Claims
- 1. An isolated nucleic acid sequence comprising a nucleic acid sequence which encodes a polypeptide having haloperoxidase activity, wherein the polypeptide is selected from the group consisting of:
(a) a polypeptide having an amino acid sequence which has at least 80% homology with the amino acid sequence of SEQ ID NO:2; (b) a polypeptide which is encoded by a nucleic acid sequence which hybridizes under medium stringency conditions with (i) the nucleotide sequence of SEQ ID NO:1, (ii) a subsequence of (i) of at least 100 nucleotides, or (iii) a complementary strand of (i) or (ii); (c) a variant of the polypeptide having an amino acid sequence of SEQ ID NO:2 comprising a substitution, deletion, and/or insertion of one or more amino acids; (d) an allelic variant of (a) or (b); (e) a fragment of (a), (b), or (d) that has haloperoxidase activity; and (f) a polypeptide having more than 50% residual activity after 15 minutes incubation at 70° C. and pH 7.
- 2. The nucleic acid sequence of claim 1, wherein the amino acid sequence of the polypeptide has at least 95% homology with the amino acid sequence of SEQ ID NO:2.
- 3. The nucleic acid sequence of claim 2, wherein the polypeptide comprises the amino acid sequence of SEQ ID NO:2.
- 4. The nucleic acid sequence of claim 1, wherein the polypeptide consists of the amino acid sequence of SEQ ID NO:2 or a fragment thereof.
- 5. The nucleic acid sequence of claim 4, wherein the polypeptide consists of the amino acid sequence of SEQ ID NO:2.
- 6. The nucleic acid sequence of claim 1, wherein the polypeptide is a variant of the polypeptide having an amino acid sequence of SEQ ID NO:2 comprising a substitution, deletion, and/or insertion of one or more amino acids.
- 7. The nucleic acid sequence of claim 1, which is contained in the plasmid contained in E. coli DH10B, deposited as DSM 13442.
- 8. An isolated nucleic acid sequence comprising a nucleic acid sequence having at least one mutation in the polypeptide coding sequence of SEQ ID NO:1, in which the mutant nucleic acid sequence encodes a polypeptide consisting of the amino acid sequence of SEQ ID NO:2.
- 9. A nucleic acid construct comprising the nucleic acid sequence of any of claims 1-8 operably linked to one or more control sequences that direct the production of the polypeptide in a suitable expression host.
- 10. A recombinant expression vector comprising the nucleic acid construct of claim 9.
- 11. A recombinant host cell comprising the nucleic acid construct of claim 9.
- 12. A method for producing a mutant nucleic acid sequence, comprising (a) introducing at least one mutation into the polypeptide coding sequence of SEQ ID NO:1, wherein the mutant nucleic acid sequence encodes a polypeptide consisting of the amino acid sequence of SEQ ID NO:2; and (b) recovering the mutant nucleic acid sequence.
- 13. A mutant nucleic acid sequence produced by the method of claim 12.
- 14. A method for producing a polypeptide, comprising (a) cultivating a strain comprising the mutant nucleic acid sequence of claim 13 encoding the polypeptide to produce a supernatant comprising the polypeptide; and (b) recovering the polypeptide.
- 15. A method for producing the polypeptide comprising (a) cultivating a host cell comprising a nucleic acid construct comprising a nucleic acid sequence of any of claims 1-8 encoding the polypeptide under conditions suitable for production of the polypeptide; and (b) recovering the polypeptide.
- 16. A method for producing a polypeptide comprising (a) cultivating a host cell under conditions conducive for production of the polypeptide, wherein the host cell comprises a mutant nucleic acid sequence having at least one mutation in the polypeptide coding sequence of SEQ ID NO:1, wherein the mutant nucleic acid sequence encodes a polypeptide consisting of the amino acid sequence of SEQ ID NO:2, and (b) recovering the polypeptide.
- 17. A method for shuffling of DNA comprising using the nucleotide sequence of SEQ ID NO: 1.
- 18. A polynucleotide encoding a polypeptide having haloperoxidase activity obtained by the method of claim 17.
- 19. A polypeptide having haloperoxidase activity encoded by the polynucleotide of claim 18.
