Claims
- 1. A pyrrolidine represented by formula (3): ##STR18## wherein: represents a 3- to 15-membered mono- or fused cyclic hydrocarbon group, a pyridyl group or a morpholino group, each of which nay be substituted with a substituent(s) selected from the group consisting of a lower alkyl group, a lower alkoxy group, phenoxy, methylenedioxy, a halogen atom, hydroxy, mercapto, in alkylthio group, nitro and a mono- and di-lower alkylamino group;
- each of n and m represents an integer and the sum of n and m is an integer of 0 to 2;
- each of X and E represents oxygen, NR' (wherein R' is hydrogen or a lower alkyl group having 1 to 6 carbon atoms), sulfur, phenylene, CH.dbd.CH or CH.sub.2 ;
- p represents air integer of 1 to 3; and,
- Y2 represents a pyridyl group or a morpholino group, each of which may be substituted with a substituent(s) selected from the group consisting of a lower alkyl group, a lower alkoxy group, phenoxy, methylenedioxy, a haloen atom, hydroxy, mercapto, an alkyltio group, nitro and a mono- and di-lower-alkylamino group;
- or a salt thereof.
- 2. A compound according to claim 1 or a salt thereof, wherein R2--(X)n--(E)m--CO-- represents phenoxyacetyl, phenylthioacetyl, pyridylthioacetyl, pyridylcarbonyl, pyridylacetyl, morpholinomethylbenzyl, phenoxybenzoyl, naphthoyl, or cinnamoyl; and Y2 represents a monovalent group from pyridyl or morpholino.
- 3. A compound according to claim 1 or a salt thereof, wherein R2--(X)n--(E)m--CO-- represents pyridylthioacetyl or pyridylcarbonyl; and Y2 represents a monovalent group from pyridyl or morpholino.
- 4. A pyrrolidine represented by formula (4): ##STR19## wherein R3 represents a phenyl group which may be substituted with a substituent(s) selects from the group consisting of a lower alkyl group, a lower alkoxy group, phenoxy, methylenedioxy, a halogen atom, hydroxy, mercapto, an alkylthio group, nitro and a mono- and di-lower alkylamino group;
- p represents an integer of 1 to 3; and,
- Y2 represents a pyridyl group or a morpholino group, each of which may be substituted with a substituent(s) selected from the group consisting of a lower alkyl group, a lower alkoxy group, phenoxy, methylenedioxy, a halogen atom, hydroxy, mercapto, an alkylthio group, nitro and a mono- and di-lower alkylamino group;
- or a salt thereof.
- 5. A compound according to claim 4 or a salt thereof, wherein R3 represents a phenyl group which may be substituted with a substituent(s) selected from the group consisting of a lower alkyl group, a lower alkoxy group, phenoxy, methylenedioxy, a halogen atom, hydroxy, mercapto, an alkylthio group, nitro and a mono- and di-lower alkylamino group; and Y2 represents a monovalent group from pyridyl or morpholino.
- 6. A pyrrolidine represented by formula (1): ##STR20## wherein: R1 represents a pyridyl group or a morpholino group, each of which may be substituted with a substituent(s) selected from the group consisting of a lower alkyl group, a lower alkoxy group, phenoxy, methylenedioxy, a halogen atom, hydroxy, mercapto, an alkylthio group, nitro and a mono- and di-lower alkylamino group;
- each of n and m represents an integer and the sum of n and m is an integer of 0 to 2;
- each of X and E represents oxygen, NR' (wherein R' is hydrogen or a lower alkyl group having 1 to 6 carbon atoms), sulfur, phenylene, CH.dbd.CH or CH.sub.2 ;
- A represents an amino acid residue or an amino acid residue, which functional group may be optionally protected, or a glycine residue which may be substituted at the amino moiety thereof with a substituent selected from the group consisting of glycine, sarcosine, N-benzylycine, N-cyclohexylglycine, N-cyclohexylnmethylglycine, N-isopropylglycine and N-isobutylglycine; and
- Y1 represents a cycloalkyl group having 3 to 8 carbon atoms;
- or a salt thereof.
- 7. A compound according to claim 6 or a salt thereof, wherein R1--(X)n--(E)m--CO-- represents pyridylthioacetyl, pyridylacetyl, morpholinomethylbenzoyl, pyridylacryloyl or pyridylcarbonyl; and A represents phenylalanine, homophenylalanine, valine, leucine, isoleucine, alanine, .beta.-cyclohexylalanine; an optionally protected lysine, ornithine, arginine, serine, homoserine, threonine, methionine, aspartic acid, glutamic acid, proline, hydroxyproline, glycine, sarcosine, N-benzylglycine, N-cyclohexylglycine, N-cyclohexylmethylglycine, N-isopropylglycine, N-isobutylglycine or dihydroindole-2-carbonyl.
- 8. A compound according to claim 6 or a salt thereof, wherein R1--(X)n--(E)m--CO-- represents pyridylthioacetyl, pyridylacetyl, morpholinomethylbenzoyl, pyridylacryloyl or pyridylcarbonyl: and A represents phenylalanine, homophenylalanine, valine, leucime, isoleucine, N-benzylglycine, proline, .beta.-cyclohexylalanine or dihydroindole-2-carbonyl.
- 9. N-cyclohexyl-2-oxo-2-[2-(1-(N-(2-pyridylcarbonyl)phenylalanyl)pyrrolidinyl)]acetamide or a salt thereof.
- 10. A compound according to claim 8 or a salt thereof, wherein A represents phenylalanine, valine, N-benzylglycine, proline, .beta.-cyclohexylalanine or dihydroindole-2-carbonyl.
- 11. A pyrrolidine represented by formula (2): ##STR21## wherein: R2 represents a pyridyl group or a morpholino group, each of which may be substituted with a substituent(s) selected from the group consisting of a lower alkyl group, a lower alkoxy group, phenoxy, methylenedioxy, a halogen atom, hydroxy, mercapto, an alkylthio group, nitro and a mono- and di-lower alkylamino group;
- each of n and m represents an integer and the sum of n and m is an integer of 0 to 2;
- each of X and B represents oxygen, NR' (wherein R' is hydrogen or a lower alkyl group having 1 to 6 carbon atoms), sulfur, phenylene, CH.dbd.CH or CH.sub.2 ; and,
- Y1 represents a cycloalkyl group having 3 to 8 carbon atoms: or a salt thereof.
- 12. A pyrrolidine according to claim 11 or a salt thereof, wherein R2--(X)n--(E)m--CO-- represents pyridylthioacetyl, pyridylacetyl, morpholinomethuylbenzoyl, pyridylcarbonyl, or pyridylacryloyl.
- 13. A pyrrolidine according to claim 11 or a salt thereof, wherein R2--(X)n--(E)m--CO-- represents morpholinomethylbenzoyl, pyridylcarbonyl or pyridylacryloyl.
- 14. A pharmaceutical composition comprising a pyrrolidine or a salt thereof according to any one of claims 1, 4, 6, or 11 together with a suitable pharmaceutical carrier.
- 15. A method for inhibiting the activity of serine protease or thiol protease which comprises administering to a mammal an effective dose of the pyrrolidine derivative or a salt thereof according to any one of claims 1, 4, 6, or 11.
- 16. N-Cyclohexyl-2-oxo-2-[2-(1-4-(morpholinomethylbenzoyl)pyrrolidinyl)]acetamide or a salt thereof.
