Claims
- 1. A process for screening a library to determine whether or not said library contains at least one compound that binds to a target molecule with at least a predetermined binding affinity, comprising:
- (a) contacting a library of compounds with a target molecule said compounds being selected from the group consisting of polymers and non-polymers and when said compound is a polymer said compound is other than an oligonucleotide, said contacting being a repeated contacting without amplification and under conditions that permit binding to the target molecule those compounds in the library which bind to the target molecule with at least the predetermined binding affinity and that eliminate from the library those compounds which do not bind to the target molecule with at least the predetermined binding affinity, said repeated contacting being with compounds of the library which have not been eliminated; and
- (b) determining during said contacting whether there is at least one of (i) a decrease in the percentage of compounds in the library which do not bind to the target molecule, or (ii) an increase in the percentage of compounds in the library which bind to the target molecule, with a change in one of said percentages indicating that said library contains at least one compound that binds to the target molecule with at least the predetermined binding affinity.
- 2. The process of claim 1 wherein the repeated contacting is a reiterative procedure involving a plurality of rounds and the determining of step (b) is effected by determining the percentage of compounds in the library which bind to the target molecule in at least two of the rounds and comparing said percentages to determine whether there is an increase in the percentage of compounds which bind to the target molecule.
- 3. The process of claim 1 wherein the repeated contacting is effected by dialysis.
- 4. The process of claim 1 wherein said target molecule is a protein.
- 5. The process of claim 1 wherein said repeated contacting comprises recovering compounds which bind to the target molecule and recontacting the recovered compounds with the target molecule.
- 6. The process of claim 1 wherein the compound is a polymer.
- 7. The process of claim 6 wherein the monomeric units are amino acids and the polymer is a peptide.
- 8. The process of claim 1 wherein the compounds are labeled.
- 9. The process of claim 1 wherein in step (b) a decrease in the percentage of compounds in the library which do not bind to the target molecule is determined by measuring the percentage of compounds present in the library which do not bind to the target molecule at least twice during said repeated contacting, and comparing said percentage to determine a decrease in said percentage.
