Claims
- 1. A method for detecting anti-adenovirus antibody in a sample, comprising (a) contacting the sample with an antigen that specifically binds to anti-adenovirus antibody under conditions that permit formation of a stable antibody antigen complex, wherein the antigen is adsorbed on a membrane, and wherein the membrane is contacted with an absorbent; and (b) detecting any complex formed.
- 2. The method of claim 1, wherein said sample comprises whole blood.
- 3. The method of claim 1, wherein said membrane is nitrocellulose paper.
- 4. The method of claim 1, wherein said antigen comprises purified adenovirus.
- 5. The method of claim 1, wherein said antigen comprises an adenovirus polypeptide comprising an epitope recognized by anti-adenovirus antibody.
- 6. The method of claim 1, wherein said membrane and absorbent are housed in a sample well.
- 7. The method of claim 1, wherein said detecting is achieved with a gold-conjugated polypeptide.
- 8. The method of claim 1, wherein said detecting is achieved with an enzyme-linked polypeptide.
- 9. The method of claim 1, wherein said membrane is nitrocellulose paper, said antigen is adenovirus-5, said absorbent is a cellulose-based material, said sample is serum or plasma, and said detecting is achieved with gold-conjugated protein A.
- 10. A method for semi-quantitating anti-adenovirus antibody in a sample, comprising (a) contacting the sample with an antigen that specifically binds to anti-adenovirus antibody under conditions that permit formation of a stable antibody antigen complex, wherein the antigen is adsorbed on a membrane, and wherein the membrane is contacted with an absorbent; (b) detecting any complex formed; and (c) comparing a detectable signal arising from the antigen-antibody complex, if any, to a detectable signal of a reference sample, whereby anti-adenovirus antibody is semi-quantitated.
- 11. The method of claim 10, wherein said sample comprises whole blood.
- 12. The method of claim 10, wherein said membrane is nitrocellulose paper.
- 13. The method of claim 10, wherein said antigen comprises purified adenovirus.
- 14. The method of claim 10, wherein said antigen comprises an adenovirus polypeptide comprising an epitope recognized by anti-adenovirus antibody.
- 15. The method of claim 10, wherein said membrane and absorbent are housed in a sample well.
- 16. The method of claim 10, wherein said detecting is achieved with a gold-conjugated polypeptide.
- 17. The method of claim 10, wherein said detecting is achieved with an enzyme-linked polypeptide.
- 18. The method of claim 10, wherein said reference sample comprises human IgG.
- 19. The method of claim 18, wherein the concentration of said human IgG is adjusted to provide a detection signal that corresponds to 12 μg/mL of said anti-adenovirus antibody in said sample.
- 20. The method of claim 10, wherein said membrane is nitrocellulose paper, said antigen is adenovirus-5, said absorbent is a cellulose-based material, said sample is serum or plasma, said reference sample is human IgG adjusted to provide a detection signal that corresponds to 12 μg/mL of said anti-adenovirus antibody in said sample and said detecting is achieved with gold-conjugated protein A.
- 21. A kit for detection or semi-quantitation of anti-adenovirus antibody in a sample, comprising a membrane, an absorbent, and an antigen that specifically binds to anti-adenovirus antibody, said kit in suitable packaging.
- 22. The kit of claim 21, wherein the membrane and the absorbent are contacted with each other.
- 23. The kit of claim 21, wherein the antigen is adsorbed on the membrane.
- 24. The kit of claim 21, further comprising at least one reference sample.
- 25. The kit of claim 21, further comprising at least one wash buffer.
- 26. The kit of claim 21, further comprising a detection molecule.
- 27. The kit of claim 21, wherein said membrane is nitrocellulose paper, said absorbent is a cellulose-based material, and said antigen is adenovirus.
- 28. The kit of claim 23, wherein said membrane is nitrocellulose paper, said absorbent is a cellulose-based material, and said antigen is adenovirus-5.
- 29. The kit of claim 28, wherein said nitrocellulose paper and cellulose-based material are housed in a sample well.
- 30. The kit of claim 28, further comprising wash buffer.
- 31. The kit of claim 30, wherein said wash buffer is HEPES buffer.
- 32. The kit of claim 28, further comprising gold-conjugated protein A.
- 33. The kit of claim 28, further comprising a reference sample comprising human IgG.
- 34. The kit of claim 33 wherein the concentration of said human IgG is adjusted to provide a detection signal that corresponds to about 12 μg/mL of said anti-adenovirus antibody in said sample.
- 35. A kit for detection or semi-quantitation of anti-adenovirus antibody in a sample, comprising nitrocellulose paper and a cellulose-based material, wherein the nitrocellulose paper and cellulose-based material are housed in a sample well, whole adenovirus-5 adsorbed on said nitrocellulose paper, HEPES buffer, gold-conjugated protein A, and human IgG, said kit in suitable packaging.
- 36. The kit of claim 35 wherein the concentration of said human IgG is adjusted to provide a detection signal that corresponds to about 12 μg/mL of said anti-adenovirus antibody in said sample.
