Claims
- 1. An oxidant scavenger comprising a nitrogen-containing macrocyclic moiety and a cell surface or extracellular matrix targeting moiety, or a pharmaceutically acceptable salt thereof.
- 2. The scavenger according to claim 1 wherein said macrocyclic moiety is a porphyrin.
- 3. The scavenger according to claim 1 wherein said porphyrin is tetrakis (4-benzoic acid) porphyrin.
- 4. An oxidant scavenger of the formula:
- 5. The scavenger according to claim 4 wherein at least one of said R1's, R2's, or R3's is different than the other of said R1's, R2's, or R3's, respectively.
- 6. The scavenger according to claim 1 or claim 4 wherein said scavenger is complexed with a metal selected from the group consisting of manganese, copper and iron.
- 7. The scavenger according to claim 1 wherein said targeting moiety is a sugar or is selected from the group consisting of:

- 8. A method of protecting cells from oxidant-induced toxicity comprising contacting said cells with a non-toxic amount of said scavenger according to claim 1 or a scavenger of the formula,
- 9. The method according to claim 8 wherein said scavenger is complexed with a metal selected from the group consisting of manganese, copper or iron.
- 10. The method according to claim 8 wherein said cells are mammalian cells or plant cells.
- 11. A method of inhibiting damage due to oxidation of a substance comprising contacting to said substance with a non-toxic amount of said scavenger according to claim 1 or a scavenger of the formula,
- 12. The method according to claim 11 wherein said mimetic is complexed with a metal selected from the group consisting of manganese, copper or iron.
- 13. A method of treating a pathological condition of a patient resulting from degradation of NO. or a biologically active form thereof comprising administering to said patient an effective amount of a compound having the activity of a catalytic antioxidant under conditions such that said treatment is effected.
- 14. The method according to claim 13 wherein said biologically active form of NO. is a nitroso, thiol nitrosyl, NO+ or NO− group.
- 15. A method of treating a pathological condition of a patient resulting from peroxynitrite accumulation comprising administering to said patient an effective amount of a compound having the activity of a catalytic antioxidant under conditions such that said treatment is effected.
- 16. The method according to claim 13 or 15 wherein said compound is a porphyrin ring-containing compound.
- 17. A method of treating an inflammatory condition in a patient in need of such treatment comprising administering to said patient an effective amount of a mimetic of SOD, EC-SOD, catalase or peroxidase under conditions such that said treatment is effected.
- 18. The method according to claim 17 wherein a GAG binding moiety is attached to said mimetic.
- 19. A method of treating a disorder resulting from aberrant smooth muscle function in a patient in need of such treatment comprising administering to said patient an effective amount of a mimetic of SOD, EC-SOD, catalase or peroxidase under conditions such that said treatment is effected.
- 20. The method according to claim 19 wherein a GAG binding moiety is attached to said mimetic.
- 21. The method according to claim 17 or 19 wherein said mimetic is a porphyrin ring-containing mimetic.
- 22. A pharmaceutical composition comprising the scavenger according to claim 1, 4, 28 or 41 and a pharmaceutically acceptable carrier.
- 23. A kit comprising the scavenger according to claim 1, 4, 28 or 41 disposed within a container means.
- 24. An isolated EC-SOD gene having the sequence shown in FIG. 24, or portion thereof at least 18 nucleotides in length.
- 25. The gene according to claim 24 wherein said portion corresponds to a noncoding region of said gene.
- 26. A method of inhibiting xanthine oxidase activity of a cell or tissue comprising contacting said cell or tissue with an amount of said scavenger according to claim 1 or a mimetic of the formula,
- 27. The method according to claim 26 wherein said pathological condition is of the lungs of said patient and said administration is to the airways of said patient and said compound has the tissue specificity of extracellular superoxide dismutase.
- 28. An oxidant scavenger of the formula:
- 29. The scavenger according to claim 28 wherein Y′″ is a secondary, tertiary or quaternary amine and each hydrogen replacement group on the amine nitrogen is a C1-C4 alkyl group.
- 30. The scavenger according to claim 28 wherein said scavenger has the structure

- 36. A method of inhibiting damage due to oxidation of a substance comprising contacting to said substance with a non-toxic amount of a scavenger of the formula,
- 37. A method of inhibiting xanthine oxidase activity of a cell or tissue comprising contacting said cell or tissue with an amount of scavenger of the formula,
- 38. A method of modulating NO. function as a neurotransmitter in a mammal comprising contacting said mammal with an amount of an oxidant scavenger of the formula,
- 39. A method of modulating NO. function as a neurotransmitter in a mammal comprising contacting said mammal with an amount of an oxidant scavenger of the formula,

- 43. A method of protecting a biological substance from oxidative stress or a method of maintaining biological function of NO., or active forms thereof, in an intracellular or extracellular space of a tissue or organ comprising contacting said substance or said tissue or organ with an amount of said scavenger according to claim 41 sufficient to effect said protection or said maintenance.
