Claims
- 1. A process for the production of clavulanic acid or a salt thereof which comprises cultivating a strain of Streptomyces clavuligerus in a suitable medium containing assimilable sources of carbon, nitrogen and mineral salt and isolating the clavulanic acid or salt thereof from the culture medium.
- 2. A process according to claim 1 wherein the Streptomyces clavuligerus is ATCC-27064 or a high yielding mutant thereof.
- 3. A process according to claim 1 wherein the cultivation takes place at a temperature of from about 15.degree. to about 40.degree. C and at a pH of between 5 and 8.5.
- 4. A process according to claim 1 wherein the cultivation takes place at a temperature of from about 20.degree. to about 35.degree. C and at a pH of between 6 and 7.5.
- 5. A process according to claim 1 wherein the cultivation takes place at a temperature of from about 25.degree. to about 30.degree. C and at a pH of between 6 and 7.5.
- 6. A process according to claim 1 wherein the clavulanic acid is recovered which comprises adjusting the culture filtrate to an acidic pH value and extracting the clavulanic acid therefrom.
- 7. A process according to claim 6 wherein the clavulanic acid is extracted into a water-immiscible organic solvent.
- 8. A process according to claim 7 wherein the water-immiscible organic solvent is n-butylacetate, methylisobutylketone, n-butanol or ethylacetate.
- 9. A process according to claim 8 wherein the solvent is n-butanol.
- 10. A process according to claim 7 for the production of a salt of clavulanic acid which comprises extracting the clavulanic acid back into an aqueous phase under approximately neutral conditions containing the desired salt ions and recovering the salt formed.
- 11. A process according to claim 1 for the production of a salt of clavulanic acid which comprises adsorbing the clarified culture filtrate at a pH from about 6 to 7 onto a basic anion exchange resin and recovering the salt formed.
- 12. A process according to claim 1 wherein an amine salt of clavulanic acid is produced which comprises extracting the culture filtrate at approximately neutral pH with an organic phase in which is disolved a water insoluble amine and recovering the amine salt of clavulanic acid.
- 13. A process according to claim 1 wherein an impure salt of clavulanic acid is recovered and thereafter purified chromatographically.
- 14. A process according to claim 13 for the production of a salt of clavulanic acid wherein the clavulanic acid is adsorbed from the culture medium onto a basic anion exchange resin from which it is recovered by elution with an inorganic salt.
- 15. A process according to claim 1 for the isolation of a salt of clavulanic acid which comprises adsorbing a culture filtrate onto a basic anion exchange resin, eluting the salt of clavulanic acid therefrom with an aqueous salt solution, desalting the resulting solution to remove the excess eluting salt and isolating the salt of clavulanic acid from the desalted solution.
- 16. A process according to claim 15 for the isolation of the lithium salt of clavulanic acid wherein the aqueous salt solution is an aqueous lithium salt solution.
- 17. A process according to claim 15 for the isolation of the sodium salt of clavulanic acid wherein the aqueous salt solution is an aqueous sodium salt solution.
- 18. A process according to claim 15 for the isolation of the potassium salt of clavulanic acid wherein the aqueous salt solution is an aqueous potassium salt solution.
- 19. A process according to claim 15 for the isolation of the calcium salt of clavulanic acid wherein the aqueous salt solution is an aqueous calcium salt solution.
