Claims
- 1. A process for the preparation of cryptates consisting essentially of subjecting a compound of the formula: ##STR20## in which R.sub.1 is a C.sub.1 -C.sub.10 alkylene group, the ring of the formula ##STR21## is a macrocycle of the formula: ##STR22## where n=0 or 1, R' is selected from the group consisting of hydrogen, --CO--NH--Y--Z, or --COOR" in which R" is C.sub.1 -C.sub.10 alkyl;
- to aminolysis by reaction with an amine of the formula NH.sub.2 --Y--Z,
- wherein Y is a divalent organic radical selected from the group consisting of (1) linear or branched C.sub.1 -C.sub.20 alkylene groups optionally containing at least one double bond and optionally being interrupted by one or more heteroatoms selected from the group consisting of O, N, S and P, (2) C.sub.5 -C.sub.8 cycloalkylene groups optionally substituted by an alkyl, aryl or sulfonate group; and (3) C.sub.6 -C.sub.14 arylene groups optionally substituted by an alkyl, aryl or sulfonate group; and
- Z is a functional group capable of bonding covalently with a biological substance.
- 2. A process for the preparation of cryptates consisting essentially of (a) reacting the halogen compound 9: ##STR23## where R.sub.2 is selected from the group consisting of H and CH.sub.3, and X is a halide ion, with the macrocycle ##STR24## in which the ring of the formula ##STR25## is a macrocycle of the formula: ##STR26## or where n=0 or 1, R' is selected from the group consisting of hydrogen, --CO--NH--Y--Z, or --COOR" in which R" is C.sub.1 -C.sub.10 alkyl;
- in solution in an anhydrous organic solvent and at the reflux temperature of the said solvent, to form the macropolycyclic complex 10:
- and (b) contacting the product of step (a) with a halogen compound of the formula XYZ,
- wherein Y is a divalent organic radical selected from the group consisting of (1) linear or branched C.sub.1 -C.sub.20 alkylene groups optionally containing at least one double bond and optionally being interrupted by one or more heteroatoms selected from the group consisting of O, N, S and P, (2) C.sub.5 -C.sub.8 cycloalkylene groups optionally substituted by an alkyl, aryl or sulfonate group; and (3) C.sub.6 -C.sub.14 arylene groups optionally substituted by an alkyl, aryl or sulfonate group; and
- Z is a functional group capable of bonding covalently with a biological substance;
- and X is a halide ion.
- 3. A process for preparing a compound of the formula ##STR27## where R.sub.1 is a C.sub.1 -C.sub.10 alkylene group, the ring of the formula ##STR28## is a macrocycle of the formula: ##STR29## consisting essentially of contacting an amine of the formula ##STR30## where R" is a C.sub.1 -C.sub.10 alkyl group, with a halogen derivative of the formula ##STR31## in which R' is selected from the group consisting of hydrogen --CO--NH--Y--Z, or --COOR"' in which R"' is C.sub.1 -C.sub.10 alkyl; wherein Y and Z are as defined in claim 1 and X is a halide ion.
- 4. The process of claim 3 wherein the molar ratio of halogen compound to amine is about 2:1.
- 5. The process of claim 3 wherein said reaction is carried out in an anhydrous organic solvent at the reflux temperature of the solvent.
- 6. The process of claim 3 wherein said reaction is carried out in the presence of a base.
- 7. The process of claim 1 wherein said aminolysis reaction is carried out under a nitrogen atmosphere.
- 8. The process of claim 1 wherein said aminolysis reaction is carried out at room temperature.
- 9. The process of claim 7 wherein said aminolysis reaction is carried out at room temperature.
- 10. The process of claim 2 wherein said halogen compound 9 and said macrocycle are contacted in an anhydrous organic solvent at reflux temperature.
- 11. The process of claim 2 wherein said halogen compound 9 and said macrocycle are contacted in the presence of a base.
