Claims
- 1. A plasmid for the production of a polypeptide analog of an animal growth hormone having substantially the same amino acid sequence as, and the biological activity of, the corresponding naturally-occurring animal growth hormone which upon introduction into a suitable Escherichia coli host cell containing the thermolabile repressor cI renders the host cell capable, upon increasing the temperature of the Escherichia coli host cell to a temperature at which the cI repressor is inactivated, of effecting expression of DNA encoding the polypeptide analog of the animal growth hormone consisting essentially of:
- a double-stranded DNA molecule which comprises in 5' to 3' order the following:
- a DNA sequence which includes the promoter and operator P.sub.L O.sub.L from .lambda. bacteriophage;
- an N utilization site selected from the group consisting of Nut.sub.L and Nut.sub.R for binding antiterminator N protein produced by the host cell;
- a DNA sequence which contains a mutant cII ribosomal binding site from .lambda. bacteriophage having the sequence: ##STR14## for rendering the mRNA transcript of the DNA encoding the polypeptide analog of the animal growth hormone capable of binding to ribosomes within the Escherichia coli host cell;
- an ATG initiation codon;
- DNA encoding the polypeptide analog of the animal growth hormone in phase with the ATG initiation codon;
- a DNA sequence which contains a T.sub.1 T.sub.2 rRNA termination sequence located less than about 100 base pairs from the 3' end of the DNA encoding the polypeptide analog of animal growth hormone;
- and which additionally includes a DNA sequence which contains the origin of replication from the bacterial plasmid pBR322 which is capable of autonomous replication in the Escherichia coli host cell and a DNA sequence which contains the fragment designated cI.sup.434, such fragment including the gene for the cI.sup.434 repressor protein and its associated promoter and operator and the distance between the 3' end of the P.sub.L O.sub.L promoter and operator sequence from .lambda. bacteriophage and the 5' end of the N utilization site being less than about 80 base pairs and the distance between the 3' end of the N utilization site and the 5' end of the ribosomal binding site being less than about 300 base pairs.
- 2. A plasmid of claim 1, wherein the T.sub.1 T.sub.2 rRNA transcription termination sequence is less than about 20 base pairs from the 3' end of the DNA encoding the polypeptide analog of the animal growth hormone.
- 3. A plasmid of claim 1, wherein the CI.sup.434 fragment is located after the 3' end of the T.sub.1 T.sub.2 rRNA transcription termination sequence.
- 4. A plasmid of claim 1 which additionally comprises a DNA sequence which contains a gene encoding a selectable or identifiable phenotypic trait which is manifested when the plasmid is present in the Escherichia coli host cell.
- 5. A plasmid of claim 4, wherein the phenotypic trait is drug resistance.
- 6. A plasmid of claim 5, wherein the drug resistance is resistance to ampicillin or tetracycline.
- 7. A circular, closed plasmid of claim 1.
- 8. A plasmid of claim 1, wherein the DNA encoding the polypeptide analog of the animal growth hormone encodes a polypeptide analog of bovine growth hormone.
- 9. A host plasmid system for production of a polypeptide analog of an animal growth hormone comprising
- a plasmid for the production of a polypeptide analog of an animal growth hormone having substantially the same amino acid sequence as, and the biological activity of, the corresponding naturally-occurring animal growth hormone which upon introduction into a suitable Escherichia coli host cell containing the thermolabile repressor cI renders the host cell capable, upon increasing the temperature of the Escherichia coli host cell to a temperature at which the cI repressor is inactivated, of effecting expression of DNA encoding the polypeptide analog of the animal growth hormone consisting essentially of:
- a double-stranded DNA molecule which comprises in 5' to 3' order the following:
- a DNA sequence which includes the promoter and operator P.sub.L O.sub.L from lambda bacteriophage;
- an N utilization site selected from the group consisting of Nut.sub.L and Nut.sub.R for binding antiterminator N protein produced by the host cell;
- a DNA sequence which contains a mutant cII ribosomal binding site from lambda bacteriophage having the sequence: ##STR15## for rendering the mRNA transcript of the DNA encoding the polypeptide analog of the animal growth hormone capable of binding to ribosomes within the Escherichia coli host cell;
- an ATG initiation codon;
- DNA encoding the polypeptide analog of the animal growth hormone in phase with the ATG initiation codon;
- a DNA sequence which contains a T.sub.1 T.sub.2 rRNA termination sequence located less than about 100 base pairs from the 3' end of the DNA encoding the polypeptide analog of animal growth hormone;
- and which additionally includes a DNA sequence which contains the origin of replication from the bacterial plasmid pBR322 which is capable of autonomous replication in the Escherichia coli host cell and a DNA sequence which contains the fragment designated cI.sup.434, such fragment including the gene for the cI.sup.434 repressor protein and its associated promoter and operator and the distance between the 3' end of the P.sub.L O.sub.L promoter and operator sequence from lambda bacteriophage and the 5' end of the N utilization site being less than about 80 base pairs and the distance between the 3' end of the N utilization site and the 5' end of the ribosomal binding site being less than about 300 base pairs in a suitable Escherichia coli host.
- 10. The host plasmid system of claim 9, wherein the animal growth hormone is bovine growth hormone.
- 11. The host plasmid system of claim 9, wherein the host is Escherichia coli A1645 (.lambda.i434cI.sup.- mini Tn10).
- 12. A method for producing a polypeptide analog of an animal growth hormone which comprises growing
- a suitable Escherichia coli host cell comprising a plasmid for the production of a polypeptide analog of an animal growth hormone having substantially the same amino acid sequence as, and the biological activity of, the corresponding naturally-occurring animal growth hormone which upon introduction into a suitable Escherichia coli host cell containing the thermolabile repressor cI renders the host cell capable, upon increasing the temperature of the Escherichia coli host cell to a temperature at which the cI repressor is inactivated, of effecting expression of DNA encoding the polypeptide analog of the animal growth hormone consisting essentially of:
- a double-stranded DNA molecule which comprises in 5' to 3' order the following:
- a DNA sequence which includes the promoter and operator P.sub.L O.sub.L from lambda bacteriophage;
- an N utilization site selected from the group consisting of Nut.sub.L and Nut.sub.R for binding antiterminator N protein produced by the host cell;
- a DNA sequence which contains a mutant cII ribosomal binding site from lambda bacteriophage having the sequence: ##STR16## for rendering the mRNA transcript of the DNA encoding the polypeptide analog of the animal growth hormone capable of binding to ribosomes within the Escherichia coli host cell;
- an ATG initiation codon;
- DNA encoding the polypeptide analog of the animal growth hormone in phase with the ATG initiation codon;
- a DNA sequence which contains a T.sub.1 T.sub.2 rRNA termination sequence located less than about 100 base pairs from the 3' end of the DNA encoding the polypeptide analog of animal growth hormone;
- and which additionally includes a DNA sequence which contains the origin of replication from the bacterial plasmid pBR322 which is capable of autonomous replication in the Escherichia coli host cell and a DNA sequence which contains the fragment designated cI.sup.434, such fragment including the gene for the cI.sup.434 repressor protein and its associated promoter and operator and the distance between the 3' end of the P.sub.L O.sub.L promoter and operator sequence from lambda bacteriophage and the 5' end of the N utilization site being less than about 80 base pairs and the distance between the 3' end of the N utilization site and the 5' end of the ribosomal binding site being less than about 300 base pairs under suitable conditions permitting production of the polypeptide analog of the animal growth hormone and recovering the resulting polypeptide analog of the animal growth hormone.
- 13. The method of claim 12 wherein the animal growth hormone is bovine growth hormone.
- 14. The method of claim 12 wherein the host is Escherichia coli A1645 (lambda i434 cI.sup.- mini Tn10).
