Claims
- 1. A plasmid comprising at least two replicons, one of which is derived from SCP2 or SCP2* and one of which is derived from SCP2@.
- 2. The plasmid of claim 1 that also comprises one or more genes selected from par and tra genes.
- 3. The plasmid of claim 1 that also comprises one or more polyketide synthase genes or non-ribosomal peptide synthase genes.
- 4. The plasmid of claim 1 that is pSMALL (pKOS97-150).
- 5. A plasmid comprising a replicon of SCP2@.
- 6. The plasmid of claim 5 that is selected from the group of plasmids consisting of SCP2@, pBoost (pKOS97-188), and pBOOST (pKOS146-145).
- 7. A method for producing a polyketide, which method comprises: transforming a host cell, said host cell comprising a first plasmid having a first SCP2-derived replicon, with a second plasmid having a second SCP2-derived replicon and capable of recombining with the first plasmid to yield a co-integrate plasmid that comprises at least two SCP2-derived replicons, wherein either said first or second plasmid comprises one or more polyketide synthase genes and wherein one of said SCP2-derived replicons is derived from SCP2@; and culturing said host cell under conditions such that said recombination occurs and said PKS genes are expressed so as to produce a functional PKS that produces said polyketide.
- 8. The method of claim 6, wherein said host cell is a Streptomyces host cell.
- 9. The method of claim 6, wherein said second plasmid is pBoost (pKOS97-188) or pBOOST (pKOS146-145).
- 10. A method of forming a co-integrate plasmid having increasing plasmid stability relative to plasmids from which said co-integrate is derived, which method comprises transforming a host cell, said host cell comprising a first plasmid having a first SCP2-derived replicon, with a second plasmid having a second SCP2-derived replicon capable of recombining with the first plasmid to yield a co-integrate plasmid that comprises at least two SCP2-derived replicons, wherein said first and second plasmids comprise one or more polyketide synthase genes and wherein one of said SCP2-derived replicons is derived from SCP2@; and culturing said host cell under conditions such that said recombination occurs.
- 11. The method of claim 10 wherein one of said first and second plasmids is selected from the group of plasmids consisting of pBoost (pKOS97-188) and pBOOST (pKOS146-145).
