3 having a first M and a second M, wherein said first M is —NH(CH2)nNH—, wherein n is an integer from about 2 to about 10, and a second M is —CO(CH2)pNH—, wherein p is an integer from about 2 to about 10 and m is 2.
- 6. A nucleic acid labeling compound according to claim 5 wherein said first M is —NH(CH2)2NH— and said second M is —CO(CH2)5NH—.
- 7. A nucleic acid labeling compound according to claim 1 wherein A is a triphosphate group having appropriate counterions and Y is OH and Z is OH.
- 8. A nucleic acid labeling compound according to claim 7 wherein said counterions are selected from the group consisting of H+, Na+, Li+, K+, (CH3CH2)3NH+ and NH4+.
- 9. A nucleic acid derivative produced by coupling a nucleic acid labeling compound according to claim 1 with a nucleic acid.
- 10. A hybridization product comprising the nucleic acid derivative according to claim 9 bound to a complementary probe.
- 11. A hybridization product according to claim 10, wherein the probe is attached to a glass chip.
- 12. A method of synthesizing a labeled nucleic acid comprising attaching a nucleic acid labeling compound according to claim 1 to a nucleic acid.
- 13. A method of detecting a nucleic acid comprising incubating a nucleic acid derivative according to claim 9 with a complementary probe.
- 14. A method according to claim 13, wherein the probe is attached to a solid support.
- 15. A method according to claim 14, wherein said solid support is glass.
- 16. A nucleic acid labeling compound of the following structure:
- 17. A nucleic acid labeling compound according to claim 16 wherein A is selected from the group consisting of H, α-thiotriphosphate and H4O9P3—; X is O; Y is H or OH; Z is H or OR10, wherein R10 is H, alkyl or aryl.
- 18. A nucleic acid labeling compound according to claim 16 wherein Q is selected from the group consisting of colloidal gold, a fluorescent group and biotin.
- 19. A nucleic acid labeling compound according to claim 18 wherein Q is a biotin.
- 20. A nucleic acid labeling compound according to claim 17 having a first M and a second M, wherein said first M is —NH(CH2)nNH—, wherein n is an integer from about 2 to about 10, and a second M is —CO(CH2)pNH—, wherein p is an integer from about 2 to about 10 and m is 2.
- 21. A nucleic acid labeling compound according to claim 20 wherein said first M is —NH(CH2)2NH— and said second M is —CO(CH2)5NH—.
- 22. A nucleic acid labeling compound according to claim 16 wherein A is a triphosphate group having appropriate counterions.
- 23. A nucleic acid labeling compound according to claim 22 wherein said counterion is selected from the group consisting of H+, Na+, Li+, K+, (CH3CH2)3NH+ and NH4+.
- 24. A nucleic acid derivative produced by coupling a nucleic acid labeling compound according to claim 16 with a nucleic acid.
- 25. A hybridization product comprising the nucleic acid derivative according to claim 24 bound to a complementary probe.
- 26. A hybridization product according to claim 25, wherein the probe is attached to a solid support.
- 27. A hybridization product according to claim 26, wherein said solid support is glass.
- 28. A method of detecting a nucleic acid comprising incubating a nucleic acid derivative according to claim 24 with a complementary probe.
- 29. A method according to claim 28, wherein the probe is attached to a solid support.
- 30. A method according to claim 29, wherein said solid support is glass.

- 32. A nucleic acid labeling compound according to claim 1 wherein L is —CH═CH—R11, wherein R11 comprises a moiety selected from the group consisting of alkyl, functionalized alkyl, alkenyl alkyl, alkynal alkyl, amido alkyl, amino alkyl, alkoxy, amino, aryl, and thio.
- 33. A nucleic acid labeling compound according to claim 32 where R11 is C(O)R12, where R12 comprises a moiety selected from the group consisting of a bond, alkyl, functionalized alkyl, alkenyl alkyl, alkynal alkyl, amido alkyl, amino alkyl, alkoxy, amino, aryl, and thio.
- 34. A nucleic acid labeling according to claim 31 wherein A is selected from the group consisting of H, α-thiotriphosphate, and H4O9P3—; X is O; Y is H or OH; Z is H or OR10, wherein R10 is H, alkyl or aryl.
- 35. A nucleic acid labeling compound according to claim 31 wherein Q is selected from the group consisting of colloidal gold, a fluorescent group and biotin.
- 36. A nucleic acid labeling compound according to claim 33, wherein R12 is a bond, having a first M and a second M, wherein said first M is —NH(CH2)nNH—, wherein n is an integer from about 2 to about 10, and a second M is —CO(CH2)pNH—, wherein p is an integer from about 2 to about 10 and m is 2.
- 37. A nucleic acid labeling compound according to claim 36 wherein said first M is —NH(CH2)2NH— and said second M is —CO(CH2)5NH— and m is 2, having the structure:
- 38. A nucleic acid labeling compound according to claim 31 wherein A is a triphosphate group having appropriate counterions.
- 39. A nucleic acid labeling compound according to claim 38 wherein said appropriate counterion is selected from the group consisting of H+, Na+, Li+, K+, (CH3)3NH+ and NH4+.
- 40. A nucleic acid derivative produced by coupling a nucleic acid labeling compound according to claim 31 with a nucleic acid.
- 41. A hybridization product comprising the nucleic acid derivative according to claim 40 bound to a complementary probe.
- 42. A hybridization product according to claim 41, wherein the complementary probe is attached to a solid support.
- 43. A hybridization product according to claim 42, wherein said solid support is glass.

- 46. A nucleic acid labeling compound according to claim 41 where R11 is C(O)R12, where R12 comprises a moiety selected from the group consisting of a bond, alkyl, functionalized alkyl, alkenyl alkyl, alkynal alkyl, amido alkyl, amino alkyl, alkoxy, amino, aryl, and thio.
- 47. A nucleic acid labeling compound according to claim 42 wherein A is selected from the group consisting of H, α-thiotriphosphate, and H4O9P3—; X is O; Y is H or OH; Z is H or OR10, wherein R10 is H, alkyl or aryl.
- 48. A nucleic acid labeling compound according to claim 43 wherein Q is selected from the group consisting of colloidal gold, a fluorescent group and biotin.
- 49. A nucleic acid labeling compound according to claim 46, wherein R12 is a bond, having a first M and a second M wherein said first M is —NH(CH2)nNH—, wherein n is an integer from about 2 to about 10, and a second M is —CO(CH2)pNH—, wherein p is an integer from about 2 to about 10 and m is 2.
- 50. A nucleic acid labeling compound according to claim 45 wherein said first M is —NH(CH2)2NH— and said second M is —CO(CH2)5NH— and m is 2 having the structure
- 51. A nucleic acid labeling compound according to claim 44 wherein A is a triphosphate group having an appropriate counterion.
- 52. A nucleic acid labeling compound according to claim 47 wherein said appropriate counterion is selected from the group consisting of H+, Na+, Li+, K+, (CH3)3NH+ and NH4+.
- 53. A nucleic acid derivative produced by coupling a nucleic acid labeling compound according to claim 44 with a nucleic acid.
- 54. A hybridization product comprising the nucleic acid derivative according to claim 53 bound to a complementary probe.
- 55. The hybridization product according to claim 54, wherein the complementary probe is attached to a solid support.
- 56. The hybridization product according to claim 55 wherein said solid support is glass.
