Claims
- 1. A process for the preparation of a substantially pure compound of the formula: ##STR22## wherein R.sub.6 and R.sub.7 are each hydrogen or R.sub.6 and R.sub.7 are independently selected from ##STR23## wherein R.sub.a and R.sub.b are independently selected from hydrogen, loweralkyl and phenyl and R.sub.c, R.sub.d and R.sub.e are independently selected from hydrogen, loweralkyl, trifluoromethyl, alkoxy, halo and phenyl; and ##STR24## wherein the naphthyl ring is unsubstituted or substituted with one, two or three substitutents independently selected from loweralkyl, trifluoromethyl, alkoxy and halo; or
- R.sub.6 is as defined above and R.sub.7 is R.sub.7a OC(O)--wherein R.sub.7a is benzyl; or
- R.sub.6 and R.sub.7 taken together with the nitrogen atom to which they are bonded are ##STR25## wherein R.sub.f, R.sub.g, R.sub.h and R.sub.i are independently selected from hydrogen, loweralkyl, alkoxy, halogen and trifluoromethyl and R.sub.8 is hydrogen or --C(O)R" wherein R" is loweralkyl, alkoxy, benzyloxy or phenyl wherein the phenyl ring is unsubstituted or substituted with one, two or three substituents independently selected from loweralkyl, trifluoromethyl, alkoxy and halo; or an acid addition salt thereof comprising
- (I) reacting a compound of the formula: ##STR26## wherein R.sub.6 and R.sub.7 are each hydrogen or R.sub.6 and R.sub.7 are independently selected from ##STR27## wherein R.sub.a and R.sub.b are independently selected from hydrogen, loweralkyl and phenyl and R.sub.c, R.sub.d and R.sub.e are independently selected from hydrogen, loweralkyl, trifluoromethyl, alkoxy, halo and phenyl; and ##STR28## wherein the naphthyl ring is unsubstituted or substituted with one, two or three substitutents independently selected from loweralkyl, trifluoromethyl, alkoxy and halo; or
- R.sub.6 is as defined above and R.sub.7 is R.sub.7a OC(O)--wherein R.sub.7a is benzyl; or
- R.sub.6 and R.sub.7 taken together with the nitrogen atom to which they are bonded are ##STR29## wherein R.sub.f, R.sub.g, R.sub.h and R.sub.i are independently selected from hydrogen, loweralkyl, alkoxy, halogen and trifluoromethyl and R.sub.8 is hydrogen with a mixture of (a) an acid selected from (i) R.sub.26 --COOH wherein R.sub.26 is loweralkyl, haloalkyl, phenyl or halophenyl, (ii) R.sub.27 --SO.sub.3 H wherein R.sub.27 is OH, F, loweralkyl, haloalkyl, phenyl, loweralkyl-substituted phenyl, halophenyl or naphthyl and (iii) R.sub.28 --PO.sub.3 H.sub.2 wherein R.sub.28 is OH, loweralkyl or phenyl or a combination thereof and (b) a boron-containing reducing agent, said mixture prepared by adding the acid or combination of acids to the boron-containing reducing agent, followed by
- (IIa) reacting the reaction mixture of step (I) with a mixture of a borohydride reagent and R.sub.29 --COOH wherein R.sub.29 is loweralkyl, haloalkyl, phenyl or halophenyl, said mixture of the borohydride reagent and R.sub.29 --COOH prepared by adding the R.sub.29 --COOH to the borohydride reagent; or followed by
- (IIb) reacting the reaction mixture of step (I) with a boron complexing agent, followed by reaction of the resulting mixture with a ketone reducing agent.
- 2. The process of claim 1 for the preparation of a substantially pure compound of the formula: ##STR30## wherein R.sub.6 and R.sub.7 are each hydrogen or benzyl and R.sub.8 is hydrogen or --C(O)R" wherein R" is t-butyloxy; or an acid addition salt thereof comprising
- (I) reacting a compound of the formula: ##STR31## wherein R.sub.6 and R.sub.7 are each benzyl and R.sub.8 is hydrogen with a mixture of NaBH.sub.4 and methanesulfonic acid prepared by adding the methanesulfonic acid to the sodium borohydride, followed by
- (II) reacting the reaction mixture of step (I) with a mixture of NaBH.sub.4 and trifluoroacetic acid prepared by adding the trifluoroacetic acid to the NaBH.sub.4.
- 3. The process of claim 1 for the preparation of a substantially pure compound of the formula: ##STR32## wherein R.sub.6 and R.sub.7 are each hydrogen or benzyl and R.sub.8 is hydrogen or --C(O)R" wherein R" is t-butyloxy; or an acid addition salt thereof comprising
- (I) reacting a compound of the formula: ##STR33## wherein R.sub.6 and R.sub.7 are each benzyl and R.sub.8 is hydrogen with a mixture of NaBH.sub.4 and methanesulfonic acid prepared by adding the methanesulfonic acid to the sodium borohydride, followed by
- (II) reacting the reaction mixture of step (I) with triethanolamine, followed by reaction of the resulting mixture with NaBH.sub.4.
