Claims
- 1. An isolated and purified plant enzyme, wherein said enzyme adds a β1,2-linked xylose to the β-linked mannose on the N-linked oligosaccharides of storage glycoproteins.
- 2. The enzyme of claim 1, wherein said enzyme is β1,2-xylosyltransferase.
- 3. The enzyme of claim 1, wherein said enzyme releases peptides having the sequence selected from the group consisting of SEQ ID Nos. 1-5 upon digestion by Endo lys C.
- 4. The enzyme of claim 1, wherein said enzyme is obtained from soybean.
- 5. The enzyme of claim 4, wherein said enzyme is obtained from the microsomal fraction of soybean cells.
- 6. The enzyme of claim 1, wherein said enzyme has a purity of about 51,000-fold to near homogeneity compared to a solubilized enzyme fraction.
- 7. The enzyme of claim 1, wherein said enzyme is separated on SDS gels into 56 and 59 kDa bands.
- 8. An oligonucleotide useful for cloning the gene encoding the enzyme of claim 1, wherein said oligonucleotides are based on the peptide having the sequence selected from the group consisting of SEQ ID Nos. 1-5.
- 9. DNA encoding the enzyme of claim 1 selected from the group consisting of:
(a) isolated DNA which encodes a xylosyltransferase; (b) isolated DNA which hybridizes to isolated DNA of (a) above and which encodes a xylosyltransferase; and (c) isolated DNA differing from the isolated DNAs of (a) and (b) above in codon sequence due to the degeneracy of the genetic code, and which encodes a xylosyltransferase.
- 10. The DNA of claim 9, wherein said DNA having the sequence shown in SEQ ID No. 6.
- 11. An antibody directed against the enzyme of claim 1.
- 12. A method for localizing xylosyltransferase in a plant cell. comprising the steps of:
contacting said cell with the antibody of claim 11; and detecting localization of said antibody, wherein the localization of said antibody indicating the localization of said enzyme.
- 13. A method for determining distribution levels of xylosyltransferase in a plant, comprising the steps of:
contacting said plant with the antibody of claim 11; and detecting binding activity of said antibody, wherein a higher activity of said antibody indicating a higher level of said xylosyltransferase in said plant.
