Claims
- 1. A method for selecting transformed Saccharomyces cerevisiae cells comprising the steps of:
- (a) subjecting Saccharomyces cerevisiae host cells having a triose phosphate isomerase gene deficiency to transforming conditions in the presence of a DNA construct comprising a S. pombe triose phosphate isomerase gene which complements said deficiency; and
- (b) subjecting the cells from step (a) to normal growth conditions in a complex nutrient medium, whereby cells stably transformed with the DNA construct remain viable and multiply in the absence of additional selective pressure, while untransformed cells fail to multiply due to said deficiency.
- 2. A method for producing a protein product in a Saccharomyces cerevisiae host cell having a triose phosphate isomerase gene deficiency comprising the steps of:
- (a) transforming said Saccharomyces cerevisiae host cell with a DNA molecule comprising a S. pombe triose phosphate isomerase gene which complements said deficiency and a sequence coding for said protein product; and
- (b) culturing the transformants from step (a) under normal growth conditions in a growth medium, whereby said gene functions as a selectable marker for cells transformed with said DNA molecule in the absence of additional selective pressure, resulting in enhanced plasmid stability.
- 3. A DNA construct comprising a Schizosaccharomyces pombe triose phosphate isomerase gene as a selectable marker and a DNA sequence coding for a heterologous protein which is expressible in a Saccharomyces cerevisiae host deficient in the triose phosphate isomerase gene, and not containing any other gene that functions as a selectable marker in said Saccharomyces cerevisiae host.
- 4. A DNA construct according to claim 3 wherein said DNA construct is pFPOT, pFATPOT, pMPOT2 or pCPOT.
- 5. A DNA construct which is the plasmid pB5.
- 6. A transformant saccaromyces strain containing the DNA construct pFATPOT according to claim 4 having the deposit accession number ATCC 20699.
- 7. A transformant Saccharomyces strain containing the DNA construct pMPOT2 according to claim 4, said construct having the deposit accession number ATCC 67788.
- 8. A transformant saccaromyces containing a DNA construct according to claim 5, 4 or 3.
- 9. A transformant saccaromyces strain containing the DNA construct pB5 according to claim 5 having the deposit accession number ATCC 20698.
- 10. A method for selecting transformed Saccharomyces cerevisiae cells comprising the steps of:
- (a) subjecting Saccharomyces cerevisiae host cells having a CDC gene deficiency to transforming conditions in the presence of a DNA construct comprising a S. cerevisiae CDC gene which complements said deficiency; and
- (b) subjecting the cells from step (a) to normal growth conditions in a complex nutrient medium, whereby cells stably transformed with the DNA construct remain viable and multiply while untransformed cells fail to multiply due to said deficiency.
- 11. A method for producing a protein product in a Saccharomyces cerevisiae host cell having a CDC gene deficiency, comprising the steps of:
- (a) transforming said Saccharomyces cerevisiae host cell with a DNA molecule comprising a S. cerevisiae CDC gene which complements said deficiency; and
- (b) culturing the transformants from step (a) under normal growth conditions in a growth medium, whereby said gene functions as a selectable marker for cells transformed with said DNA molecule, resulting in enhanced plasmid stability.
