The present disclosure relates to a quinazoline compound, a preparation method therefor, and a use thereof.
Inflammatory diseases have become a significant threat to global public health. Inflammation is closely related to the pathological process of various diseases, including many infectious and autoimmune diseases, malignant tumors, cardiovascular diseases, diabetes, and other chronic non-communicable diseases of significance. Autoimmune diseases refer to a class of diseases induced by the immune system attacking the body's own organs, tissues, or cells, causing damage. These primarily include psoriasis, rheumatoid arthritis (RA), systemic lupus erythematosus (SLE), and multiple sclerosis (MS), etc.
Inflammatory cytokines TNF-α, IL-6, and IL-1β are abundantly expressed in the serum and local lesion tissues of patients with autoimmune diseases. Inhibiting the formation of these three factors can effectively reduce the inflammatory response and reduce the damage to bodily tissues caused by inflammation. Hydroxychloroquine (HCQ) was introduced in 1944, initially gaining attention as an anti-malarial drug. It was later discovered that HCQ also has anti-inflammatory properties, and thus it has been used as an anti-inflammatory agent to treat certain inflammatory diseases. It is widely used in the treatment of systemic lupus erythematosus, rheumatoid arthritis, Sjogren's syndrome, porphyria cutanea tarda, etc.
Modifying HCQ to develop newer drugs with better activity has been a hot research topic in the field of medicinal chemistry.
The technical problem to be solved by the present disclosure is to provide a structurally novel quinazoline compound with anti-inflammatory effects. The compounds of the present disclosure exhibit good inhibitory effects on cytokines TNF-α, IL-6, and IL-1β.
The present disclosure provides a quinazoline compound of formula I, a pharmaceutically acceptable salt thereof, a solvate thereof, or a solvate of the pharmaceutically acceptable salt thereof:

The term “solvate” refers to a substance formed when a compound crystallizes with a solvent, which includes, but is not limited to, water, methanol, ethanol, etc. Solvates are classified into stoichiometric solvates and non-stoichiometric solvates.
The term “solvate of the pharmaceutically acceptable salt” refers to a substance formed by the combination of a compound with a pharmaceutically acceptable (relatively non-toxic, safe, and suitable for patient use) acid or base, and a solvent (including but not limited to water, methanol, ethanol, etc.). Here, the “pharmaceutically acceptable salt” carries the same meaning as defined above, and the solvent can be either stoichiometric or non-stoichiometric. The solvate of the pharmaceutically acceptable salt include, but is not limited to, hydrochloride monohydrate.
The term “therapeutically effective amount” refers to an amount of a compound administered to a patient that is sufficient to effectively treat a disease. The therapeutically effective amount will vary depending on the compound, type of disease, severity of the disease, patient's age, etc., but can be adjusted by those skilled in the art as required.
The term “pharmaceutical excipient” refers to the formulating agents and additives used in the manufacture of drugs and the compounding of prescriptions. They are substances contained in a pharmaceutical formulation, apart from the active ingredients. For specific reference, see the Pharmacopoeia of the People's Republic of China (2020 Edition) or the Handbook of Pharmaceutical Excipients (Raymond C Rowe, 2009).

The reagents and raw materials used in the present disclosure are commercially available.
The advantageous progressive effect of the present disclosure is that the quinazoline compound of the present disclosure is novel in structure and exhibits good inhibitory effects on cytokines TNF-α, IL-6, and IL-1β.
The present disclosure is further described through examples below, but is not limited to the scope of the examples provided. In the following examples, experimental methods without specified conditions are carried out according to conventional methods and conditions, or are selected according to the product instructions.
Under a hydrogen atmosphere at room temperature, 5-hydroxy-2-pentanone (5.0 g, 48.96 mmol) and ammonium formate (10.0 g, 158.58 mmol) were added to a mixture of palladium on carbon (1 g) in methanol (200 mL). The reaction mixture was stirred at room temperature for 48 hours. The reaction mixture was concentrated under reduced pressure, and the residual liquid was slurried with ethyl acetate (200 mL). The mixture was filtered and concentrated under reduced pressure to dryness to obtain the crude 4-amino-1-pentanol (4 g, yield of 79.22%). LC-MS: m/z 104.1 [M+H]+.
Under an argon atmosphere at room temperature, 4-amino-1-pentanol (4.0 g, 38.83 mmol), imidazole (5.28 g, 77.67 mmol), and tert-butyldiphenylchlorosilane (16.02 g, 58.25 mmol) were added to a solution of dichloromethane (200 mL). The reaction mixture was stirred at room temperature for 18 hours. After the reaction was completed, the reaction mixture was concentrated, and then water and ethyl acetate were added thereto. The mixture was extracted with ethyl acetate three times. The organic phase was dried, filtered, and concentrated. The residue was separated by column chromatography to obtain the product 5-((tert-butyldiphenylsilyl)oxy)pentan-2-amine (5.4 g, yield of 40.79%). LC-MS: m/z 342.2 [M+H]+.
The starting material 5-((tert-butyldiphenylsilyl)oxy)pentan-2-amine (3.8 g, 11.14 mmol) was dissolved in 40 mL of 1,4-dioxane. Pd2(dba)3 (50 mg), BINAP (40 mg), Cs2CO3 (7.25 g, 22.25 mmol), and 4,7-dichloroquinoline (2.0 g, 10.1 mmol) were added thereto, respectively. The reaction mixture was heated to 110° C. under an argon atmosphere and reacted for 18 hours. After the reaction was completed, the reaction mixture was concentrated. The residue was separated by column chromatography to obtain the product (4 g, yield of 78.72%). LC-MS: m/z 503.2 [M+H]+.
The starting material N-(5-((tert-butyldiphenylsilyl)oxy)pentan-2-yl)-7-chloroquinolin-4-amine (4.0 g, 7.95 mmol) was dissolved in 40 mL of methanol. Ammonium fluoride (2.0 g, 54.05 mmol) was added thereto, and the reaction mixture was heated to reflux under an argon atmosphere and reacted for 2 hours. After the reaction was completed, the reaction mixture was concentrated. The residue was separated by preparative liquid chromatography to obtain the product (1.6 g, yield of 76.19%). LC-MS: m/z 265.1 [M+H]+.
The starting material 4-((7-chloroquinolin-4-yl)amino)pentan-1-ol (2.0 g, 7.58 mmol) was dissolved in 40 mL of dichloromethane. Thionyl chloride (2.0 g, 16.81 mmol) and a catalytic amount of N,N-dimethylformamide (0.1 mL) were added thereto. The reaction mixture was reacted at room temperature under an argon atmosphere for 2 hours. After the reaction was completed, the reaction mixture was concentrated. The residue was quenched with sodium bicarbonate solution and extracted with ethyl acetate. The organic phase was dried, filtered, and concentrated to obtain the intermediate product (2.1 g, yield of 97.95%), which was used directly in the next reaction step. LC-MS: m/z 283.1 [M+H]+.
The starting material 7-chloro-N-(5-chloropentan-2-yl)quinolin-4-amine (80 mg, 0.28 mmol) was dissolved in 20 mL of acetonitrile. KI (10 mg), K2CO3 (100 mg, 0.73 mmol), and thiomorpholine-1,1-dioxide (150 mg, 1.11 mmol) were added thereto. The reaction mixture was heated to 60 to 70° C. under an argon atmosphere and reacted for 48 hours. After the reaction was completed, the reaction mixture was concentrated. The residue was separated by column chromatography to obtain the product (15 mg, yield of 13.89%). 1H NMR (400 MHz, CDCl3) δ 8.47 (d, J=5.6 Hz, 1H), 7.98 (d, J=1.8 Hz, 1H), 7.87 (d, J=9.0 Hz, 1H), 7.35 (dd, J=8.9, 2.0 Hz, 1H), 6.45 (d, J=5.7 Hz, 1H), 5.57 (s, 1H), 3.78 (dd, J=13.4, 6.8 Hz, 1H), 3.05 (d, J=6.1 Hz, 4H), 2.99 (d, J=5.9 Hz, 4H), 2.56 (t, J=6.9 Hz, 2H), 1.86-1.76 (m, 1H), 1.76-1.56 (m, 4H), 1.39 (d, J=6.3 Hz, 3H). LC-MS: m/z 382.1 [M+H]+.
To prepare the compounds in examples 2 to 12, the same method as described in example 1 was applied. Specific experimental data for these compounds are shown in Table 1.
The starting material 5-((tert-butyldiphenylsilyl)oxy)pentan-2-amine (2.8 g, 8.20 mmol) was dissolved in 40 mL of isopropanol. DIPEA (3.5 g, 27.08 mmol) and 4,7-dichloroquinazoline (2.0 g, 10.05 mmol) were then added thereto. The reaction mixture was heated to 90° C. under an argon atmosphere and reacted for 18 hours. After the reaction was completed, the reaction mixture was concentrated. The residue was subjected to preparative separation to obtain the product (3.1 g, yield of 75.01%). LC-MS: m/z 504.2 [M+H]+.
The starting material N-(5-((tert-butyldiphenylsilyl)oxy)pentan-2-yl)-7-chloroquinazolin-4-amine (3.1 g, 6.15 mmol) was dissolved in 40 mL of methanol. Ammonium fluoride (2.0 g, 54.05 mmol) was added thereto, and the reaction mixture was heated to reflux under an argon atmosphere and reacted for 2 hours. After the reaction was completed, the reaction mixture was concentrated. The residue was separated by preparative liquid chromatography to obtain the product (1.3 g, yield of 79.75%). LC-MS: m/z 266.1 [M+H]+.
The starting material 4-((7-chloroquinolin-4-yl)amino)pentan-1-ol (1.0 g, 3.76 mmol) was dissolved in 20 mL of dichloromethane. Thionyl chloride (1.0 g, 8.40 mmol) and a catalytic amount of N,N-dimethylformamide (0.1 mL) were added thereto. The reaction mixture was reacted at room temperature under an argon atmosphere for 2 hours. After the reaction was completed, the reaction mixture was concentrated. The residue was quenched with sodium bicarbonate solution and extracted with ethyl acetate. The organic phase was dried, filtered, and concentrated to obtain the intermediate product (1.0 g, yield of 93.66%), which was used directly in the next reaction step. LC-MS: m/z 284.1 [M+H]+.
The starting material 7-chloro-N-(5-chloropentan-2-yl)quinazolin-4-amine (100 mg, 0.35 mmol) was dissolved in 20 mL of acetonitrile. n-Bu4NBr (10 mg), K2CO3 (100 mg, 0.73 mmol), and imidazolidine-2,4-dione (100 mg, 1.00 mmol) were added thereto, respectively. The reaction mixture was heated to 60 to 70° C. under an argon atmosphere and reacted for 48 hours. After the reaction was completed, the reaction mixture was concentrated. The residue was separated by column chromatography to obtain the product (54 mg, yield of 44.12%). 1H NMR (400 MHZ, CDCl3) δ 8.62 (s, 1H), 7.81 (t, J=5.5 Hz, 2H), 7.41 (dd, J=8.8, 2.1 Hz, 1H), 6.13 (d, J=8.0 Hz, 1H), 5.74 (s, 1H), 4.66-4.52 (m, 1H), 4.01 (d, J=0.9 Hz, 2H), 3.68-3.56 (m, 2H), 1.90-1.80 (m, 2H), 1.72-1.60 (m, 2H), 1.32 (d, J=6.5 Hz, 3H). LC-MS: m/z 348.1 [M+H]+.
To prepare the compounds in example 14, the same method as described in example 13 was applied. Specific experimental data are shown in Table 2.
The starting material 4-((7-chloroquinazolin-4-yl)amino)pentan-1-ol (50 mg, 0.19 mmol) was dissolved in 20 mL of anhydrous tetrahydrofuran. PPh3 (280 mg, 1.07 mmol), DIAD (240 mg, 1.19 mmol), and pyrrolidin-2,5-dione (100 mg, 1.01 mmol) were added thereto, respectively. The reaction mixture was reacted at room temperature under an argon atmosphere for 48 hours. After the reaction was completed, the reaction mixture was concentrated. The residue was separated by column chromatography to obtain the product (57 mg, yield of 87.10%). 1H NMR (400 MHZ, CDCl3) δ 8.63 (s, 1H), 7.82 (d, J=2.0 Hz, 1H), 7.78 (d, J=8.8 Hz, 1H), 7.42 (dd, J=8.8, 2.1 Hz, 1H), 5.97 (d, J=7.9 Hz, 1H), 4.63-4.51 (m, 1H), 3.67-3.52 (m, 2H), 2.74 (s, 4H), 1.76-1.60 (m, 4H), 1.32 (d, J=6.5 Hz, 3H). LC-MS: m/z 348 [M+H]+.
At room temperature, the starting materials 2-(benzylamino)ethanol (17.5 g, 116 mmol), 5-chloropentan-2-one (28 g, 232 mmol), potassium carbonate (24 g, 174 mmol), and potassium iodide (1.9 g, 12 mmol) were added to N,N-dimethylformamide (300 mL). The reaction mixture was reacted overnight at 120° C. under an argon atmosphere. After N,N-dimethylformamide was evaporated until dryness, water was added thereto, and the mixture was extracted with dichloromethane three times. The organic phase was washed with saturated brine, dried over anhydrous sodium sulfate, filtered, and then subjected to rotary evaporation until dryness. The residue was purified by column chromatography to obtain a yellow oil, 5-(benzyl(2-hydroxyethyl)amino)pentan-2-one (23 g, 98 mmol, yield of 84.5%). LC-MS: m/z 236 [M+H]+.
The starting material 5-(benzyl(2-hydroxyethyl)amino)pentan-2-one (9 g, 38 mmol) was dissolved in methanol (200 mL). Ammonium acetate (17.7 g, 230 mmol) was added at room temperature and the reaction mixture was stirred for 30 minutes. Subsequently, sodium cyanoborohydride (5 g, 80 mmol) was added thereto, and the reaction mixture was stirred at room temperature overnight. After methanol was evaporated until dryness, 40% sodium hydroxide aqueous solution (200 mL) was added thereto, and the mixture was extracted three times with dichloromethane. The organic phase was washed with saturated brine, dried over anhydrous sodium sulfate, filtered, and then subjected to rotary evaporation until dryness. The residue was purified by column chromatography to obtain a yellow oil, 2-[4-aminopentyl(benzyl)amino]ethanol (2.45 g, 10.4 mmol, yield of 27.1%). LC-MS: m/z 237 [M+H]+.
The starting materials 2-[4-aminopentyl(benzyl)amino]ethanol (2.4 g, 10 mmol) and 4,7-dichloroquinazoline (2.26 g, 11.4 mmol) were added to N,N-diisopropylethylamine (7.5 g). The reaction mixture was stirred at 90° C. overnight. After N,N-diisopropylethylamine was evaporated until dryness, water was added thereto, and the mixture was extracted with dichloromethane three times. The organic phase was washed with saturated brine, dried over anhydrous sodium sulfate, filtered, and then subjected to rotary evaporation until dryness. The residue was purified by column chromatography to obtain a yellow oil, 2-(benzyl(4-((7-chloroquinazolin-4-yl)amino)pentyl)amino)ethanol (2.2 g, 5.5 mmol, yield of 54%). LC-MS: m/z 399 [M+H]+.
The starting materials 2-(benzyl(4-((7-chloroquinazolin-4-yl)amino)pentyl)amino)ethanol (2.2 g, 5.5 mmol) and imidazole (0.752 g, 11.0 mmol) were dissolved in anhydrous dichloromethane (30 mL). tert-Butyldimethylchlorosilane (1 g, 6.6 mmol) was added dropwise under an ice bath, and then the reaction mixture was stirred at room temperature overnight. The reaction mixture was washed with water three times, dried over anhydrous sodium sulfate, filtered, and then subjected to rotary evaporation until dryness. The residue was purified by column chromatography to obtain a white solid, N1-benzyl-N1-(2-((tert-butyldimethylsilyl)oxy)ethyl)-N4-(7-chloroquinazolin-4-yl)pentane-1,4-diamine (1.4 g, 2.7 mmol, yield of 50%). LC-MS: m/z 513 [M+H]+.
The starting material N1-benzyl-N1-(2-((tert-butyldimethylsilyl)oxy)ethyl)-N4-(7-chloroquinazolin-4-yl)pentane-1,4-diamine (1.4 g, 2.7 mmol) was dissolved in anhydrous dichloromethane (30 mL). 1-Chloroethyl chloroformate (0.51 g, 3.6 mmol) was added dropwise under an ice bath, and the reaction mixture was stirred at room temperature for 2 hours. The dichloromethane was evaporated, and the residue was added with methanol and refluxed for 2 hours. After the methanol was evaporated, the residue was purified by column chromatography to obtain a yellow oil, 2-((4-((7-chloroquinazolin-4-yl)amino)pentyl)amino)ethanol (0.388 g, 1.26 mmol, yield of 46%). LC-MS: m/z 309 [M+H]+.
The starting materials 2-((4-((7-chloroquinazolin-4-yl)amino)pentyl)amino)ethanol (65 mg, 0.21 mmol), 2-(chloromethyl)oxazole (30 mg, 0.26 mmol), and potassium carbonate (44 mg, 0.32 mmol) were added to acetonitrile (5 mL). The reaction mixture was stirred at 80° C. overnight. After the solvent was evaporated until dryness, water was added thereto, and the mixture was extracted three times with dichloromethane. The organic phase was washed with saturated brine, dried over anhydrous sodium sulfate, filtered, and then subjected to rotary evaporation until dryness. The residue was purified by a silica gel preparative plate to obtain a yellow oil, 2-((4-((7-chloroquinazolin-4-yl)amino)pentyl)(oxazol-2-ylmethyl)amino)ethan -1-ol (39 mg, 0.10 mmol, yield of 47.52%). 1H NMR (400 MHz, CDCl3) δ 8.62 (s, 1H), 7.85 (d, J=2.0 Hz, 1H), 7.82 (d, J=8.9 Hz, 1H), 7.63 (s, 1H), 7.42 (dd, J=8.8, 2.0 Hz, 1H), 7.08 (s, 1H), 6.00 (d, J=6.4 Hz, 1H), 4.58-4.47 (m, 1H), 3.90 (s, 2H), 3.65 (t, J=5.1 Hz, 2H), 2.79 (t, J=4.4 Hz, 2H), 2.73-2.64 (m, 2H), 1.85-1.80 (m, 2H), 1.71-1.62 (m, 3H), 1.34 (d, J=6.5 Hz, 3H). To prepare the compounds in examples 17 to 43, the same method as described in example 16 was applied. Specific experimental data are shown in Table 4.
At room temperature, potassium carbonate (2.07 g, 15 mmol) and 2-(5-bromopentyl)isoindole-1,3-dione (2.96 g, 10 mmol) were added to a solution of 2-(benzylamino)ethan-1-ol (1.51 g, 10 mmol) in acetonitrile (30 mL). The reaction mixture was heated to 80° C. and stirred for 16 hours. Subsequently, the reaction mixture was cooled to room temperature, filtered, and the filtrate was concentrated under reduced pressure until dryness to obtain a pale yellow liquid, 2-(5-(benzyl(2-hydroxyethyl)amino)pentyl)isoindole -1,3-dione (3.66 g, 10 mmol, yield of 100%). LC-MS: m/z 367.2 [M+H]+.
2-(5-(Benzyl(2-hydroxyethyl)amino)pentyl)isoindole-1,3-dione (3.66 g, 10 mmol) was dissolved in methanol (40 mL), then 10% Pd/C (0.9 g) was added thereto. The reaction was conducted under 1 atom of H2 pressure at 30° C. for 48 hours. LCMS showed that the reaction was completed, and the reaction mixture was filtered to remove Pd/C, and the mother liquor was concentrated under reduced pressure until dryness to obtain a pale yellow liquid, 2-(5-((2-hydroxyethyl)amino)pentyl)isoindole-1,3-dione (2.76 g, 10 mmol, yield of 99%), which was used directly in the next reaction step. LC-MS: m/z 277.2 [M+H]+.
2-(5-((2-Hydroxyethyl)amino)pentyl)isoindole-1,3-dione (2.76 g, 10 mmol) was dissolved in dichloromethane (30 mL). Di-tert-butyl dicarbonate (2.62 g, 12 mmol) and triethylamine (1.5 g, 15 mmol) were then added thereto. After the reaction mixture was stirred at room temperature for 16 hours, LCMS showed that the reaction was completed. The reaction mixture was concentrated under reduced pressure using a rotary evaporator, and the residue was purified by a Biotage preparative chromatography column (25 g silica gel, MeOH/DCM=0 to 5%, v/v) to obtain tert-butyl (5-(1,3-dioxoisoindol-2-yl)pentyl)(2-hydroxyethyl)carbonate (3.0 g, 7.958 mmol, yield of 79.58%) as a yellow oil. LC-MS: m/z 377.2 [M+H]+.
tert-Butyl (5-(1,3-dioxoisoindol-2-yl)pentyl)(2-hydroxyethyl)carbonate (3.0 g, 7.958 mmol) was dissolved in methanol (20 mL), then hydrazine hydrate (10 mL, 50%) was added thereto. After the reaction mixture was stirred at 30° C. for 6 hours, LCMS showed that the reaction was completed. The reaction mixture was concentrated using a rotary evaporator, and then 1 M NaOH solution was added to the residue. The mixture was extracted with dichloromethane (20 mL×3), and the organic phases were combined, dried over anhydrous Na2SO4, and concentrated until dryness to obtain tert-butyl (5-aminopentyl)(2-hydroxyethyl)carbonate (1.1 g, 4.453 mmol, yield of 55.96%) as a yellow oil. LC-MS: m/z 247.3 [M+H]+.
At room temperature, tert-butyl (5-aminopentyl)(2-hydroxyethyl)carbonate (1.1 g, 4.453 mmol) and DIPEA (1.29 g, 10.00 mmol) were added to a solution of 4,7-dichloroquinazoline (0.88 g, 4.453 mmol) in isopropanol (10 mL). The reaction mixture was stirred at 80° C. for 16 hours and then concentrated until dryness. The reaction mixture was added with saturated sodium bicarbonate aqueous solution (100 mL), then extracted with dichloromethane (100 mL×2). The organic phases were combined, dried over anhydrous sodium sulfate, and concentrated until dryness. The residue was purified by a silica gel column (eluent: petroleum ether: ethyl acetate =10:1) to obtain tert-butyl (5-((7-chloroquinazolin-4-yl)amino)pentyl)(2-hydroxyethyl)carbonate (1.2 g, 2.934 mmol, yield of 65.89%) as a yellow oil. LC-MS: m/z 409.2 [M+H]+.
At room temperature, a solution of hydrochloric acid in 1,4-dioxane (10 mL) was added to a solution of tert-butyl (5-((7-chloroquinazolin-4-yl)amino)pentyl)(2-hydroxyethyl)carbonate (1.2 g, 2.94 mmol) in methanol (5 mL). The reaction mixture was stirred at room temperature for 1.5 hours and then concentrated until dryness to obtain 2-((5-((7-chloroquinazolin-4-yl)amino)pentyl)amino)ethan-1-ol (0.905 g, 2.94 mmol, yield of 100%) as a yellow solid. LC-MS: m/z 309.2 [M+H]+.
2-((5-((7-Chloroquinazolin-4-yl)amino)pentyl)amino)ethan-1-ol (0.08 g, 0.2591 mmol) and methyl 4-(bromomethyl)benzoate (0.060 g, 0.2619 mmol) were dissolved in acetonitrile (3 mL), and potassium carbonate (0.04 g, 0.2899 mmol) was added thereto. The reaction mixture was stirred at 80° C. for 16 hours. The reaction mixture was cooled to room temperature, concentrated until dryness, and purified by a preparative thin-layer silica gel plate (eluent: dichloromethane: methanol =10:1) to obtain a yellow oily product, methyl 4-(((5-((7-chloroquinazolin-4-yl)amino)pentyl)(2-hydroxyethyl)amino)methyl)benzoate (23 mg, 0.05033 mmol, yield of 19.43%). 1H NMR (400 MHZ, d-DMSO) 8 8.45 (s, 1H), 8.32-8.39 (t, J=6.0 Hz, 1H), 8.27 (d, J=8.9 Hz, 1H), 7.85 (d, J=8.2 Hz, 2H), 7.70 (d, J=2.2 Hz, 1H), 7.55 (dd, J=8.8, 2.2 Hz, 1H), 7.43 (d, J=8.1 Hz, 2H), 4.33 (t, J=5.2 Hz, 1H), 3.84 (s, 3H), 3.63 (s, 2H), 3.51 (q, J=6.8 Hz, 2H), 3.45 (q, J=6.4 Hz, 2H), 2.44 (m, 4H), 1.67-1.54 (m, 2H), 1.53-1.40 (m, 2H), 1.38-1.28 (m, 2H). LC-MS: m/z 457.2 [M+H]+.
To prepare the compounds in examples 45 to 49, the same method as described in example 44 was applied. Specific experimental data are shown in Table 5.
The raw materials 4-chloro-7-fluoroquinazoline (0.9 g, 5 mmol) and 5-aminopentan -1-ol (0.5 g, 5 mmol) were dissolved in 10 mL of isopropanol. N,N-Diisopropylethylamine (0.7 g, 5 mmol) was added to the reaction mixture, and the reaction mixture was heated to 90° C. under an argon atmosphere and reacted for 6 hours. After the reaction was completed, the reaction mixture was concentrated. The residue was separated by column chromatography to obtain the product (622 mg, yield of 50%). LC-MS: m/z 250.1 [M+H]+.
The raw material 5-((7-fluoroquinazolin-4-yl)amino)pentan-1-ol (0.6 g, 2.4 mmol) was dissolved in 20 mL of anhydrous dichloromethane. Then, sulfonyl chloride (1.2 g, 10 mmol) and a catalytic amount of N,N-dimethylformamide (0.3 mL) were added thereto. The reaction mixture was reacted at room temperature under an argon atmosphere for 4 hours. After the reaction was completed, the reaction mixture was concentrated. The residue was adjusted to a pH of 8 with an aqueous solution of sodium bicarbonate and extracted with dichloromethane. The organic phase was dried, filtered, and concentrated. The residue was then separated by column chromatography to obtain the product (493 mg, yield of 76.5%). LC-MS: m/z 268.0 [M+H]+.
The raw material N-(5-chloropentyl)-7-fluoroquinazolin-4-amine (80 mg, 0.3 mmol) was dissolved in 5 mL of dry tetrahydrofuran. Imidazolidine-2,4-dione (60 mg, 0.6 mmol), potassium carbonate (130 mg, 0.9 mmol), and tetrabutylammonium hydrogen sulfate (20 mg, 0.06 mmol) were added thereto. The reaction mixture was heated to 60° C. under an argon atmosphere and reacted overnight. After the reaction was completed, the reaction mixture was concentrated. The residue was separated by a preparative silica gel plate to obtain the product (42 mg, yield of 42.4%). 1H NMR (400 MHZ, MeOD) 8 8.45 (s, 1H), 8.23 (dd, J=9.9, 5.8 Hz, 1H), 7.37-7.32 (m, 2H), 3.93 (s, 2H), 3.64 (t, J=7.1 Hz, 2H), 3.52 (t, J=7.1 Hz, 2H), 1.83-1.75 (m, 2H), 1.74-1.66 (m, 2H), 1.50-1.41 (m, 2H). LC-MS: m/z 332.0 [M+H]+.
To prepare the compounds in examples 51 to 77, the same method as described in example 50 was applied. Specific experimental data are shown in Table 6.
(1) Cell culture and inoculation: A stock solution of PMA at 1 mg/mL was diluted with serum-free RP1640 medium to prepare a 100 ng/mL PMA solution. Normal cultured THP-1 cells were centrifuged, resuspended in a complete medium (RP1640 medium containing 10% FBS), and counted. The cell density was adjusted to 2×106 cells/mL. An equal volume of 100 ng/mL PMA solution was then added, resulting in a cell density of 1×106 cells/mL and a final PMA concentration of 50 ng/mL. 100 μL of the cell suspension was seeded into each well of a 96-well cell culture plate with 1×105 cells per well. The 96-well cell culture plate was then incubated at 37° C. in a 5% CO2 atmosphere for 24 hours.
(2) Drug treatment of cells: On the second day, LPS was added to the complete medium to adjust the final concentration to 100 ng/mL, and the compound was prepared using this LPS-containing medium. The compound was added to the appropriate cell wells according to the prepared concentration gradient. The positive control group (LPS+) was a culture medium solution containing a final concentration of 100 ng/mL LPS and 0.1% DMSO without drugs, while the negative control group (LPS-) was a culture medium solution containing 0.1% DMSO without LPS. The volume in each well was 100 μL.
(3) Test sample collection: The mixture was centrifuged at 1000 rpm for 5 minutes and the cell supernatant and cell precipitate were collected separately. The cell supernatant was used to detect TNF-α and IL-6, and the cell precipitate was collected for IL-1β detection. After collecting the cell supernatant, the remaining liquid in the wells was discarded and 100 μL of cell lysis solution (20 mM Tris 8.0; 2 mM EDTA; 0.5% Triton X-100) was added to each well for complete lysis.
(4) Sample detection: The enzyme-linked immunosorbent assay (ELISA) was used to detect the content of each cytokine. The specific steps were performed according to the instructions provided in the corresponding kit manuals. The catalog numbers of kits are as follows:

In this assay, the compounds of the present disclosure exhibited IC50 values below 2000 μM. Some compounds showed IC50 values below 10 μM. Tables 7 and 8 provide the specific IC50 values of the compounds of the present disclosure tested in this assay.
Although the specific embodiments of the present disclosure have been described above, those skilled in the art should understand that these are merely illustrative examples. Various changes or modifications can be made to these embodiments without departing from the principles and essence of the present disclosure.
