Claims
- 1. A process for the preparation of a compound of formula I: or a pharmaceutical acceptable salt thereof wherein:
- X is --NR.sub.1 --, --S(O).sub.g --, or --O--;
- R.sub.1 is --H, C.sub.1-6 alkyl, optionally substituted with one or more --OH, --CN or halo,
- --(CH.sub.2).sub.h -aryl, --COR.sub.1-1, --COOR.sub.1-2, --CO--(CH.sub.2).sub.h --COR.sub.1-1, --SO.sub.2 l--C.sub.1-6 alkyl,
- --SO.sub.2 --(CH.sub.2).sub.h -aryl, or --(CO).sub.i -Het;
- R.sub.1-1 is --H, C.sub.1-6 alkyl, optionally substituted with one or more --OH, --CN or halo,
- --(CH.sub.2).sub.h -aryl, or --(CH.sub.2).sub.h --OR.sub.1-3 ;
- R.sub.1-2 is C.sub.1-6 alkyl, optionally substituted with one or more OH, CN or halo,
- --(CH.sub.2).sub.h -aryl, or --(CH.sub.2).sub.h --OR.sub.1-3 ;
- R.sub.1-3 is --H, C.sub.1-6 alkyl, --(CH.sub.2).sub.h -aryl, or --CO(C.sub.1-6 alkyl);
- R.sub.2 is --H, C.sub.1-6 alkyl --(CH.sub.2).sub.h -aryl, or halo;
- R.sub.3 and R.sub.4 are independently --H or halo;
- R.sub.5 is --H, C.sub.1-12 alkyl, optionally substituted with one or more halo,
- C.sub.3-2 cycloalkyl, C.sub.1-6 alkoxy; aryl is a phenyl, pyridyl or naphthyl moiety, optionally substituted with one or more
- --F, --Cl, --Br, --I, --CN, --OH, --SH, C.sub.1-6 alkyl, C.sub.1-6 alkoxy, or C.sub.1-6 thioalkyl;
- Het is 5 to 10 membered heterocyclic rings having one to three O, N and S atom;
- g is 0, 1 or 2; h is 1, 2, 3 or 4; i is 0 or 1; m is 0, 1, 2, or 3; n is 0, 1, 2, or 3; the dotted line ------ is a single or a double bound; and with the proviso that m and n taken together are 3; which comprises:
- a) introducing --C(.dbd.O)R.sub.5 or --C(.dbd.O)R.sub.5-1 into the amino group of a compound of the formula II; ##STR6## b) when X.sub.1 is a protected X in Formulae deprotecting the protecting group by catalytic hydrogenation of a compound of formula III; ##STR7## c) when ------ is a double bond and m and n taken together is 3, reacting a compound of structure 53 with a compound of structure 54; ##STR8## d) when ------ is a single bond, catalytically hydrogenating a compound of formula I;
- e) when ------is a double bond, eliminating an element of water from a compound of formula IV, ##STR9## d) when X is NR.sub.1 and R.sub.1 is --COR.sub.1-1, --COOR.sub.1-2, --CO--(CH.sub.2).sub.h --COR.sub.1-1, --SO.sub.2 --C.sub.1-6 alkyl, --SO.sub.2 --(CH.sub.2).sub.h -aryl, or --(CO)-Het, reacting a compound of formula V ##STR10## with L--COR.sub.1-1, L--COOR.sub.1-2, L--CO--(CH.sub.2).sub.h --COR.sub.1-1, L--SO.sub.2 --C.sub.1-6 alkyl,
- L--SO.sub.2 --(CH.sub.2).sub.h -aryl, or L--(CO).sub.i -Het,
- wherein X.sub.1 is X or protected X; R.sub.5-1 is R.sub.5 or protected R.sub.5 ;
- R.sub.1, R.sub.1-1, R.sub.1-2 R.sub.2, R.sub.3, R.sub.4, R.sub.5, aryl, Het, X and ------ are as defined above, L is a leaving group; and
- e) thereafter, if necessary, removing any protecting groups.
- 2. A process for preparing a compound of structure 47 which comprises: ##STR11## a) condensing compounds of structures 42 and 43 to provide a compound of structure 44 at about -78.degree. C. to about -40.degree. C. in the presence of a lithium base;
- b) eliminating an element of water to provides compound 45;
- c) treating 45 with n-butyllithium in an appropriate solvent, followed by addition of (R)-glycidyl butyrate to provide oxazolidinone 46a in enantiomerically enriched form at the 5-position of the oxazolidinone ring;
- d) converting 46a to the corresponding alkyl or aryl sulfonate 46b by treatment with alkyl or aryl sulfonyl chloride in the presence of triethylamine or pyridine;
- e) treating 46b with an alkali metal azide such as sodium or potassium azide with an optional catalyst to afford azide derivatives;
- f) reducing the azide derivatives to the corresponding amine 46c by hydrogenation in the presence of a palladium, platinum or nickel catalyst in an appropriate solvent, or preparing amine 46c by treating 46b with an appropriate solvent which is saturated with ammonia and heating the mixture to 100.degree. C. in a sealed tube;
- g) acylating 46c with an acid chloride or anhydride in the presence of a base such as pyridine or triethylamine at temperatures ranging from -40.degree. C. to 40.degree. C. to provide the N-acyl oxazolidinone 46d;
- h) reducing the double bond in the heterocyclic ring to a single bond to give 47 by catalytic hydrogenation when it is desirable;
- i) where X is a sulfur atom, oxidizing the sulfur group to provide the corresponding sulfones and sulfoxides when it is desirable; and
- j) where X is N with a protecting group, optionally removing the protecting group after the step (g) or (h);
- wherein X is O, S or N with a protecting group, R.sub.5 is as defined in claim 1; and R' is alkyl or phenyl.
- 3. A process for preparing a compound of structure 52 which comprises:
- a) reacting a compound of structure 48 with a protected aniline 19 in the presence of an appropriate base, followed by the addition of zinc chloride and an appropriate catalyst to provide a compound of structure 49;
- b) acylating structure 49 to provide structure 50;
- c) treating 50 with n-butyllithium in an appropriate solvent, followed by addition of (R)-glycidyl butyrate to provide oxazolidinone 51a in enantiomerically enriched form at the 5-position of the oxazolidinone ring;
- d) converting 51a to the corresponding alkyl or aryl sulfonate 51b by treatment with alkyl or aryl sulfonyl chloride in the presence of triethylamine or pyridine;
- e) treating 51b with an alkali metal azide such as sodium or potassium azide with an optional catalyst to afford azide derivatives;
- f) reducing the azide derivatives to the corresponding amine 51c by hydrogenation in the presence of a palladium, platinum or nickel catalyst in an appropriate solvent, or preparing amine 51c by treating 51b with an appropriate solvent which is saturated with ammonia and heating the mixture to 100.degree. C. in a sealed tube;
- g) acylating 51c with an acid chloride or anhydride in the presence of a base such as pyridine or triethylamine at temperatures ranging from -40.degree. C. to 40.degree. C. to provide the N-acyl oxazolidinone 51d;
- h) reducing the double bond in the heterocyclic ring to a single bond to give 52 by catalytic hydrogenation when it is desirable;
- i) where X is a sulfur atom, oxidizing the sulfur group to provide the corresponding sulfones and sulfoxides when it is desirable; and
- j) where X is N with a protecting group, optionally removing the protecting group after the step (g) or (h); ##STR12## wherein X is O, S or N with a protecting group, R.sub.3, R.sub.4 and R.sub.5 are as defined in claim 1; and
- R' is alkyl or phenyl.
- 4. A process for preparing a compound of structure 64 or 65 which comprises:
- a) reacting a compound of structure 59 with a protected aniline 19 to afford structure 60;
- b) acylating 60 to provides structure 61;
- c) treating 61 with an appropriate acid to provide a mixture of 62 and 63;
- d) separating 62 and 63 by chromatography; and
- e) treating 62 or 63 with n-butyllithium in an appropriate solvent, followed by addition of (R)-glycidyl butyrate to provide oxazolidinone 64a or 65a in enantiomerically enriched form at the 5-position of the oxazolidinone ring,
- f) converting 64a or 65a to the corresponding alkyl or aryl sulfonlate 64b or 65b by treatment with alkyl or aryl sulfonyl chloride in the presence of triethylamine or pyridine;
- g) treating 64b or 65b with an alkali metal azide such as sodium or potassium azide with an optional catalyst to afford azide derivatives;
- h) reducing the azide derivatives to the corresponding amine 64c or 65c by hydrogenation in the presence of a palladium, platinum or nickel catalyst in an appropriate solvent, or preparinl, amine 64c or 65c by treating 64b or 65b with an appropriate solvent which is saturated with ammonia and heating the mixture to 100.degree. C. in a sealed tube;
- i) acylating 64c or 65c with an acid chloride or anhydride in the presence of a base such as pyridine or triethylamine at temperatures ranging from -40.degree. C. to 40.degree. C.; and
- j) removing --CO.sub.2 CH.sub.2 Ph on the nitrogen to provide 64d or 65d;
- k) when it is desirable, reducing the double bond in the heterocyclic ring to a single bond by catalytic hydrogenation before or after step (j);
- wherein R.sub.3, R.sub.4 and R.sub.5 are as defined in claim 1, R' is alkyl or phenyl. ##STR13##
