Claims
- 1. A process for preparing a compound of Formula I: ##STR40## wherein: R.sup.1 is:
- a) OH,
- b) C.sub.1 -C.sub.8 alkyl,
- c) C.sub.1 -C.sub.8 alkoxyl,
- d) C.sub.3 -C.sub.7 cycloalkyl,
- e) aryl,
- f) heteroaryl, or
- g) heterocyclyl;
- C.sub.1 -C.sub.8 alkoxy, C.sub.1 -C.sub.8 alkyl, or C.sub.3 -C.sub.8 cycloalkyl, are unsubstituted or substituted with one, two or three substituents selected from the group consisting of: OH, CO.sub.2 R.sup.4, Br, Cl, F, I, CF.sub.3, N(R.sup.5).sub.2, C.sub.1 -C.sub.8 alkoxy, C.sub.3 -C.sub.8 cycloalkyl, aryl, heteroaryl, heterocyclyl, CO(CH.sub.2).sub.n CH.sub.3, and CO(CH.sub.2).sub.n CH.sub.2 N(R.sup.5).sub.2;
- aryl is defined as phenyl or naphthyl, which is unsubstituted or substituted with one, two or three substituents selected from the group consisting of: OH, CO.sub.2 R.sup.4, Br, Cl, F, I, CF.sub.3, N(R.sup.5).sub.2, C.sub.1 -C.sub.8 alkoxy, C.sub.1 -C.sub.8 alkyl, C.sub.3 -C.sub.8 cycloalkyl, CO(CH.sub.2).sub.n CH.sub.3, and CO(CH.sub.2).sub.n CH.sub.2 N(R.sup.5).sub.2, or when aryl is substituted on adjacent carbons they can form a 5- or 6-membered fused ring having one, two or three heteroatoms selected from O, N, and S, this ring is unsubstituted or substituted on carbon or nitrogen with one, two or three substituents selected from the group consisting of: OH, CO.sub.2 R.sup.4, Br, Cl, F, I, CF.sub.3, N(R.sup.7).sub.2, C.sub.1 -C.sub.8 alkoxy, C.sub.1 -C.sub.8 alkyl, C.sub.3 -C.sub.8 cycloalkyl, CO(CH.sub.2).sub.n CH.sub.3, and CO(CH.sub.2).sub.n CH.sub.2 N(R.sup.5).sub.2;
- heteroaryl is defined as a 5- or 6-membered aromatic ring containing 1, 2 or 3 heteroatoms selected from O, N and S, which is unsubstituted or substituted with one, two or three substituents selected from the group consisting of: OH, CO.sub.2 R.sup.4, Br, Cl, F, I, CF.sub.3, N(R.sup.5).sub.2, C.sub.1 -C.sub.8 alkoxy, C.sub.1 -C.sub.8 alkyl, C.sub.3 -C.sub.8 cycloalkyl, CO(CH.sub.2).sub.n CH.sub.3, and CO(CH.sub.2).sub.n CH.sub.2 N(R.sup.5).sub.2, and additionally the 5- or 6-membered aromatic ring can be benzofused and unsubstituted or substituted with one, two or three substituents as described above;
- heterocyclyl is defined as a 5- or 6-membered, non-aromatic ring containing 1, 2 or 3 heteroatoms selected from O, N and S , which may contain one or two double bonds and which is unsubstituted or substituted with one, two or three substituents selected from the group consisting of: OH, CO.sub.2 R.sup.4, Br, Cl, F, I, CF.sub.3, N(R.sup.5).sub.2, C.sub.1 -C.sub.8 alkoxy, C.sub.1 -C.sub.8 alkyl, C.sub.3 -C.sub.8 cycloalkyl, CO(CH.sub.2).sub.n CH.sub.3, and CO(CH.sub.2).sub.n CH.sub.2 N(R.sup.5).sub.2, and additionally the 5- or 6-membered ring can be benzofused and unsubstituted or substituted with one, two or three substituents as described above;
- R.sup.2 is:
- a) C.sub.1 -C.sub.8 alkyl,
- b) C.sub.3 -C.sub.7 cycloalkyl,
- c) aryl,
- d) heteroaryl, or
- e) heterocyclyl;
- n is: 0 to 5;
- t is: 0, 1 or 2;
- R.sup.4 is: H, or C.sub.1 -C.sub.8 alkyl; or
- R.sup.5 is: H, or C.sub.1 -C.sub.8 alkyl, or aryl;
- R.sup.7 is: H, C.sub.1 -C.sub.8 alkyl, aryl, when two R.sup.7 substituents are on the same nitrogen they can join to form a ring of 3 to 6 atoms;
- comprising reacting a compound of Formula II in a solvent, wherein R.sup.1 and R.sup.2 substituents of Formula II are the same as R.sup.1 and R.sup.2 substituents defined in Formula I except that OH and C.sub.1 -C.sub.8 alkoxyl substituents are not R.sup.1 substituents of Formula II, ##STR41## with a solution of periodic acid, a catalytic amount of a chromium reagent in a solvent at a temperature range of about -20.degree. C. to about 40.degree. C. for about 15 minutes to about 24 hours to oxidize to the compound of Formula I.
- 2. The process as recited in claim 1, wherein the solvent is selected from the group consisting of: acetonitrile, tetrahydrofuran, diethyl ether, MTBE (methyl t-butyl ether), DME (dimethoxyethane), DIGLYME (2-methoxyethyl ether), TRIGLYME (triethylene glycol dimethyl ether), dioxane, and a mixture of said solvents, including a mixture of said solvents with water.
- 3. The process as recited in claim 2, wherein the periodic acid (H.sub.5 IO.sub.6) is utilized in about 2.0 to about 4.0 equivalents.
- 4. The process as recited in claim 3, wherein the chromium reagent is selected from the group consisting of: CrO.sub.3, Na.sub.2 Cr.sub.2 O.sub.7, K.sub.2 Cr.sub.2 O.sub.7, and CrX.sub.3, where X is Cl, Br, F, NO.sub.2, OAc, or ClO.sub.4.
- 5. The process as recited in claim 4, wherein the chromium reagent is utilized in about 0.1 to about 10 mole percent.
- 6. The process as recited in claim 5, wherein the temperature range is about -10.degree. C. to about 30.degree. C.
- 7. The process as recited in claim 6, wherein the periodic acid (H.sub.5 IO.sub.6) is utilized in about 2.5 equivalents.
- 8. The process as recited in claim 7, wherein the chromium trioxide (CrO.sub.3) is utilized in about 1.0 to about 2.0 mole percent.
- 9. The process as recited claim 8, wherein the temperature range is about -10.degree. C. to about 0.degree. C.
- 10. The process as recited in claim 9, wherein the reaction time is about 45 minutes to about 1.5 hours.
