Claims
- 1. Substantially pure purine nucleoside phosphorylase having the following properties:
- (1) temperature stability: stable even upon heating at 60.degree. C. for 15 minutes;
- (2) specific activity: 400 or more (U/mg) at 80% of enzyme purity;
- (3) an optimum pH range of 7 to 8, and a stable pH range of 5 to 9; and
- (4) a molecular weight of about 31,000 measured by SDS-polyacrylamide gel electrophoresis, said phosphorylase being capable of catalyzing the reaction purine nucleoside+phosphoric acid .revreaction.purine base+pentose 1-phosphate.
- 2. Substantially pure pyrimidine nucleoside phosphorylase having the following properties:
- (1) temperature stability: stable even upon heating at 60.degree. C. for 15 minutes;
- (2) specific activity: 250 or more (U/mg) at 80% of enzyme purity;
- (3) an optimum pH range of 7 to 9, and a stable pH range of 5 to 9; and
- (4) a molecular weight of about 45,000 measured by SDS-polyacrylamide gel electrophoresis,
- said phosphorylase being capable of catalyzing the reaction pyrimidine nucleoside+phosphoric acid .revreaction.pyrimidine base+pentose 1-phosphate.
- 3. The phosphorylaseof claim 1, having the following additional properties:
- (1) an optimum temperature range of 60 to 80.degree. C.;
- (2) specific activity: 450 (U/mg) at 90% of enzyme purity; and
- (3) specificity for inosine, 2'-deoxyinosine, guanosine and 2'-deoxyguanosine.
- 4. The phosphorylase of claim 1, which has the same properties as those of purine nucleoside phosphorylase obtainable by a process comprising extracting the purine nucleoside phosphorylase from a culture of Bacillus stearothermophilus having the accession number FERM BP-2758.
- 5. The phosphorylase of claim 2, having the following additional properties:
- (1) an optimum temperature range of 60 to 70.degree. C.;
- (2) specific activity: 297 (U/mg) at 90% of enzyme purity; and
- (3) specificity for uridine, 2'-deoxyuridine, ribofuranosylthymine and thymidine.
- 6. The phosphorylase of claim 2, which has the same properties as those of pyrimidine nucleoside phosphorylase obtainable by a process comprising extracting the pyrimidine nucleoside phosphorylase from a culture of Bacillus stearothermophilus having the accession number FERM BP-2758.
- 7. In a process for enzymatically producing nucleosides by the use of nucleoside phosphorylase, the improvement which comprises using, as the nucleoside phosphorylase, the purine nucleoside phosphorylase according to claim 1.
- 8. The process of claim 7, wherein the phosphorylase has the same properties as those of purine nucleoside phosphorylase obtainable by a process comprising extracting the purine nucleoside phosphorylase from a culture of Bacillus stearothermophilus having the accession number FERM BP-2758.
- 9. In a process for enzymatically producing nucleosides by the use of nucleoside phosphorylase, the improvement which comprises using, as the nucleoside phosphorylase, the purine nucleoside phosphorylase according to claim 3.
- 10. The process of claim 9, wherein the phosphorylase has the same properties as those of purine nucleoside phosphorylase obtainable by a process comprising extracting the purine nucleoside phosphorylase from a culture of Bacillus stearothermophilus having the accession number FERM BP-2758.
- 11. In a process for enzymatically producing nucleosides by the use of nucleoside phosphorylase, the improvement which comprises using, as the nucleoside phosphorylase, the pyrimidine nucleoside phosphorylase according to claim 2.
- 12. The process of claim 11, wherein the phosphorylase has the same properties as those of pyrimidine nucleoside phosphorylase obtainable by a process comprising extracting the pyrimidine nucleoside phosphorylase from a culture of Bacillus stearothermophilus having the accession number of FERM BP-2758.
- 13. In a process for enzymatically producing nucleosides by the use of nucleoside phosphorylase, the improvement which comprises using, as the nucleotide phosphorylase, the pyrimidine nucleoside phosphorylase according to claim 5.
- 14. The process of claim 13, wherein the phosphorylase has the same properties as those of pyrimidine nucleoside phosphorylase obtainable by a process comprising extracting the pyrimidine nucleoside phosphorylase from a culture of Bacillus stearothermophilus having the accession number of FERM BP-2758.
- 15. In a process for enzymatically producing nucleosides by the use of nucleoside phosphorylase, the improvement which comprises using a nucleoside phosphorylase comprising the purine nucleoside phosphorylase according to claim 1, and the pyrimidine nucleoside phosphorylase according to claim 2.
- 16. In a process for enzymatically producing nucleosides by the use of nucleoside phosphorylase, the improvement which comprises using a nucleoside phosphorylase comprising the purine nucleoside phosphorylase according to claim 3 and the pyrimidine nucleoside phosphorylase according to claim 4.
- 17. The process of claim 15, wherein the purine nucleoside phosphorylase has the same properties as those of purine nucleoside phosphorylase obtainable by a process comprising extracting the purine nucleoside phosphorylase from a culture of Bacillus stearothermophilus having the accession number FERM BP-2758, and the pyrimidine nucleoside phosphorylase has the same properties as those of pyrimidine nucleoside phosphorylase obtainable by a process comprising extracting the pyrimidine nucleoside phosphorylase from a culture of Bacillus stearothermophilus having the accession number of FERM BP-2758.
- 18. The process of claim 16, wherein the purine nucleoside phosphorylase has the same properties as those of purine nucleoside phosphorylase obtainable by a process comprising extracting the purine nucleoside phosphorylase from a culture of Bacillus stearothermophilus having the accession number FERM BP-2758, and the pyrimidine nucleoside phosphorylase has the same properties as those of pyrimidine nucleoside phosphorylase obtainable by a process comprising extracting the pyrimidine nucleoside phosphorylase from a culture of Bacillus stearothermophilus having the accession number of FERM BP-2758.
