Claims
- 1. A heat pasteurized, aqueous solution useful for preparation of a hemoglobin blood substitute, said solution comprising conformationally stabilized hemoglobin consisting essentially of tetrameric hemoglobin units which are both stabilized against dissociation into dimeric hemoglobin units, and are also conformationally stabilized against conformational change between the T-conformation and the R-conformation, said tetrameric hemoglobin units containing sub units having globin chains and having secondary amino covalent chemical linkages between the globin chains of the sub units so as to effect the stabilization, the linkages having been formed by reaction of the hemoglobin with a polyaldehyde to form Schiff base linkages, followed by reduction to secondary amino linkages; the hemoglobin having been protected during heat pasteurization by formation of a hemoglobin-carbon monoxide complex; said aqueous solution being free of detectable, transmissible viral infection.
- 2. The solution of claim 1 wherein the tetrameric hemoglobin is stabilized into the T-conformation.
- 3. The solution of claim 2 wherein the stabilized hemoglobin product is formed by reaction of the hemoglobin with a polyaldehyde produced by oxidative ring opening of an oligosaccharide having at least two sugar moieties.
- 4. The solution of claim 3 wherein the polyaldehyde is one produced by oxidative ring opening of an oligosaccharide having up to seven sugar moieties per molecule.
- 5. The solution of claim 4 wherein the oligosaccharide is raffinose.
- 6. A process for preparing the pasteurized, aqueous solution of claim 1, which comprises reacting hemoglobin with a molar excess of carbon monoxide to form CO-hemoglobin;
- stabilizing the hemoglobin against dissociation into dimers and against conformational change between the T-configuration and the R-configuration by reaction with a polyaldehyde to form Schiff base links followed by reduction thereof to secondary amino linkages;
- heating the solution to a temperature of at least 60.degree. C. for a period sufficient for inactivation of viral contaminants therein;
- and reconverting the CO-hemoglobin complex to a stabilized oxy- or deoxyhemoglobin.
- 7. The process of claim 6 wherein the hemoglobin is stabilized in the T-configuration.
- 8. The process of claim 7 wherein the polyaldehyde used to effect stabilization is a ring-opened oligosaccharide.
- 9. The process of claim 8 wherein the polyaldehyde is ring opened raffinose.
- 10. The process of claim 9 wherein the heating to effect pasteurization takes place for a period of at least 10 hours.
- 11. An aqueous solution of a hemoglobin product capable of withstanding temperatures of up to 60.degree. C. for ten hours without substantial denaturation or decomposition to effect pasteurization thereof, the hemoglobin product comprising a carbon monoxylated, stabilized hemoglobin produced by reacting a hemoglobin with a molar excess of carbon monoxide, and by reacting a hemoglobin with a polyaldehyde which is an oxidatively ring opened oligosaccharide, followed by reduction of the Schiff base stabilizing bonds so formed to secondary amino linkages.
