Claims
- 1. An isolated plasmid comprising a gene, said gene encoding a polypeptide having Rep protein activity and, said polypeptide comprising an amino acid sequence that is at least 90% homologous to the amino acid sequence of SEQ ID NO: 8.
- 2. The plasmid according to claim 1, wherein said polypeptide comprises an amino acid sequence that is at least 99% homologous to the amino acid sequence of SEQ ID NO: 8.
- 3. The plasmid according to claim 3, wherein said polypeptide comprises the amino acid sequence of SEQ ID NO: 8.
- 4. The plasmid according to claim 1, wherein said plasmid is isolated from Corynebacterium thermoaminogenes AJ12309.
- 5. The plasmid according to claim 1, which has the restriction map shown in FIG. 2.
- 6. The plasmid according to claim 1, wherein said gene is obtained from a plasmid having the restriction map shown in FIG. 2.
- 7. The method of isolating the plasmid according to claim 1, comprising
(A) culturing a Corynebacterium thermoaminogenes in a culture medium, (B) obtaining fractions by an alkali method, and (C) isolating said plasmid.
- 8. The method according to claim 7, further comprising analyzing the fractions by agarose gel electrophoresis.
- 9. An isolated polynucleotide comprising a nucleic acid sequence that encodes a polypeptide having Rep protein activity and, said polypeptide comprising an amino acid sequence that is at least 90% homologous to the amino acid sequence of SEQ ID NO. 8.
