Claims
- 1. A method for creating engineered recombinant plasmids that will be partition proficient during Gram-positive host cell divisions comprising ligating a DNA fragment comprising a Gram-positive partition segment carried on at least one HaeIII - PstI fragment of approximately 260 base pairs isolated from the engineered recombinant Bacillus Subtilis plasmid pOG2381 (ATCC 39038) or the progeny thereof whereby said recombinant plasmids become partition proficient in a Gram-positive bacterial host.
- 2. The method according to claim 1 wherein said DNA fragment comprises a Gram-positive partition segment carried on at least one HindIII - HaeIII fragment of approximately 350 base pairs isolated from the engineered recombinant Bacillus subtilis plasmid pOG2381 (ATCC 39038) or the progeny thereof which are partition proficient in a Gram-positive bacterial host.
- 3. The method according to claim 1 wherein said DNA fragment comprises a Gram-positive partition segment carried on at least one MboI or Sau3A fragment of approximately 1.2 kb derived from the engineered recombinant Bacillus subtilis plasmid pOG2381 (ATTCC 39038) or the progeny thereof which are partition proficient in a Gram-positive bacterial host.
- 4. DNA fragments comprising Gram-positive partition segments carried on HindIII - HaeIII fragments of approximately 350 base pairs isolated from engineered recombinant plasmid pOG2381 (ATCC 39038) or the progeny thereof which are partition proficient in a Gram-positive bacterial host.
- 5. DNA fragments comprising Gram-positive partition segments carried on HaeIII - PstI fragments of approximately 260 base pairs isolated from engineered recombinant plasmid pOG2381 (ATCC 39038) or the progeny thereof which are partition proficient in a Gram-positive bacterial host.
- 6. A method for increasing, in a population of Gram-positive organisms harboring engineered recombinant plasmids which carry a selectable marker, the fraction of said organisms which are designated Bacillus subtilis strain BD224 (ATCC 39038) harboring a partition proficient plasmid, which comprises growing said organisms in the absence of the selection pressure corresponding to said selectable marker.
- 7. Transformable and compatible Gram-positive organisms wherein said organisms are Bacillus subtilis strain BD244 transformed by the partition proficient engineered recombinant plasmid pOG2381 (ATCC 39038) and the progeny thereof which are partition proficient.
- 8. Gram-positive partition proficient engineered recombinant plasmid pOG2381 (ATCC 39038) and the progeny thereof which are partition proficient.
