Claims
- 1. A process for the production of D-.alpha.-amino acids characterized in that D-N-carbamoyl-.alpha.-amino acids represented by the formula:
- R--CH(NHCONH.sub.2)--COOH
- wherein
- R is phenyl; phenyl substituted with hydroxy; alkyl having 1 to 4 carbon atoms; alkyl having 1 to 4 carbon atoms substituted with hydroxy, alkylthio, carboxyl, amino, phenyl or phenyl substituted with hydroxy or amido; aralkyl having 7 to 8 carbon atoms; or thienyl; provided that when R is phenyl substituted with hydroxyl, optionally one or more hydroxyl groups are attached at any of the o-, m-, and p-positions;
- are converted into the corresponding D-.alpha.-amino acids in an aqueous medium, using an enzyme which is produced by a transformant obtained by transforming a host bacterial cell with a recombinant DNA comprising a vector and a DNA fragment having a gene encoding said enzyme, said enzyme being capable of converting D-N-carbamoyl-.alpha.-amino acids by removal of their carbamoyl groups into the corresponding D-.alpha.-amino acids, after which the D-.alpha.-amino acids are collected, wherein the recombinate DNA is a plasmid selected from the group consisting of pAHD 101, pAD 108, pPHD 301, and pPD 304.
- 2. A process for the production of D-.alpha.-amino acids characterized in that D-N-carbamoyl-.alpha.-amino acids represented by the formula:
- R--CH(NHCONH.sub.2)--COOH
- wherein
- R is phenyl; phenyl substituted with hydroxy; alkyl having 1 to 4 carbon atoms; alkyl having 1 to 4 carbon atoms substituted with hydroxy, alkylthio, carboxyl, amino, phenyl or phenyl substituted with hydroxy or amido; aralkyl having 7 to 8 carbon atoms; or thienyl; provided that when R is phenyl substituted with hydroxyl, one or more hydroxyl groups are attached at any of the o-, m-, and p-positions;
- are converted into the corresponding D-.alpha.-amino acids in an aqueous medium, using an enzyme which is produced by a transformant obtained by transforming a host bacterial cell with a recombinant DNA comprising a vector and a DNA fragment having a gene encoding a D-N-carbamoyl-.alpha.-amino acid amidohydrolase capable of converting D-N-carbamoyl-.alpha.-amino acids by removal of their carbamoyl groups into the corresponding D-.alpha.-amino acids, after which the D-.alpha.-amino acids are collected, wherein said transformant is selected from the group consisting of Escherichia coli JM 109 pAHD 101, Escherichia coli JM 109 pAD 108 (FERM BP-3184), Escherichia coli JM 109 pPHD 301 and Escherichia coli JM 109 pPD 304 (FERM BP-3183).
- 3. A process for the production of a D-.alpha.-amino acids characterized in that D-N-carbamoyl-.alpha.-amino acids represented by the formula:
- R--CH(NHCONH.sub.2)--COOH
- wherein
- R is phenyl; phenyl substituted with hydroxy; alkyl having 1 to 4 carbon atoms; alkyl having 1 to 4 carbon atoms substituted with hydroxy, alkylthio, carboxyl, amino, phenyl or phenyl substituted with hydroxy or amido; aralkyl having 7 to 8 carbon atoms; or thienyl; provided that when R is phenyl substituted with hydroxyl, optionally one or more hydroxyl groups are attached at any of the o-, m-, and p-positions;
- are converted into the corresponding D-.alpha.-amino acids in an aqueous medium, using an enzyme which is produced by a bacterium selected from the group consisting of Pseudomonas sp. KNK 0030A (FERM BP-3181), and Pseudomonas sp. KNK 505 (FERM BP-3182), said enzyme being capable of converting D-N-carbamoyl-.alpha.-amino acids by removal of their carbamoyl groups into the corresponding D-.alpha.-amino acids, after which the D-.alpha.-amino acids are collected.
