- 113. A method of preparing a sample of a phosphate linked oligonucleotide having a substantially reduced content of acrylonitrile adduct comprising:
providing a sample containing a plurality of oligomers, said oligomers linked to a solid support medium, said oligomers having a plurality of phosphorus protecting groups; contacting said oligomers with a deprotecting agent to remove substantially all of said phosphorus protecting groups from said oligomers;
said deprotecting reagent comprising at least one amine, the conjugate acid of said amine having a pKa of from about 8 to about 11; said deprotecting reagent optionally further comprising one or more solvents selected from the group consisting of alkyl solvents, haloalkyl solvents, cyanoalkyl solvents, aryl solvents and aralkyl solvents; optionally washing said oligomers; and reacting said oligomers with a cleaving reagent.
- 114. The method of claim 113, wherein said oligomers are not cleaved from the solid support medium by the deprotecting reagent.
- 115. The method of claim 113, wherein the amine is an aliphatic amine.
- 116. A method of preparing a sample of a phosphate linked oligonucleotide having a substantially reduced content of acrylonitrile adduct comprising:
providing a sample containing a plurality of solid support-linked oligomers, said oligomers having a plurality of phosphorus protecting groups; contacting said oligomers with a deprotecting agent to remove substantially all of said phosphorus protecting groups from said oligomers; washing said oligomers with a washing reagent, said washing reagent comprising at least one amine, the conjugate acid of said amine having a pKa of from about 8 to about 11; said deprotecting reagent optionally further comprising one or more solvents selected from the group consisting of alkyl solvents, haloalkyl solvents, cyanoalkyl solvents, aryl solvents and aralkyl solvents; and reacting said oligomers with a cleaving reagent.
- 117. The method of claim 116, wherein said oligomers are not cleaved from the solid support by the washing reagent.
- 118. The method of claim 116, wherein the amine is an aliphatic amine.
- 119. A method for deprotecting a phosphate-linked oligomer, said oligomer having a plurality of protected phosphorus linkages of Formula II:
- 120. The method of claim 119, wherein Rt is —CH2CH2—CN.
- 121. A method for deprotecting a phosphate-linked oligomer, said oligomer having a plurality of protected phosphorus linkages of Formula II:
- 122. The method of claim 121, wherein said oligomers are not cleaved from the solid support medium by the deprotecting reagent.
- 123. The method of claim 121, wherein the amine is an aliphatic amine.
