Claims
- 1. A subtilase enzyme variant having improved wash performance in detergents, comprising modification(s) in position(s) 252, 255 and/or 259 (in BASBPN numbering).
- 2. A subtilase enzyme variant having improved wash performance in detergents, comprising at least one modification chosen from the group comprising (in BASBPN numbering):
252L+255I; 252V+255A; 252M+255C+259H; 252S+255e+259C; 252K+255s+259C; or a variant comprising one or more conservative modification(s) in any of the above mentioned variants (e.g., a conservative modification of a 252I (a hydrophobic amino acid)+255I variant include variants such as 252I (a hydrophobic amino acid)+255I, and 252V (a hydrophobic amino acid)+255I.
- 3. The subtilase enzyme variant according to claim 2, wherein the modification is chosen from the group comprising (in BASBPN numbering):
N252L+T255I; N252V+T255A; N252M+T255C+S259H; N252S+T255E+S259C; N252K+T255S+S259C; or a variant comprising one or more conservative modification(s) in any of the above mentioned variants (e.g., a conservative modification of a N252L (a hydrophobic amino acid)+T255I variant include variants such as N252I (a hydrophobic amino acid)+T255I, and N252V (a hydrophobic amino acid)+T255I.
- 4. The variant of any of claims 1-3, wherein the parent subtilase is chosen from the sub-group I-S1.
- 5. The variant of claim 4, wherein the parent subtilase is chosen from the group comprising ABSS168, BASBPN, BSSDY, and BLSCAR or functional variants thereof having retained the characteristic of sub-group I-S1.
- 6. The variant of any of claims 1-3, wherein the parent subtilase is chosen from the sub-group I-S2.
- 7. The variant of claim 6, wherein the parent subtilase is chosen from the group comprising BLS147, BLS309, BAPB92, TVTHER AND BYSYAB or functional variants thereof having retained the characteristic of sub-group I-S2.
- 8. The variant of any of claims 1-7, wherein said modification(s) is/are combined with one or more modification(s) in any other position(s).
- 9. The variant of claim 8, wherein said modification(s) is/are combined with modification(s) in one or more of the positions 27, 36, 57, 76, 97, 101, 104, 120, 123, 167, 170, 206, 218, 222, 224, 235 and 274.
- 10. The variant of claim 9, wherein said subtilase belongs to the I-S2 sub-group and said further change is chosen from the group comprising K27R, *36D, S57P, N76D, G97N, S101G, V104A, V104N, V104Y, H120D, N123S, Y167A, Y167I, R170S, R170L, R170N, Q206E, N218S, M222S, M222A, T224S, K235L, and T274A.
- 11. The variant of claim 10, comprising any of the variants V104N+S111G, K27R+V104Y+N123S+T274A, or N76D+V104A, or other combinations of these mutations (V104N, S101G, K27R, V104Y, N123S, T274A, N76D, V104A), in combination with any one or more of the substitutions, deletions and/or insertions mentioned in any of claims 1 to 10.
- 12. The subtilase variant of any of the preceding claims, wherein said modificaton(s) is/are combined with modification(s) in one or more of the positions 129, 131, 133 and 194.
- 13. The variant of claim 12, wherein said subtilase belongs to the I-S2 sub-group and said further change is chosen from the group comprising P129K, P131H, A133P, A133D and A194P.
- 14. An isolated DNA sequence encoding a subtilase variant of any of claims 1-13.
- 15. An expression vector comprising an isolated DNA sequence of claim 14.
- 16. A microbial host cell transformed with an expression vector of claim 15.
- 17. The microbial host of claim 16, which is a bacterium, preferably a Bacillus, especially B. lentus.
- 18. The microbial host of claim 16, which is a fungus or yeast, preferably a filamentous fungus, especially an Aspergillus.
- 19. A method for producing a variant of any of claims 1-13, wherein a host of any of claims 16-18 is cultured under conditions conducive to the expression and secretion of said variant, and the variant is recovered.
- 20. A composition comprising a subtilase variant of any of claims 1-13.
- 21. The composition according to claim 20, which additionally comprises a cellulase, lipase, cutinase, oxidoreductase, another protease, or an amylase.
- 22. The composition of claim 20 or 21, wherein the composition is a detergent composition.
- 23. Use of a subtilase variant according to any of claims 1-13 or an enzyme composition according to any of claims 20-22 in a laundry and/or a dishwash detergent.
- 24. A process for the identification of a protease variant exhibiting improved wash performance in detergents, which comprises
(a) effecting a mutation in DNA encoding a subtilase enzyme or its pre- or preproenzyme at one or more of the positions corresponding to amino acid (in BASBPN numbering):
(i) N252L+T255I; (ii) N252V+T255A; (iii) N252M+T255C+S259H; (iv) N252S+T255E+S259C; (v) N252K+T255S+S259C; or (vi) a variant comprising one or more conservative modification(s) in any of the above mentioned variants; (b) transforming a Bacillus strain with said mutated DNA; (c) selecting strains producing such protease variants; (d) fermenting/growing such a strain; (e) recovering said protease variant; and (f) testing for improved wash performance in detergents.
- 25. A modified subtilase, comprising a mutation in an amino acid sequence of a subtilase, wherein the mutation comprises one or more of the following substititions:
(a) a substitution of the amino acid at position 252 with K, L, M, S or V, (b) a substitution of the amino acid at position 255 with A, C, E, I, or S, (c) a substitution of the amino acid at position 259 with C or H, wherein the positions are numbered according to the amino acid sequence of the mature subtilisin BPN′.
- 26. The modified subtilase of claim 25, wherein the mutation comprises a substitution of the amino acid at position 252 with K, L, M, S or V.
- 27. The modified subtilase of claim 26, wherein the mutation comprises a substitution of the amino acid at position 252 with K.
- 28. The modified subtilase of claim 25, wherein the mutation comprises a substitution of the amino acid at position 255 with A, C, E, I, or S.
- 29. The modified subtilase of claim 25, wherein the mutation comprises a substitution of the amino acid at position 259 with C or H.
- 30. The modified subtilase of claim 25, comprising at least one further mutation at one or more other positions.
- 31. The modified subtilase of claim 30, wherein the one or more positions are selected from the group consisting of 27, 36, 57, 76, 97, 101, 104, 120, 123, 167, 170, 206, 218, 222, 224, 235 and 274.
- 32. The modified subtilase of claim 31, wherein the at least one further mutation is selected from the group consisting of K27R, *36D, S57P, N76D, G97N, S101G, V104A, V104N, V104Y, H120D, N123S, Y167A, Y167I, R170L, R170N, R170S, Q206E, N218S, M222A, M222S, T224S, K235L, and T274A.
- 33. The modified subtilase of claim 32, wherein the at least one further mutation is selected from the group consisting of K27R+V104Y+N123S+T274A, N76D+V104A, V104N+S101G, and any other combination of K27R, N76D, S101G, V104A, V104N, V104Y, N123S, and T274A.
- 34. The modified subtilase of claim 30, wherein the one or more positions are selected from is the group consisting of 129, 131, 133 and 194.
- 35. The modified subtilase of claim 34, wherein the at least one further mutation is selected from the group consisting of P129K, P131H, A133D, A133P, and A194P.
- 36. The modified subtilase of claim 25, wherein the subtilase is a sub-group I-S1 subtilase.
- 37. The modified subtilase of claim 36, wherein the subtilase is selected from the group consisting of subtilisin I168, subtilisin BPN′, subtilisin DY, and subtilisin Carlsberg.
- 38. The modified subtilase of claim 25, wherein the subtilase is a sub-group I-S2 subtilase.
- 39. The modified subtilase of claim 38, wherein the subtilase is selected from the group consisting of subtilisin 147, subtilisin 309, subtilisin PB92, and subtilisin YaB.
- 40. A composition comprising a modified subtilase of claim 25 and a surfactant.
- 41. The composition of claim 40, which additionally comprises an amylase, cellulase, cutinase, lipase, oxidoreductase, or another protease.
- 42. An isolated DNA sequence encoding a modified subtilase of claim 25.
- 43. An expression vector comprising an isolated DNA sequence of claim 42.
- 44. A microbial host cell transformed with an expression vector of claim 43.
- 45. The microbial host cell of claim 44, which is a bacterium.
- 46. A method for producing a modified subtilase, comprising
(a) culturing a microbial host cell of claim 44 under conditions conducive to the expression and secretion of the modified subtilase, and (b) recovering the modified subtilase.
- 47. A modified subtilase, comprising a mutation in an amino acid sequence of a subtilase, wherein the mutation comprises one of the following:
(a) 252H+255A+259C,

- 53. The modified subtilase of claim 49, wherein the one or more positions are selected from the group consisting of 129, 131, 133 and 194.
- 54. The modified subtilase of claim 53, wherein the at least one further mutation is selected from the group consisting of P129K, P131H, A133D, A133P, and A194P.
- 55. The modified subtilase of claim 47, wherein the subtilase is a sub-group I-S1 subtilase.

- 65. A method for producing a modified subtilase, comprising
(a) culturing a microbial host cell of claim 63 under conditions conducive to the expression and secretion of the modified subtilase, and (b) recovering the modified subtilase.
