Claims
- 1. Anti CK-M.sub.T antibody which can inhibit CK-M.sub.T subunit activity, but not inhibit CK-M.sub.S subunit activity, wherein said antibody can inhibit substantially 100% of CK-MM.sub.3 isoform activity and about 52% of CK-MM.sub.2 isoform activity, but not inhibit CK-MM.sub.1 isoform activity.
- 2. A reagent for measuring creatine kinase activity comprising an antibody, wherein said antibody inhibits creatine kinase M.sub.T subunit, but does not inhibit creatine kinase M.sub.S subunit and wherein said antibody can inhibit substantially 100% of CK-MM.sub.3 isoform activity and about 52% of CK-MM.sub.2 isoform activity, but not inhibit CK-MM.sub.1 isoform activity.
- 3. The reagent according to claim 1 wherein said antibody is obtained from a hybridoma cell strain which can produce a monoclonal antibody inhibiting creatine kinase (CK) M.sub.T activity.
- 4. The reagent according to claim 1 being composed of two packages, one of which comprises 0.1 to 40 unit/ml of glucokinase (GlcK) or hexokinase (HK), 0.1 to 40 unit/ml of glucose-6 phosphate dehydrogenase (G6PDH), 0.001 to 5 mg/ml of M.sub.T subunit inhibiting antibody (Ab), 0.1 to 20 mM of adenosine diphosphate (ADP), 0.05 to 20 mM of nicotinamide adenine dinucleotide (NAD.sup.+) or nicotinamide adenine dinucleotide phosphate (NADP.sup.+), 1 to 200 mM of glucose, 0.2 to 20 mM of adenosine monophosphate (AMP), 0.001 to 0.1 mM of diadenosine pentophosphate (Ap5A), 0.5 to 50 mM of N-acetylcysteine (NAC) or another thiol compound, 0.5 to 30 mM of magnesium salts, 0.5 to 50 mM of sodium azide, 0.1 to 20 mM of ethylenediamine tetraacetate (EDTA) and 5 to 50 mM of a buffer solution (pH 6.7), and the other reagent comprises 2 to 70 mM of creatine phosphate (CrP), 0.5 to 50 mM of sodium azide and 5 to 500 mM of a buffer solution.
- 5. A method for measuring creatine kinase activity comprising determining an inhibition of creatine kinase of body fluids using a reagent for measuring creatine kinase activity, wherein said reagent comprises an antibody which inhibits creatine kinase M.sub.T subunit, but does not inhibit creatine kinase M.sub.S subunit and wherein said antibody can inhibit substantially 100% of CK-MM.sub.3 isoform activity and about 52% of CK-MM.sub.2 isoform activity, but not inhibit CK-MM.sub.1 isoform activity.
- 6. The method according to claim 4 wherein said antibody is obtained from a hybridoma cell strain which can produce a monoclonal antibody inhibiting creatine kinase (CK) M.sub.T activity.
- 7. The method according to claim 4 wherein said reagent is composed of two packages, one of which comprises 0.1 to 40 unit/ml of glucokinase (Glck) or hexokinase (HK), 0.1 to 40 unit/ml of glucose-6 phosphate dehydrogenase (G6PDH), 0.001 to 5 mg/ml of M.sub.T subunit inhibiting antibody (Ab), 0.1 to 20 mM of adenosine diphosphate (ADP), 0.05 to 20 mM of nicotinamide adenine dinucleotide (NAD.sup.+) or nicotinamide adenine dinucleotide phosphate (NADP.sup.+), 1 to 200 mM of glucose, 0.2 to 20 mM of adenosine monophosphate (AMP), 0.001 to 0.1 mM of diadenosine pentophosphate (Ap5A), 0.5 to 50 mM of N-acetylcysteine (NAC) or another thiol compound, 0.5 to 30 mM of magnesium salts, 0.5 to 50 mM of sodium azide, 0.1 to 20 mM of ethylenediamine tetraacetate (EDTA) and 5 to 50 mM of a buffer solution (pH 6.7), and the other reagent comprises 2 to 70 mM of creatine phosphate (CrP), 0.5 to 50 mM of sodium azide and 5 to 500 mM of a buffer solution.
