Claims
- 1. A compound represented by formula IIa ##STR63## or a pharmaceutically acceptable salt thereof, in which R.sub.1 represents hydrogen, halo, cyano, a cyano C.sub.1-6 alkyl group, a C.sub.1-12 alkyl group (unsubstituted or substituted by one or more hydroxy groups), a C.sub.1-6 alkoxy group, phenoxy (unsubstituted or substituted), phenyl (unsubstituted or substituted), a C.sub.2-6 alkoxycarbonyl group, an amino group of formula --NR.sub.13 R.sub.14 (in which R.sub.13 and R.sub.14 are independently hydrogen or a C.sub.1-4 alkyl group or R.sub.13 and R.sub.14 together with the nitrogen atom to which they are attached represent a saturated 3-7 membered heterocyclic ring which does or does not further contain sulfur, oxygen or an additional nitrogen atom, wherein the ring is unsubstituted or substituted by one or more C.sub.1-4 alkyl groups), a group of formula --N(R.sub.15)SO.sub.2 R.sub.16 (in which R.sub.15 represents hydrogen or C.sub.1-6 alkyl and R.sub.16 represents hydroxy, a C.sub.1-6 alkyl group or unsubstituted or substituted phenyl), a halogenated C.sub.1-4 alkoxy group, a halogenated C.sub.1-4 alkyl group, arylalkoxy (unsubstituted or substituted), hydroxy, a phenyl C.sub.1-6 alkyl group (unsubstituted or substituted), a (C.sub.2-6 alkoxycarbonyl)vinyl group; a group of formula --S(O).sub.n R.sub.7 (in which R.sub.7 represents a C.sub.1-6 alkyl group and n is 0, 1 or 2 or n is 2 and R.sub.7 represents a group of formula --N(R.sub.17)R.sub.18 in which R.sub.17 and R.sub.18 independently represent hydrogen, a C.sub.1-6 alkyl group or unsubstituted or substituted phenyl), a C.sub.2-6 alkoxycarbonyl C.sub.1-6 alkyl group, a carboxy C.sub.1-6 alkyl group, a carbamoyl group of formula --CONR.sub.11 R.sub.12 (in which R.sub.11 and R.sub.12 are independently hydrogen, a C.sub.1-6 alkyl group, a C.sub.3-6 cycloalkyl group or unsubstituted or substituted phenyl or R.sub.11 and R.sub.12 together with the nitrogen atom to which they are attached represent a saturated 3-7 membered heterocyclic ring which does or does not further contain sulfur, oxygen or an additional nitrogen atom, wherein the the ring is unsubstituted or substituted by one or more C.sub.1-4 alkyl groups), a group of formula --OSO.sub.2 R.sub.21 in which R.sub.21 represents a C.sub.1-6 alkyl group or a phenyl group (unsubstituted or substituted), 4,5-dihydrothiazol-2-yl, 4,4-dimethyl-2-oxazolin-2-yl or a group of formula --NR.sub.60 R.sub.61 in which R.sub.60 represents hydrogen or a C.sub.1-6 alkyl group and R.sub.61 represents a C.sub.1-6 alkanoyl group or benzoyl (unsubstituted or substituted);

- R.sub.19 represents hydrogen, a C.sub.1-6 alkyl group, aryl or an aryl C.sub.1-6 alkyl group.
- 2. A compound according to claim 1 in which R.sub.1 represents hydrogen, halo, a C.sub.1-6 alkyl group (which may be substituted by one or more hydroxy groups), benzyloxy (unsubstituted or substituted), a C.sub.1-4 alkoxy group, a C.sub.1-4 alkylthio group, a C.sub.1-4 perhaloalkyl group, cyano, a C.sub.2-6 alkoxycarbonyl group, 4,5-dihydrothiazol-2-yl, or a group of formula --OSO.sub.2 R.sub.21 in which R.sub.21 represents a C.sub.1-6 alkyl group or a phenyl group (unsubstituted or substituted).
- 3. A compound according to claim 1 in which R.sub.2 represents hydrogen or methyl and R.sub.3 represents hydrogen, a C.sub.1-4 alkyl group, a C.sub.1-4 alkoxy group or halo.
- 4. A compound according to claim 1 in which T represents O or S.
- 5. A compound according to claim 1 in which L.sub.2 represents a C.sub.1-4 alkylene chain unsubstituted or substituted by one or more C.sub.1-4 alkyl groups.
- 6. A compound according to claim 1 in which R.sub.6 represents hydrogen or a C.sub.1-4 alkyl group (unsubstituted or substituted by one or more hydroxy groups or a C.sub.2-6 alkoxycarbonyl group).
- 7. A compound according to claim 1 in which Q represents a C.sub.2-6 alkylene chain unsubstituted or substituted by one or more hydroxy groups.
- 8. A compound according to claim 1 in which Y represents an imidazole group of formula (1) or (2) ##STR66## in which R.sub.8 represents hydrogen or a C.sub.1-4 alkyl group; and R.sub.9 and R.sub.10 each represent hydrogen or a group of formula --L.sub.5 --NR.sub.40 R.sub.41 wherein L.sub.5 represents a C.sub.1-4 alkylene chain (unsubstituted or substituted by one or more alkyl groups);
- R.sub.40 represents hydrogen or a C.sub.1-4 alkyl group and
- R.sub.41 represents a group of formula SO.sub.2 R.sub.42 in which R.sub.42 represents a C.sub.1-6 alkyl group or phenyl (unsubstituted or substituted) or a group of formula COR.sub.43 in which R.sub.43 represents a C.sub.1-6 alkyl group or phenyl (unsubstituted or substituted); and
- R.sub.19 represents hydrogen.
- 9. A compound according to claim 1 in which Y represents a group of formula (1), R.sub.9 represents hydrogen or methyl and R.sub.9 and R.sub.10 independently represent hydrogen or a group of formula --L.sub.5 --NR.sub.40 R.sub.41 in which L.sub.5 represents methylene, ethylene or trimethylene, R.sub.40 represents hydrogen and R.sub.41 represents phenylsulphonyl or benzoyl.

- 11. A pharmaceutical composition comprising a therapeutically effective amount of a compound of formula IIa as defined in claim 1 in combination with a pharmaceutically acceptable diluent or carrier.

- 13. A method of preparing a compound of formula IIa as defined in claim 4 which method comprises:
- a) reacting an imine of formula III ##STR71## in which R.sub.1, R.sub.2, R.sub.3, L.sub.1, T, Q and Y are as previously defined and --L.sub.2 '--C(R.sub.60).dbd. represents a group which on reduction gives L.sub.2, with a reducing agent, in the presence of a solvent for the compound of formula III, at a temperature in the range 0-150.degree. C., at atmospheric pressure; or
- b) condensing a compound of formula IV ##STR72## in which R.sub.1, R.sub.2, R.sub.3, L.sub.1, T, and --L.sub.2 '--C(R.sub.60).dbd. are as previously defined, with a compound of formulaV
- H.sub.2 N--Q--Y V
- by heating the two compounds at a temperature in the range 0-200.degree. C., in the absence or presence of a solvent for the reactants, and then reducing the intermediate obtained directly with a reducing agent, in the presence of a solvent for the reactants, at a temperature in the range 0-150.degree. C., at atmospheric pressure; or
- c) reacting an imine of formula VI ##STR73## in which R.sub.1, R.sub.2, R.sub.3, L.sub.1, T, L.sub.2 and Y are as previously defined and .dbd.C(R.sub.61)--Q'-- represents a group which on reduction gives Q, with a reducing agent, in the presence of a solvent for the compounds of formula VI, at a temperature in the range 0-200.degree. C., at atmospheric pressure; or
- d) condensing a compound of formula VII ##STR74## in which R.sub.1, R.sub.2, R.sub.3, L.sub.1, T and L.sub.2 are as previously defined and R.sub.6 represents hydrogen, with a compound of formula VIII ##STR75## in which R.sub.61, Q' and Y are as previously defined, for example by heating the two compounds at a temperature in the range of 0-200.degree. C., in the absence or presence of a solvent for the reactants, at atmospheric pressure, in the absence or presence of a dehydrating agent, and then reducing the intermediate obtained directly, by reaction with a reducing agent, in the presence of a solvent for the reactants, at a temperature in the range 0-150.degree. C., at atmospheric pressure; or
- e) reacting a compound of formula IX ##STR76## in which R.sub.1, R.sub.2, R.sub.3, L.sub.1, T, L.sub.2, R.sub.6 and Y are as previously defined and --C(O)--Q"-- represents a group which on reduction gives Q, with a reducing agent, in the absence or presence of a solvent for the compound of formula IX, at a temperature in the range 0-200.degree. C., at atmospheric pressure; or
- f) by reacting a compound of formula X ##STR77## in which R.sub.1, R.sub.2, R.sub.3, L.sub.1, T, R.sub.6, Q and Y are as previously defined and --L.sub.2 "--CO-- represents a group which on reduction gives L.sub.2, with a reducing agent, in the absence or presence of a solvent for the compound of formula X, at a temperature in the range 0-200.degree. C., at atmospheric pressure; or
- g) by reacting a compound of formula XI ##STR78## in which R.sub.1, R.sub.2, R.sub.3, L.sub.1, T, L.sub.2 and R.sub.6 are as previously defined with a compound of formula XII

- in which Z represents a leaving group, in the absence or presence of a base, in the presence of a solvent for the reactants, at a temperature in the range 0-200.degree. C.; or
- h) deprotecting compounds of formula XIII ##STR79## in which R.sub.1, R.sub.2, R.sub.3, L.sub.1, T, L.sub.2, Q and Y are as previously defined and PG represents an amine protecting group; or
- i) reacting a compound of formula XVII ##STR80## in which R.sub.1, R.sub.2, R.sub.3, L.sub.1, T, L.sub.2 and R.sub.6 are as previously defined, with a compound of formula XXVII ##STR81## in which R.sub.8, R.sub.9 and R.sub.10 are as previously defined, at a temperature in the range 0-250.degree. C., in the absence or presence of a solvent for the reactants, in the absence or presence of a base, at atmospheric pressure, to give compounds of formula IIa in which Q represents a group of formula --CH.sub.2 --CH(OH)--CH.sub.2 -- and Y represents a group of formula (1); or
- j) by reacting a compound of formula XXXI ##STR82## in which Z is a leaving group, with a compound of formula XXXII ##STR83## at a temperature in the range -50 to 250.degree. C. in the presence of a base, in the presence of an inert organic liquid which is a solvent for XXXI and XXXII, to give compounds of formula IIa in which L.sub.1 is other than a bond and T represents oxygen; or
- k) condensing a compound of formula XXXIII ##STR84## in which Z represents a leaving group with a compound of formula XXXII ##STR85## in which L.sub.2, R.sub.6, Q and Y are as defined previously, at a temperature in the range -50 to 250.degree. C., in the presence of a base and in the presence of an inert organic liquid which is a solvent for XXXII and XXXIII, to give compounds of formula IIa in which T represents a carbonyl group; or
- l) alkylation of a corresponding compound of formula IIa in which R.sub.6 is hydrogen, to give compounds of formula IIa in which R.sub.6 is a group other than hydrogen.
- 14. The process as defined in claim 13, wherein, in step d), compounds VII and VIII are heated at a temperature of from 15 to 150.degree. C.
- 15. The process as defined in claim 13, wherein in step f), compound X is reacted with the reducing agent at a temperature of from 15 to 150.degree. C.
- 16. The process as defined in claim 13, wherein in step l), compound XVII is reacted with compound XXVII at a temperature of from 50 to 150.degree. C.
- 17. The process as defined in claim 13, wherein in step j), compound XXXI is reacted with compound XXXII at a temperature of from 15 to 150.degree. C.
