Claims
- 1. A compound of the formula ##STR53## wherein R.sub.1 is hydrogen, alkyl, alkanoyl, aroyl or aralkyl; R.sub.2 and R.sub.3, independently, are hydrogen, alkyl, cycloalkyl, alkenyl, acyl, aryl or aralkyl; and R.sub.4 is hydrogen, alkyl, hydroxyalkyl, phenyl-hydroxyalkyl, halophenyl-hydroxyalkyl, alkylphenyl-hydroxyalkyl, alkoxyphenyl-hydroxyalkyl, alkoxyalkyl, aryloxy-hydroxyalkyl, alkoxy-hydroxyalkyl, acyloxyalkyl, arylcarbonylalkyl, alkoxycarbonylalkyl, aralkyl, alkenyl, alkylcycloalkyl, alkynyl, thienyl-alkyl, furyl-alkyl, arylcarboxamidoalkyl, acylalkyl, cyclic-alkyloxoalkyl, cyclic-alkylhydroxyalkyl, alkenyloxyalkyl, aralkenyl, aryloxyalkyl, N-alkyl-pyrrolidinylalkyl, trifluoroalkyl of 2 to 6 carbon atoms, aryl-N-imidazolonylalkyl, or ##STR54## wherein R.sub.6 and R.sub.7, independently, are hydrogen or alkyl, or taken together with the nitrogen form a morpholine, N-methyl-piperazino, piperazino or pyrrolidino ring, and X is O or S,
- an optical isomer, a geometric isomer, or a pharmaceutically acceptable acid addition salt thereof.
- 2. A compound in accordance with claim 1, wherein X is O.
- 3. A compound in accordance with claim 2, which is the trans isomer.
- 4. A compound in accordance with claim 3 in which R.sub.1 is hydrogen, R.sub.2 and R.sub.3 are alkyl, and R.sub.4 is alkyl, hydroxyalkyl, phenyl-hydroxyalkyl, aralkyl, alkoxyalkyl, aryloxyalkyl, arylcarbonylalkyl, or alkyl-cycloalkyl.
- 5. A compound in accordance with claim 4 in which R.sub.4 is alkyl.
- 6. A compound in accordance with claim 4, wherein R.sub.4 is phenyl hydroxyalkyl.
- 7. A compound in accordance with claim 4, wherein R.sub.4 is aralkyl.
- 8. A compound in accordance with claim 4, wherein R.sub.4 is alkoxyalkyl.
- 9. A compound in accordance with claim 4, wherein R.sub.4 is aryloxyalkyl.
- 10. A compound in accordance with claim 4, wherein R.sub.4 is arylcarbonylalkyl.
- 11. A compound in accordance with claim 4, wherein R.sub.4 is alkyl-cycloalkyl.
- 12. A compound in accordance with claim 4, wherein R.sub.4 is hydroxyalkyl.
- 13. A compound in accordance with claim 5, 3-ethyl-2,6-dimethyl-4,4a,5,6,7,8,8a,9-octahydro-4a,8a-trans-1H-pyrrolo[2,3-g]isoquinolin-4-one.
- 14. A compound in accordance with claim 5, 2,3,6-trimethyl-4,4a,5,6,7,8,8a,9-octahydro-4a,8a-trans-1H-pyrrolo[2,3-g]isoquinolin-4-one.
- 15. A compound in accordance with claim 13, (-)-3-ethyl-2,6-dimethyl-4,4a,5,6,7,8,8a,9-octahydro-4a,8a-trans-1H-pyrrolo[2,3-g]isoquinolin-4-one.
- 16. A compound in accordance with claim 6, 2-methyl-3-ethyl-6-(2-hydroxy-2-phenylethyl)-4,4a,5,6,7,8,8a,9-octahydro-4a,8a-trans-1H-pyrrolo[2,3-g]isoquinolin-4-one.
- 17. A compound in accordance with claim 7, 2-methyl-3-ethyl-6-(2-phenylethyl)-4,4a,5,6,7,8,8a,9-octahydro-4a,8a-trans-1H-pyrrolo[2,3-g]isoquinolin-4-one.
- 18. A compound in accordance with claim 7, 2-methyl-3-ethyl-6-benzyl-4,4a,5,6,7,8,8a,9-octahydro-4a,8a-trans-1H-pyrrolo[2,3-g]isoquinolin-4-one.
- 19. A compound in accordance with claim 8, 2-methyl-3-ethyl-6-(2-ethoxyethyl)-4,4a,5,6,7,8,8a,9-octahydro-4a,8a-trans-1H-pyrrolo[2,3-g]isoquinolin-4-one.
- 20. A compound in accordance with claim 9, 2-methyl-3-ethyl-6-(3-phenoxypropyl)-4,4a,5,6,7,8,8a,9-octahydro-4a,8a-trans-1H-pyrrolo[2,3-g]isoquinolin-4-one.
- 21. A compound in accordance with claim 10, 2-methyl-3-ethyl-6-[4-(4-fluorophenyl)-4-oxobutyl]-4,4a,5,6,7,8,8a,9-octahydro-4a,8a-trans-1H-pyrrolo[2,3-g]isoquinolin-4-one.
- 22. A compound in accordance with claim 10, 2-methyl-3-ethyl-6-[3-(4-fluorophenyl)-3-oxopropyl]-4,4a,5,6,7,8,8a,9-octahydro-4a,8a-trans-1H-pyrrolo[2,3-g]isoquinolin-4-one.
- 23. A compound in accordance with claim 11, 2-methyl-3-ethyl-6-(cyclopropylmethyl)-4,4a,5,6,7,8,8a,9-octahydro-1H-pyrrolo[2,3-g]isoquinolin-4-one.
- 24. A compound in accordance with claim 12, 3-ethyl-2-methyl-6-(2-hydroxy-3-methylbutyl)-4,4a,5,6,7,8,8a,9-octahydro-1H-pyrrolo[2,3-g]isoquinolin-4-one.
- 25. A compound in accordance with claim 12, 3-ethyl-2-methyl-6-(2-hydroxy-3,3-dimethylbutyl)-4,4a,5,6,7,8,8a,9-octahydro-1H-pyrrolo[2,3-g]isoquinolin-4-one.
- 26. A compound in accordance with claim 1 wherein X is S.
- 27. A compound in accordance with claim 26 which is the trans isomer.
- 28. A compound in accordance with claim 27 wherein R.sub.1 is hydrogen, R.sub.2 and R.sub.3 are alkyl, and R.sub.4 is alkyl, aralkyl, or aryl-carbonylalkyl.
- 29. A compound in accordance with claim 28, wherein R.sub.4 is alkyl.
- 30. A compound in accordance with claim 28, wherein R.sub.4 is aralkyl.
- 31. A compound in accordance with claim 28, wherein R.sub.4 is arylcarbonylalkyl.
- 32. A compound in accordance with claim 29, 3-ethyl-2,6-dimethyl-4,4a,5,6,7,8,8a,9-octahydro-4a,8a-trans-1H-pyrrolo[2,3g]isoquinolin-4-thione.
- 33. A compound in accordance with claim 30, 3-ethyl-2-methyl-6-(2-phenylethyl)-4,4a,5,6,7,8,8a,9-octahydro-4a,8a-trans-1H-pyrrolo[2,3-g]isoquinolin-4-thione.
