Claims
- 1. An isolated in vitro population of transformation-competent Zea mays cells, wherein the walls of said cells have been partially degraded by the removal of pectin, so as to yield a population of transformation-competent Zea mays cells, wherein said cells are stably transformed with recombinant DNA by electroporation to yield transgenic Zea mays cells which can be regenerated into a population of fertile, transgenic Zea mays plants containing said recombinant DNA which is heritable.
- 2. An isolated in vitro population of transformed Zea mays cells, produced by a process consisting essentially of: incubating a population of Zea mays cells with at least one pectin-degrading enzyme in an aqueous osmoticum, so as to partially degrade the walls of the cells, to yield a population of transformation-competent Zea mays cells; and transforming said cells with recombinant DNA by electroporation to yield transgenic Zea mays cells which can be regenerated into a population of fertile, transgenic Zea mays plants containing said recombinant DNA which is heritable.
- 3. The population of claim 2 where said pectin-degrading enzyme comprises pectinase, endopectin lyase, endopolygalacturonase or mixtures thereof.
- 4. The population of claim 3 wherein about 300-500 units of enzymatic activity, measured as maceration activity, present in said aqueous osmoticum per ml of packed Zea mays cells.
- 5. The population of claim 3 wherein said pectin-degrading enzyme is pectolyase.
- 6. The population of claim 2 wherein the aqueous osmoticum further comprises cellulase, xylanase or mixtures thereof.
- 7. The population of claim 2 wherein the incubation is carried out for about 0.75-3.0 hours at about 20.degree.-30.degree. C.
