Claims
- 1. A method of making microspherical particles comprising:
- a) preparing a solvent solution comprising a water miscible organic solvent, a liquid silicone compound and an amphiphilic compound having a functional group for covalent coupling to a ligand;
- b) combining said solvent solution with an aqueous medium to form an oil-in-water microemulsion having as the dispersed phase microspherical particles comprising a core and a monolayer on the surface of the core, wherein the core comprises the liquid silicone compound and the monolayer on the surface of the core comprises the amphiphilic compound;
- c) covalently coupling the ligand to the functional group of the amphiphilic compound, and;
- d) isolating the microspherical particles covalently coupled to the ligand.
- 2. The method of claim 1 wherein said solvent solution further comprises a water-insoluble dye.
- 3. The method of claim 2 wherein said liquid silicone compound is selected from the group consisting of polydimethyldiphenyl siloxane and fluorosilicones and said amphiphilic compound is a phospholipid selected from the group consisting of phosphatidyl choline, phosphatidyl glycerol, phosphatidyl ethanolamine and distearoyl derivatives thereof.
- 4. The method of claim 3 wherein said ligand is selected from the group consisting of antibodies, antigens, haptens, biotin, avidin and oligonucleotides.
- 5. The method of claim 1 wherein said water miscible organic solvent is removed prior to covalently coupling the ligand to the amphiphilic compound.
