Claims
- 1. A teicoplanin amide derivative of formula I ##STR18## wherein R represents hydrogen or a protecting group of the amine function;
- Y represents a compound of formula
- --NR.sub.1 --alk.sub.1 --[X--alk.sub.2 ].sub.p --[T--alk.sub.3 ].sub.q --W
- wherein
- R.sub.1 represents hydrogen or (C.sub.1 -C.sub.4)alkyl;
- alk.sub.1, alk.sub.2 and alk.sub.3 each independently represents a linear or branched alkylene of 2 to 10 carbon atoms;
- p represents an integer ranging from 1 to 50;
- q represents an integer ranging from 0 to 12;
- X represents a --NR.sub.2 -- group or an oxygen atom wherein R.sub.2 represents hydrogen, (C.sub.1 -C.sub.4)alkyl, a group alk.sub.4 NR.sub.3 R.sub.4 wherein alk.sub.4 represents a linear or branched alkylene of 2 to 4 atoms, R.sub.3 is hydrogen or (C.sub.1 -C.sub.4)alkyl and R.sub.4 is hydrogen, (C.sub.1 -C.sub.4)alkyl or a 5-6 membered cycloalkyl; or R.sub.1 and R.sub.2 taken together represent a (C.sub.2 -C.sub.4)alkylene moiety connecting the two nitrogen atoms with the proviso that in such case p is 1;
- T represents a --NR.sub.5 -- group or an oxygen atom wherein R.sub.5 is hydrogen, (C.sub.1 -C.sub.4)alkyl, a group alk.sub.5 NR.sub.6 R.sub.7 wherein alk.sub.5 represents a linear or branched alkylene of 2 to 4 atoms, R.sub.6 is hydrogen or (C.sub.1 -C.sub.4)alkyl and R.sub.7 is hydrogen, (C.sub.1 -C.sub.4)alkyl or a 5-6 membered cycloalkyl; or R.sub.2 and R.sub.5 taken together represent a (C.sub.2 -C.sub.4)alkylene moiety connecting the two nitrogen atoms with the proviso that in such case both p and q are 1;
- W represents hydroxy, NR.sub.8 R.sub.9 or N.sup..sym. R.sub.11 R.sub.12 R.sub.13 An.sup..crclbar., R.sub.8 represents hydrogen or (C.sub.1 -C.sub.6)alkyl, R.sub.9 represents hydrogen, (C.sub.1 -C.sub.6)alkyl, (C.sub.5 -C.sub.6)cycloalkyl or COOR.sub.10, R.sub.10 represents (C.sub.1 -C.sub.6)acyloxy(C.sub.1 -C.sub.4)alkyl, and, R.sub.11, R.sub.12, and R.sub.13 each independently represents (C.sub.1 -C.sub.4)alkyl and An.sup..crclbar. is an anion derived from a pharmaceutically acceptable acid; with the proviso that when simultaneously X is NR.sub.2, p is 1 and q is zero, then W is not represented by hydroxy;
- A represents H or --N[(C.sub.9 -C.sub.12)aliphatic acyl]- beta-D-2-deoxy-2-aminoglucopyranosyl,
- B represents hydrogen or N-acetyl-beta-D-2-deoxy-2-aminoglucopyranosyl;
- M represents hydrogen or alpha-D-mannopyranosyl or the pharmaceutically addition salt thereof;
- with the further proviso that B represents hydrogen only when A and M are simultaneously hydrogen (SEQ ID NO 1).
- 2. A compound of claim 1 wherein the (C.sub.9 -C.sub.12) aliphatic acyl radicals of the symbol A is one of the following:
- (Z)-4-decenoyl, 8-methylnonanoyl, decanoyl, 8-methyldecanoyl, 9-methyldecanoyl, 6-methyloctanoyl, nonanoyl, 10-methylundecanoyl and dodecanoyl.
- 3. A compound of claim 1 wherein X represents --NR.sub.2 -- and T represents --NR.sub.5 --; alk.sub.1, alk.sub.2, and alk.sub.3, each represent a (C.sub.2 -C.sub.10) linear chain, p is an integer from 1 to 5, and q is an integer from 0 to 12.
- 4. A compound of claim 1 wherein both X and T represent oxygen atoms and p and q are such that the sum of p+q represent an integer from 2 to 50.
- 5. A compound of claim 1 wherein X represents --NR.sub.2 -- wherein R.sub.2 is hydrogen, a (C.sub.1 -C.sub.4 )alkyl or a group alk.sub.4 NR.sub.3 R.sub.4 wherein alk.sub.4, R.sub.3 and R.sub.4 are defined as in claim 1.
- 6. A compound of claim 1 wherein p is 1 and X is --NR.sub.2 -- wherein R.sub.2 taken together with R.sub.1 represent a (C.sub.2 -C.sub.3)alkylene moiety connecting the nitrogen atoms.
- 7. A compound of claim 1 wherein p is 1, q is 1 and X and T are --NR.sub.2 -- and --NR.sub.5 -- respectively, wherein R.sub.2 and R.sub.5 taken together represent a (C.sub.2 -C.sub.3)alkylene moiety connecting the nitrogen atoms.
- 8. A compound of claim 6 wherein the alkylene moiety is a group --CH.sub.2 --CH.sub.2 --.
- 9. A compound of claim 1 wherein X and T are oxygen atoms and W is hydroxy or --NR.sub.8 R.sub.9 -- wherein R.sub.8 is hydrogen or (C.sub.1 -C.sub.4)alkyl and R.sub.9 is hydrogen, (C.sub.1 -C.sub.4)alkyl, cyclopentyl or cyclohexyl.
- 10. A compound of claim 1 wherein W represents --NR.sub.8 R.sub.9 -- wherein R.sub.8 is as defined and R.sub.9 is COOR.sub.10 in which R.sub.10 is a (C.sub.1 -C.sub.6)acyloxy-(C.sub.1 -C.sub.4)alkyl group.
- 11. A compound of claim 10 wherein the (C.sub.1 -C.sub.4)alkyl group of the (C.sub.1 -C.sub.6)acyloxy-(C.sub.1 -C.sub.4)alkyl moiety is a methylene optionally substituted with a (C.sub.1 -C.sub.3)linear or branched alkyl chain.
- 12. A compound according to claim 1 in which R is represented by hydrogen; A is represented by hydrogen; B is represented by hydrogen; M is represented by hydrogen and Y is represented by --NH--(CH.sub.2).sub.3 --N[(CH.sub.2).sub.3 --NH.sub.2 ].sub.2.
- 13. A compound according to claim 1 in which R is represented by hydrogen; A is represented by hydrogen; B is represented by hydrogen; M is represented by hydrogen; and Y is represented by --NH(CH.sub.2).sub.3 --NH(CH.sub.2).sub.3 --NH(CH.sub.2).sub.3 --NH.sub.2.
- 14. An antibacterial formulation comprising a compound according to claim 1 present in an antibacterially effective amount in admixture with a pharmaceutically acceptable carrier.
- 15. An antibacterial formulation comprising a compound according to claim 12 present in an antibacterially effective amount in admixture with a pharmaceutically acceptable carrier.
- 16. An antibacterial formulation comprising a compound according to claim 13 present in an antibacterially effective amount in admixture with a pharmaceutically acceptable carrier.
- 17. A method for the treatment of bacterial infections comprising administering to a patient in need thereof an antibacterially effective amount of a compound according to claim 1.
- 18. A method for the treatment of bacterial infections comprising administering to a patient in need thereof an antibacterially effective amount of a compound according to claim 12.
- 19. A method for the treatment of bacterial infections comprising administering to a patient in need thereof an antibacterially effective amount of a compound according to claim 13.
