Claims
- 1. A dry immunoassay analytical element for assaying a ligand, comprising a support bearing:
- (a) a label zone comprising an enzyme labeled ligand or an enzyme labeled receptor;
- (b) a spreading zone;
- (c) a receptor zone comprising a fixed concentration of an immobilized receptor for the ligand; and
- (d) a gravure zone comprising a diaryl telluride having the structure of formula (I):
- Ar.sup.1 --Te--Ar.sup.2 (I)
- wherein Ar.sup.1 and Ar.sup.2 are the same or different aryl or heteroaryl groups selected from the group consisting of: ##STR4## wherein X is O, S, Se, or Te, and R.sup.11, R.sup.12, R .sup.13, R.sup.21, R.sup.22, R.sup.23, R.sup.31, R.sup.32, R.sup.33, R.sup.41, R.sup.42 and R.sup.43 are the same or different and each selected from the group consisting of hydrogen, alkyl having 1-5 carbon atoms, OH, OR.sup.1, SH, NH.sub.2, NHR.sup.1, NR.sup.1.sub.2, NR.sup.1 R.sup.2, CO.sub.2 H or its salts, CO.sub.2 R.sup.1, SO.sub.3 H and its salts, PO.sub.3 H.sub.2 and its salts, and SR.sup.1 wherein R.sup.1 and R.sup.2 are different and each selected from the group consisting of alkyl having a carbon chain of 1 to 14 carbon atoms optionally carrying one or several hydrophilic groups, phenyl, and substituted phenyl;
- R.sup.14, R.sup.15, R.sup.24, and R.sup.25 are the same or different and each selected from the group consisting of hydrogen, alkyl having 1-5 carbon atoms, alkoxy having 1-5 carbon atoms, CO.sub.2 H or its salts, CO.sub.2 R.sup.1, SO.sub.3 H and its salts, PO.sub.3 H.sub.2 and its salts.
- 2. The element according to claim 1 wherein the diaryl telluride is selected from the group consisting of: ##STR5##
- 3. The element according to claim 1 further comprising a vanadyl salt.
- 4. The element according to claim 2 further comprising a vanadyl salt.
- 5. The element according to claim 1 wherein a peroxidase or a derivative of peroxidase is the label in the labeled ligand or labeled receptor.
- 6. The element according to claim 5 wherein the peroxidase is horseradish peroxidase or a derivative of horseradish peroxidase.
- 7. The element according to claim 1 wherein the gravure zone is contiguous with the spreading zone surface that provides contact with a liquid sample.
- 8. The element according to claim 7 wherein the diaryl telluride is selected from the group consisting of:
- 9. The element according to claim 8 further comprising a vanadyl salt.
- 10. A method for assaying an immunologically reactive ligand in an aqueous sample, comprising the steps of: (1) providing a dry immunoassay analytical element comprising a support bearing:
- (a) a label zone comprising an enzyme labeled ligand or an enzyme labeled receptor;
- (b) a spreading zone;
- (c) a receptor zone comprising a fixed concentration of an immobilized receptor for the ligand;
- (d) a gravure zone comprising a diaryl telluride having the structure of formula (I):
- Ar.sup.1 --Te--Ar.sup.2 (I)
- wherein Ar.sup.1 and Ar.sup.2 are the same or different aryl or heteroaryl groups selected from the group consisting of: ##STR6## wherein X is O, S, Se, or Te, and R.sup.11, R.sup.12, R.sup.13, R.sup.21, R.sup.22, R.sup.23, R.sup.31, R.sup.32, R.sup.33, R.sup.41, R.sup.42 and R.sup.43 are the same or different and each selected from the group consisting of hydrogen, alkyl having 1-5 carbon atoms, OH, OR.sup.1, SH, NH.sub.2, NHR.sup.1, NR.sup.1.sub.2, NR.sup.1 R.sup.2, CO.sub.2 H or its salts, CO.sub.2 R.sup.1, SO.sub.3 H and its salts, PO.sub.3 H.sub.2 and its salts, and SR.sup.1 wherein R.sup.1 and R.sup.2 are different and each selected from the group consisting of alkyl having a carbon chain of 1 to 14 carbon atoms optionally carrying one or several hydrophilic groups, phenyl, and substituted phenyl;
- R.sup.14, R.sup.15, R.sup.24, and R.sup.25 are the same or different and each selected from the group consisting of hydrogen, alkyl having 1-5 carbon atoms, alkoxy having 1-5 carbon atoms, CO.sub.2 H or its salts, CO.sub.2 R.sup.1, SO.sub.3 H and its salts, PO.sub.3 H.sub.2 and its salts;
- (2) contacting a finite area of the spreading zone of the element with the sample thereby forming (a) an immobilized ligand-receptor complex, (b) an immobilized enzyme labeled ligand-receptor complex, or (c) a mixture of (a) and (b), or an immobilized receptor-ligand-labeled receptor complex;
- (3) contacting the finite area with a substrate solution for the enzyme label thereby catalyzing the development of a signal; and
- (4) determining the concentration of the ligand by detecting the signal.
- 11. The method according to claim 10 wherein the diaryl telluride is selected from the group consisting of: ##STR7##
- 12. The method according to claim 10 wherein the element further comprises a vanadyl salt.
- 13. The method according to claim 11 wherein the element further comprises a vanadyl salt.
- 14. The method according to claim 10 wherein a peroxidase or a derivative of peroxidase is the label in the labeled ligand or labeled receptor.
- 15. The method according to claim 14 wherein the peroxidase is horseradish peroxidase or a derivative of horseradish peroxidase.
- 16. The method according to claim 15 wherein the signal is a calorimetric signal.
- 17. The method according to claim 10 wherein the gravure zone is contiguous with the spreading zone surface that provides contact with the aqueous sample.
- 18. The method according to claim 17 wherein the diaryl telluride is selected from the group consisting of:
- 19. The method according to claim 18 wherein the element further comprises a vanadyl salt.
- 20. A dry immunoassay analytical element for assaying a ligand, comprising a support bearing: (a) a spreading zone;
- (b) a receptor zone comprising a fixed concentration of an immobilized receptor for the ligand; and
- (c) a gravure zone comprising a labeled ligand and a diaryl telluride having the structure of formula (I):
- Ar.sup.1 --Te--Ar.sup.2 (I)
- wherein Ar.sup.1 and Ar.sup.2 are the same or different aryl or heteroaryl groups selected from the group consisting of: ##STR8## wherein X is O, S, Se, or Te, and R.sup.11, R.sup.12, R.sup.13, R.sup.21, R.sup.22, R.sup.23, R.sup.31, R.sup.32, R.sup.33, R.sup.41, R.sup.42 and R.sup.43 are the same or different and each selected from the group consisting of hydrogen, alkyl having 1-5 carbon atoms, OH, OR.sup.1, SH, NH.sub.2, NHR.sup.1, NR.sup.1.sub.2, NR.sup.1 R.sup.2, CO.sub.2 H or its salts, CO.sub.2 R.sup.1, SO.sub.3 H and its salts, PO.sub.3 H.sub.2 and its salts, and SR.sup.1 wherein R.sup.1 and R.sup.2 are different and each selected from the group consisting of alkyl having a carbon chain of 1 to 14 carbon atoms optionally carrying one or several hydrophilic groups, phenyl, and substituted phenyl;
- R.sup.14, R.sup.15, R.sup.24, and R.sup.25 are the same or different and each selected from the group consisting of hydrogen, alkyl having 1-5 carbon atoms, alkoxy having 1-5 carbon atoms, CO.sub.2 H or its salts, CO.sub.2 R.sup.1, SO.sub.3 H and its salts, PO.sub.3 H.sub.2 and its salts.
- 21. A method for assaying an immunologically reactive ligand in an aqueous sample, comprising the steps of:
- (1) providing a dry immunoassay analytical element comprising a support bearing:
- (a) a spreading zone;
- (b) a receptor zone comprising a fixed concentration of an immobilized receptor for the ligand; and
- (c) a gravure zone comprising a labeled ligand and a diaryl telluride having the structure of formula (I):
- Ar.sup.1 13 Te--Ar.sup.2 (I)
- wherein Ar.sup.1 and Ar.sup.2 are the same or different aryl or heteroaryl groups selected from the group consisting of: ##STR9## wherein X is O, S, Se, or Te, and R.sup.11, R.sup.12, R.sup.13, R.sup.21, R.sup.22, R.sup.23, R.sup.31, R.sup.32, R.sup.33, R.sup.41, R.sup.42 and R.sup.43 are the same or different and each selected from the group consisting of hydrogen, alkyl having 1-5 carbon atoms, OH, OR.sup.1, SH, NH.sub.2, NHR.sup.1, NR.sup.1 .sub.2, NR.sup.1 R.sup.2, CO.sub.2 H or its salts, CO.sub.2 R.sup.1, SO.sub.3 H and its salts, PO.sub.3 H.sub.2 and its salts, and SR.sup.1 wherein R.sup.1 and R.sup.2 are different and each selected from the group consisting of alkyl having a carbon chain of 1 to 14 carbon atoms optionally carrying one or several hydrophilic groups, phenyl, and substituted phenyl;
- R.sup.14, R.sup.15, R.sup.24, and R.sup.25 are the same or different and each selected from the group consisting of hydrogen, alkyl having 1-5 carbon atoms, alkoxy having 1-5 carbon atoms, CO.sub.2 H or its salts, CO.sub.2 R.sup.1, SO.sub.3 H and its salts, PO.sub.3 H.sub.2 and its salts;
- (2) contacting a finite area of the spreading zone of the element with the sample thereby forming (a) an immobilized ligand-receptor complex, (b) and immobilized enzyme labeled ligand-receptor complex, or (c) a mixture of (a) and (b), or an immobilized receptor-ligand-labeled receptor complex;
- (3) contacting the finite area with a substrate solution for the enzyme label thereby catalyzing development of a signal; and
- (4) determining the concentration of the ligand by detecting the signal.

This application is a continuation-in-part of Ser. No. 08/476,155, now U.S. Pat. No. 5,686,254 filed on Jun. 7, 1995 in the name of Margaret E. Logan et al. and entitled Reduction in First Slide Bias and Improved Enzyme Stability by the Incorporation of Diaryl Tellurides in Thin-Film Immunoassay Elements.
