Claims
- 1. A process of preparing d1-(methyl)-16, 16-(dimethyl)-11 alpha, 15-alpha-dihydroxy-9-oxo-2,13-trans, trans-prostadienoate and its congeners, which comprises
- (i) reacting (i) a 2-Halo-alkanedioate diester of the formula ##STR13## wherein R.sub.2 and R.sub.3 are selected from the class consisting essentially of C.sub.1 -C.sub.6 alkyl, and R.sub.2 may be the same as or different from R.sub.3, and Halo is selected from the class consisting of chloro-,bromo-, and iodo-, with (ii) an alkali metal thio-monocyclic-aryloxide, said alkali metal being selected from the class consisting of lithium, sodium and potassium, and said aryl moiety being selected from the class consisting essentially of phenyl and phenyl substituted with halogen, C.sub.1 -C.sub.4 alkyl, C.sub.1 -C.sub.4 alkoxy or trifluoromethyl, to form a compound having the formula ##STR14## wherein Aryl is selected from the class consisting essentially of phenyl and phenyl substituted with halogen, C.sub.1 -C.sub.4 alkyl, C.sub.1 -C.sub.4 alkoxy or trifluoromethyl,
- (ii) hydrolizing the preceding compound with a strong alkali to form a compound having the formula ##STR15## (iii) reacting the preceding compound with a lower anhydride of formula ##STR16## or an acyl halide of the formula ##STR17## to form a mixed anhydride having the formula ##STR18## wherein R.sub.6 and R.sub.7 are selected from the class consisting essentially of C.sub.1 -C.sub.3 alkyl or halo-substituted C.sub.1 -C.sub.3 alkyl, and R.sub.6 may be the same as or different from R.sub.7, and X is halogen selected from the class consisting of chloro, bromo and iodo,
- (iv) reacting said mixed anhydride with furan to form an acyl furan compound having the formula ##STR19## (v) reacting the carbonyl group of an acyl furan having the formula ##STR20## wherein R.sub.5 is selected from the class consisting essentially of hydrogen and a C.sub.1 -C.sub.6 alkyl, with a carbonyl reducing agent to form a compound having the formula ##STR21## (vi) subjecting the preceding compound to an acid catalyzed rearrangement to form a cyclopentenone having the formula ##STR22## (vii) reacting the preceding compound with a peroxy organic acid followed by thermolysis to form a cyclopentenonylalkenyl compound having the formula ##STR23## (viii) protecting the 4-hydroxyl group of the cyclopentenonyl moiety of the preceding compound with a protective group P.sub.2, to form a compound having the formula ##STR24## (ix) reacting the preceding compound with a reagent capable of furnishing a prostaglandin beta-chain having the formula ##STR25## wherein P.sub.3 is a protective group, and X is a metal support capable of introducing said side chain as a prostaglandin betachain to form a compound having the formula ##STR26## and (x) reacting said compound with a weak organic acid to remove the protective groups P.sub.2 and P.sub.3 to obtain a compound having the formula ##STR27## comprising at least one of the optically active isomers of said compound.
- 2. The process of claim 1 wherein any of the compounds prepared in steps (vi) or (vii) are resolved into their optically active isomers before being further reacted.
- 3. The process of claim 1 wherein R.sub.5 is methyl, the compounds prepared in step (x) are resolved by chromatography and the optically active 11-alpha, 15-alpha-dihydroxy isomer is recovered.
- 4. The process of claim 1 wherein, in the compound prepared by step (vi), R.sub.1 is hydrogen, and said compound is esterified before step (vii).
- 5. The process of claim 1 wherein the reagent-reactant of step (ix) is a lithio-cuprate.
- 6. The process of claim 1, including the additional step of chromatographic separation of the individual 15-alpha and 15-beta isomers of the product of step (x).
- 7. The process of claim 1, wherein any of the compounds prepared in steps (vi) or (vii) are resolved into their optically active isomers, and the product of step (x) is chromatographically separated into individual 15-alpha and 15-beta isomers, each being obtained in the nat. configuration.
- 8. The process according to claim 1 wherein said alcohol protecting group is selected from the group consisting of tetrahydropyranyl, trialkysilyl, or alpha-alkoxyethyl moieties.
