Abstract
Substituted 1H-Pyridinyl-2-ones are useful as GABAA-Alpha 2/3 ligands.
Description




This application is a 371 of PCT/aB 98/01593, filed Jun. 1, 1998.




The present invention relates to a class of substituted 1H-pyridinyl-2-one derivatives and to their use in therapy. More particularly, this invention is concerned with substituted 1H-pyridinyl-2-one derivatives which are ligands for GABA


A


receptors and are therefore useful in the therapy of deleterious mental states.




Receptors for the major inhibitory neurotransmitter, gamma-aminobutyric acid (GABA), are divided into two main classes: (1) GABA


A


receptors, which are members of the ligand-gated ion channel superfamily; and (2) GABA


B


receptors, which may be members of the G-protein linked receptor superfamily. Since the first cDNAs encoding individual GABA


A


receptor subunits were cloned the number of known members of the mammalian family has grown to thirteen (six α subunits, three β subunits, three γ subunits and one δ subunit). It may be that further subunits remain to be discovered; however, none has been reported since 1993.




Although knowledge of the diversity of the GABA


A


receptor gene family represents a huge step forward in our understanding of this ligand-gated ion channel, insight into the extent of subtype diversity is still at an early stage. It has been indicated that an α subunit, a β subunit and a γ subunit constitute the minimum requirement for forming a fully functional GABA


A


receptor expressed by transiently transfecting cDNAs into cells. As indicated above, a δ subunit also exists, but is present only to a minor extent in GABA


A


receptor populations.




Studies of receptor size and visualisation by electron microscopy conclude that, like other members of the ligand-gated ion channel family, the native GABA


A


receptor exists in pentameric form. The selection of at least one α, one β and one γ subunit from a repertoire of thirteen allows for the possible existence of more than 10,000 pentameric subunit combinations. Moreover, this calculation overlooks the additional permutations that would be possible if the arrangement of subunits around the ion channel had no constraints (i.e. there could be 120 possible variants for a receptor composed of five different subunits).




Receptor subtype assemblies which do exist include, amongst many others, α1β2γ2, α2β2/3γ2, α3βγ2/3, α2βγ1, α5β3γ2/3, α6βγ2, α6βδ and α4βδ. Subtype assemblies containing an α1 subunit are present in most areas of the brain and are thought to account for over 40% of GABA


A


receptors in the rat. Subtype assemblies containing α2 and α3 subunits respectively are thought to account for about 25% and 17% of GABA


A


receptors in the rat. Subtype assemblies containing an α5 subunit are expressed predominantly in the hippocampus and cortex and are thought to represent about 4% of GABA


A


receptors in the rat.




A characteristic property of all known GABA


A


receptors is the presence of a number of modulatory sites, one of which is the benzodiazepine (BZ) binding site. The BZ binding site is the most explored of the GABA


A


receptor modulatory sites, and is the site through which anxiolytic drugs such as diazepam and temazepam exert their effect. Before the cloning of the GABA


A


receptor gene family, the benzodiazepine binding site was historically subdivided into two subtypes, BZ1 and BZ2, on the basis of radioligand binding studies. The BZ1 subtype has been shown to be pharmacologically equivalent to a GABA


A


receptor comprising the α1 subunit in combination with a β subunit and γ2. This is the most abundant GABA


A


receptor subtype, and is believed to represent almost half of all GABA


A


receptors in the brain.




Two other major populations are the α2βγ2 and α3βγ2/3 subtypes. Together these constitute approximately a further 35% of the total GABA


A


receptor repertoire. Pharmacologically this combination appears to be equivalent to the BZ2 subtype as defined previously by radioligand binding, although the BZ2 subtype may also include certain α5-containing subtype assemblies. The physiological role of these subtypes has hitherto been unclear because no sufficiently selective agonists or antagonists were known.




It is now believed that agents acting as BZ agonists at α1βγ2, α2βγ2 or α3βγ2 subunits will possess desirable anxiolytic properties. The α1-selective GABA


A


receptor agonists alpidem and zolpidem are clinically prescribed as hypnotic agents, suggesting that at least some of the sedation associated with known anxiolytic drugs which act at the BZ1 binding site is mediated through GABA


A


receptors containing the α1 subunit. Accordingly, it is considered that GABA


A


receptor agonists which bind more effectively to the α2 and/or α3 subunit than to α1 will be effective in the treatment of anxiety with a reduced propensity to cause sedation. Also, agents which are antagonists or inverse agonists at α1 might be employed to reverse sedation or hypnosis caused by α1 agonists.




The compounds of the present invention, being selective ligands for GABA


A


receptors, are therefore of use in the treatment and/or prevention of a variety of disorders of the central nervous system. Such disorders include anxiety disorders, such as panic disorder with or without agoraphobia, agoraphobia without history of panic disorder, animal and other phobias including social phobias, obsessive-compulsive disorder, stress disorders including post-traumatic and acute stress disorder, and generalized or substance-induced anxiety disorder; neuroses; convulsions; migraine; and depressive or bipolar disorders, for example single-episode or recurrent major depressive disorder, dysthymic disorder, bipolar I and bipolar II manic disorders, and cyclothymic disorder.

For use in medicine, the salts of the compounds of formula I will be pharmaceutically acceptable salts. Other salts may, however, be useful in the preparation of the compounds according to the invention or of their pharmaceutically acceptable salts. Suitable pharmaceutically acceptable salts of the compounds of this invention include acid addition salts which may, for example, be formed by mixing a solution of the compound according to the invention with a solution of a pharmaceutically acceptable acid such as hydrochloric acid, sulphuric acid, methanesulphonic acid, fumaric acid, maleic acid, succinic acid, acetic acid, benzoic acid, oxalic acid, citric acid, tartaric acid, carbonic acid or phosphoric acid. Furthermore, where the compounds of the invention carry an acidic moiety, suitable pharmaceutically acceptable salts thereof may include alkali metal salts, e.g. sodium or potassium salts; alkaline earth metal salts, e.g. calcium or magnesium salts; and salts formed with suitable organic ligands, e.g. quaternary ammonium salts.




The present invention includes within its scope prodrugs of the compounds of formula I above. In general, such prodrugs will be functional derivatives of the compounds of formula I which are readily convertible in vivo into the required compound of formula I. Where the compounds according to the invention have at least one asymmetric centre, they may accordingly exist as enantiomers. Where the compounds according to the invention possess two or more asymmetric centres, they may additionally exist as diastereoisomers. It is to be understood that all such isomers and mixtures thereof in any proportion are encompassed within the scope of the present invention.




The compounds of the present invention possess desirable binding properties at various GABA

receptor subtypes. The compounds in accordance with the present invention have good affinity as ligands for the α2 and/or α3 subunit of the human GABA


A


receptor. Typically, the compounds of this invention display more effective binding to the α2 and/or α3 subunit than to the α1 subunit. Typically, the compounds of the present invention have a binding affinity (K

The invention also provides pharmaceutical compositions comprising one or more compounds of this invention and a pharmaceutically acceptable carrier. Preferably these compositions are in unit dosage forms such as tablets, pills, capsules, powders, granules, sterile parenteral solutions or suspensions, metered aerosol or liquid sprays, drops, ampoules, auto-injector devices or suppositories; for oral, parenteral, intranasal, sublingual or rectal administration, or for administration by inhalation or insufflation. For preparing solid compositions such as tablets, the principal active ingredient is mixed with a pharmaceutical carrier, e.g. conventional tableting ingredients such as corn starch, lactose, sucrose, sorbitol, talc, stearic acid, magnesium stearate, dicalcium phosphate or gums, and other pharmaceutical diluents, e.g. water, to form a solid preformulation composition containing a homogeneous mixture of a compound of the present invention, or a pharmaceutically acceptable salt thereof. When referring to these preformulation compositions as homogeneous, it is meant that the active ingredient is dispersed evenly throughout the composition so that the composition may be readily subdivided into equally effective unit dosage forms such as tablets, pills and capsules. This solid preformulation composition is then subdivided into unit dosage forms of the type described above containing from 0.1 to about 500 mg of the active ingredient of the present invention. Typical unit dosage forms contain from 1 to 100 mg, for example 1, 2, 5, 10, 25, 50 or 100 mg, of the active ingredient. The tablets or pills of the novel composition can be coated or otherwise compounded to provide a dosage form affording the advantage of prolonged action. For example, the tablet or pill can comprise an inner dosage and an outer dosage component, the latter being in the form of an envelope over the former. The two components can be separated by an enteric layer which serves to resist disintegration in the stomach and permits the inner component to pass intact into the duodenum or to be delayed in release. A variety of materials can be used for such enteric layers or coatings, such materials including a number of polymeric acids and mixtures of polymeric acids with such materials as shellac, cetyl alcohol and cellulose acetate.




The liquid forms in which the novel compositions of the present invention may be incorporated for administration orally or by injection include aqueous solutions, suitably flavoured syrups, aqueous or oil suspensions, and flavoured emulsions with edible oils such as cottonseed oil, sesame oil, coconut oil or peanut oil, as well as elixirs and similar pharmaceutical vehicles. Suitable dispersing or suspending agents for aqueous suspensions include synthetic and natural gums such as tragacanth, acacia, alginate, dextran, sodium carboxymethylcellulose, methylcellulose, polyvinyl-pyrrolidone or gelatin.




In the treatment of anxiety, a suitable dosage level is about 0.01 to 250 mg/kg per day, preferably about 0.05 to 100 mg/kg per day, and especially about 0.05 to 5 mg/kg per day. The compounds may be administered on a regimen of 1 to 4 times per day.




The present invention also provides a compound of formula I for use in a method of treatment of the human or animal body, in particular for use in the treatment of anxiety.




The present invention further provides the use of a compound of formula I for the manufacture of a medicament for the treatment of disorders for which the administration of a ligand for the GABA

receptor α2 and/or α3 subunits is required, for example, for the treatment of anxiety.




There is also disclosed a prophylactic or therapeutic method of treatment of a subject suffering from a condition for which the administration of a ligand for the GABA

receptor α2 and/or α3 subunit is required, which comprises administering to that subject a prophylactically or therapeutically effective amount of a compound of formula I. An example of such a condition is anxiety.

The order in which these compounds are combined is not critical. The reaction is generally carried out at reflux for several hours.

Compounds of formula I″ constitute a further feature of the present invention as they act as ligands of GABA A receptors containing the α2 and/or α3 subunits. Preferred substitution patterns of these compounds are the same as for the compounds of formula I mutatis mutandis.




It will be understood that any compound of formula I initially obtained from any of the above processes may, where appropriate, subsequently be elaborated into a further compound of formula I by techniques known from the art.




Where the above-described processes for the preparation of the compounds according to the invention give rise to mixtures of stereoisomers, these isomers may be separated by conventional techniques such as preparative chromatography. The novel compounds may be prepared in racemic form, or individual enantiomers may be prepared either by enantiospecific synthesis or by resolution. The novel compounds may, for example, be resolved into their component enantiomers by standard techniques such as preparative HPLC, or the formation of diastereomeric pairs by salt formation with an optically active acid, such as (−)-di-p-toluoyl-d-tartaric acid and/or (+)-di-p-toluoyl-1-tartaric acid, followed by fractional crystallization and regeneration of the free base. The novel compounds may also be resolved by formation of diastereomeric esters or amides, followed by chromatographic separation and removal of the chiral auxiliary.




During any of the above synthetic sequences it may be necessary and/or desirable to protect sensitive or reactive groups on any of the molecules concerned. This may be achieved by means of conventional protecting groups, such as those described in

The following Examples illustrate the preparation of compounds according to the invention.

3-(4-Methoxyphenyl)-1-methyl-5-(thiazol-2-yl)-1H-[2,4′]bipyridinyl-6-one




Step1: N,N-Carbonyldiimidazole (20.3 g) was added portionwise to a stirred suspension of isonicotinic acid (15.4 g) in dry tetrahydrofuran (200 cm

). The gently effervescent mixture was stirred at room temperature for 90 minutes, forming an orange solution. A solution of lithium diisopropylamide was prepared by the addition of n-butyllithium (2.5 M, hexanes; 100 cm

) to a stirred solution of diisopropylamine (35 cm

Step 1: To a cooled (0° C.) solution of 3,5-diphenyl-6-chloro-1H-pyrazin-2-one (1.2 g ; prepared by the method of Cheeseman et al, J. Chem. Soc. Chem. Comm. 1971, (18), 2977-2979) in 4:1 ethylene glycol dimethylether:dimethylformamide (25 cm

)was added sodium hydride (0.28 g). The reaction was stirred for 5 minutes at 0° C. under nitrogen whereupon lithium bromide (1.17 g) was added. After a further 5 minutes stirring at room temperature, under nitrogen, methyl iodide (4.78 g) was added. The mixture was stirred at room temperature overnight under nitrogen. The reaction was diluted with water and extracted with ethyl acetate. The organic phase was separated, dried over sodium sulphate and evaporated in vacuo. A compound which is a derivative of formula I or a salt thereof: wherein:R is hydrogen, C1-6alkyl, C2-6alkenyl, C2-6alkynyl, C1-6alkoxy, C2-6alkenyloxy or C2-6alkynyloxy, and when R is not hydrogen, R is optionally independently substituted by one or more halogen atoms or hydroxy group; V is CH; W is O; X is a five-membered heteroaromatic group containing one, two, three or four heteroatoms independently selected from N, O and S providing that not more than one heteroatom is selected from O and S, the heteroaromatic group being unsubstituted or substituted with one or more groups independently selected from halogen, C1-6alkyl, C2-6alkenyl, C2-6alkynyl, C3-6cycloalkyl, C4-6cycloalkenyl and CF3; Y is hydrogen, NR1R2, C1-6alkyl, C2-6alkenyl, C2-6alkynyl, Ar, O(CH2)nAr1, or CkH2k−4Ar2; R1 and R2 are independently selected from C1-6alkyl and (CH2)mAr3; Ar is thienyl, furyl or a six-membered heteroaromatic ring containing one or two nitrogen atoms which is unsubstituted or substituted with one or more groups independently selected from halogen and C1-6alkyl groups and which is optionally fused to a benzene ring; or napththyl or phenyl rings which rings are unsubstituted or substituted with one or more groups independently selected from halogen, cyano, amino, nitro, C1-6alkyl, C2-6alkenyl, C2-6alkynyl, CF3, CF3O, C1-6alkoxy, C2-6alkenyloxy, C2-6alkynyloxy, C1-6alkylthio, C2-6alkenylthio C2-6alkynylthio, hydroxy, hydroxyC1-6alkyl, NR3R4, OC(O)NR3R4, C1-6alkoxyphenylC1-6alkoxy, cyanoC1-6alkyl, cyanoC2-6alkenyl, cyanoC2-6alkynyl, pyridyl, phenyl, C1-6alkoxycarbonyl, C1-6alkoxycarbonylC1-6alkyl, C1-6alkoxycarbonylC2-6alkenyl, C1-6alkoxycarbonylC2-6alkynyl and —O(CH2)pO— and which is optionally fused to a benzene ring; Ar1, Ar2, Ar3 are independently selected from pyridyl; and phenyl which is unsubstituted or substituted with one or more groups independently selected from halogen, cyano, amino, nitro, C1-6alkyl, C2-6alkenyl, C2-6alkynyl, CF3, C1-6alkoxy, C2-6alkenyloxy, C2-6alkynyloxy, C1-6alkylthio, C2-6alkenylthio, C2-6alkynylthio and —O(CH2)pO—; Z is Het1; R3, and R4 are independently hydrogen or C1-6alkyl; R5 and R6 are independently selected from hydrogen, C1-6alkyl, C2-6alkenyl, C2-6alkynyl, C1-6hydroxyalkyl and (CH2)mAr3; Ar4 is phenyl which is unsubstituted or substituted with one or more groups independently selected from halogen, cyano, amino, nitro, C1-6alkyl, C2-6alkenyl, C2-6alkynyl, CF3, C1-6alkoxy, C2-6alkenyloxy, C2-6alkynyloxy, C1-6alkylthio, C2-6alkenylthio, C2-6alkynylthio and —O(CH2)pO—; Het1 is a four- or five-membered saturated ring containing a nitrogen atom optionally substituted by a hydroxy group; a six-membered saturated ring containing a nitrogen atom, and optionally a further nitrogen atom or an oxygen atom; an unsaturated five-membered heterocyclic group containing one, two, three or four heteroatoms independently selected from N, O and S providing that not more than one heteroatom is selected from O and S; or an unsaturated six-membered heterocyclic group containing one or two nitrogen atoms; each of which moieties is unsubstituted or substituted by one or more groups independently selected from halogen, cyano, amino, C1-6alkyl, C2-6alkenyl, C2-6alkynyl, CF3, C1-6alkoxy, C2-6alkenyloxy, C2-6alkynyloxy; k is 2; m and n are 1; and p is 1, or 2.
  • 2. A compound according to claim 1 wherein:R is hydrogen, C1-6 alkyl, C2-6 alkenyl, C2-6 alkynyl, C1-6 alkoxy, C2-6 alkenyloxy or C2-6 alkynyloxy; V is CH; and W is O.
  • 3. A compound according to claim 1, wherein:Y is hydrogen, NR1R2, C2-6 alkynyl, Ar, O(CH2)nAr1 or CkH2k−4Ar2; R1 and R2 are independently selected from C1-6 alkyl and (CH2)mAr3; Ar is pyridyl or pyrimidinyl and is unsubstituted or substituted with halogen or C1-6 alkyl; unsubstituted naphthyl; or phenyl which is unsubstituted or substituted with Rx and/or Ry and/or Rz wherein Rx and Ry are independently chosen from halogen, cyano, amino, nitro, C1-6 alkyl, C2-6 alkenyl, C2-6 alkynyl, CF3, OCF3, C1-6 alkoxy, C2-6 alkenyloxy, C2-6 alkynyloxy, C1-6 alkylthio, C2-6 alkenylthio, C2-6 alkynylthio, hydroxy, hydroxyC1-6alkyl, NR3R4, OC(O)NR3R4, C1-6 alkoxyphenylC1-6alkoxy, cyanoC1-6alkly, cyanoC2-6alkenyl, cyanoC2-6alkynyl, pyridyl, phenyl, C1-6 alkoxycarbonyl, C1-6 alkoxycarbonylC1-6alkyl, C1-6 alkoxycarbonylC2-6alkenyl, C1-6 alkoxycarbonylC2-6alkynyl and —O(CH2)pO— and Rz is halogen, C1-6 alkyl or C1-6 alkoxy and Ar is optionally fused to a benzene ring; Ar1, Ar2 and A3 are independently: pyridyl; or phenyl which is unsubstituted or substituted with halogen, cyano, amino, nitro, C1-6 alkyl, C2-6 alkenyl, C2-6 alkynyl, CF3, C1-6 alkoxy, C2-6 alkenyloxy, C2-6 alkynyloxy, C1-6 alkythio, C2-6 alkenylthio, C2-6 alkynylthio or —O(CH2)pO—; R3 and R4 are independently chosen from hydrogen and C2-6alkyl; k is 2; m and n are 1; and p is 1 or 2.
  • 4. A compound according to claim 1 which is:3-(4-methoxyphenyl)-1-methyl-5-(thiazol-2-yl)-1H-[2,4′]bipyridinyl-6-one; 5-(4-methoxyphenyl)-1-methyl-6-(5-methylfuran-2-yl)-3-(3-methyl-[1,2,4]oxadiazol-5-yl)-1H-pyridin-2-one; 3-(4-methoxyphenyl)-1-methyl-5-(4-methylthiazol-2-yl)-1H-[2,4′]bipyridinyl-6-one; 5-(4-methoxyphenyl)-1-methyl-3-(thiazol-2-yl)-6-(3-thienyl)-1H-pyridin-2-one; 5-(4-methoxyphenyl)-1-methyl-3-(5-cyclopropyl-[1,2,4]oxadiazol-3-yl)-6-(3-thienyl)-1H-pyridin-2-one; 5-(4-methoxyphenyl)-1-methyl-6-(5-methylfuran-2-yl)-3-(thiazol-2-yl)-1H-pyridin-2-one; 3-(4-methoxyphenyl)-1-methyl-5-(thiophen-2-yl)-1H-[2,4′]bipyridinyl-6-one; 3-(4-methoxyphenyl)-1-methyl-5-(4-cyclopropylthiazol-2-yl)-1H-[2,4′]bipyridinyl-6-one; 1-methyl-3-(4-pyridyl)-5-(thiazol-2-yl)-1H-[2,4′]bipyridinyl-6-one; 3-(4-methoxyphenyl)-1-methyl-5-(4-methylthiophen-2-yl)-1H-[2,4′]bipyridinyl-6-one; 5-(4-methoxyphenyl)-1-methyl-3-(4-methylthiazol-2-yl)-6-pyridazin-4-yl-1H-pyridin-2-one; 1-methyl-3-(4-methylthiazol-2-yl)-6-(pyridazin-4-yl)-5-(2,4,6-trifluorophenyl)-1H-pyridin-2-one; 5-benzyloxy-1-methyl-3-(4-methylthiazol-2-yl)-6-(pyridin-4-yl)-1H-pyridin-2-one; 1-methyl-3-(1-methylpyrazol-3-yl)-5-(4-methoxyphenyl)-6-(pyridin-4-yl)-1H-pyridin-2-one; 5-(3,4-methylenedioxyphenyl )-1-methyl-3-(4-methylthiazol-2-yl)-6-(pyridin-4-yl)-1H-pyridin-2-one; 5-(4-methoxyphenyl)-1-methyl-3-(4-methylthiazol-2-yl)-6-(pyrazin-2-yl)-1H-pyridin-2-one; 5-(4-methoxyphenyl)-3-(3-methylisothiazol-5-yl)-6-(4-pyridyl)-1-methyl-1H-pyridin-2-one; 1-methyl-3-(4-methylthiazol-2-yl)-5-(N-methyl-N-benzylamino)-6-(4-pyridyl)-1H-pyridin-2-one; 5-(4-methoxyphenyl)-1-methyl-3-(4-methylthiazol-2-yl)-6-(4-morpholino)-1H-pyridin-2-one; 1-methyl-3-(4-methylthiazol-2-yl)-6-(4-morpholino)-1H-[3,4′]bipyridin-2-one; 1-methyl-3-(4-methylthiazol-2-yl)-6-(1-pyrrolidino)-1H-[3,4,′]bipyridin-2-one; or a salt thereof.
  • 5. A pharmaceutical composition comprising a compound according to claim 1, or a salt thereof, and a pharmaceutically acceptable carrier.
  • 6. A compound according to claim 1, or a salt thereof, for use in a method of treatment of the human or animal body.
  • 7. Use of a compound according to claim 1, or a salt thereof, for the manufacture of a medicament for the treatment of disorders for which the administration of a ligand for the GABAA receptor α2 and/or α3 subunits is required.
  • 8. A prophylactic or therapeutic method of treatment of a subject suffering from a condition for which the administration of a ligand for the GABAA receptor α2 and/or α3 subunit is required, which comprises administering to that subject a prophylactically or therapeutically effective amount of a compound according to claim 1, or a salt thereof.
  • 9. A process for producing a compound according to claim 1 in which V is CH which comprises:reacting a compound of formula VII with a compound of formula  wherein R, X, Y and Z are as defined in claim 1 and R″ and R′″ are independently halogen or C1-6 alkyl.
