Claims
- 1. A method of preserving an organ or tissue, comprising the steps of:
(a) providing a base electrolyte solution; (b) mixing a first portion of the base solution with a first additive to produce a first mixed solution; (c) contacting the first mixed solution with an organ or tissue during at least one stage of procurement, preservation or transplantation of that organ or tissue; (d) mixing a second portion of the base solution with a second, different additive to produce a second, different mixed solution; and (e) contacting the second mixed solution with an organ or tissue during at least one stage of procurement, preservation or transplantation of that organ or tissue.
- 2. The method of claim 1, wherein said organ or tissue is isolated.
- 3. The method of claim 1, wherein said organ or tissue is in a cadaver.
- 4. The method of claim 1, wherein said organ or tissue is in a living patient.
- 5. The method of claim 4, wherein said patient is undergoing bloodless surgery.
- 6. The method of claim 1, wherein the organ or tissue of step (c) and the organ or tissue of step (e) are the same organ or tissue.
- 7. The method of claim 1, wherein the organ or tissue of step (c) and the organ or tissue of step (e) are different organs or tissue.
- 8. The method of claim 1, wherein said base solution comprises an intracellular base solution.
- 9. The method of claim 1, wherein said base solution comprises an extracellular base solution.
- 10. The method of claim 1, wherein at least one said contacting step comprises purging vasculature of said organ or tissue of blood in preparation for preservation; and
wherein said base solution is an extracellular base solution, and the additive of said at least one said contacting step forms a purge solution from said extracellular base solution.
- 11. The method of claim 1, wherein at least one said contacting step comprises protecting and maintaining cellular stability during cold storage of the organ or tissue; and
wherein said base solution is an intracellular base solution, and the additive of said at least one said contacting step is a cytoprotective additive that forms a maintenance solution from said intracellular base solution.
- 12. The method of claim 1, wherein at least one said contacting step comprises restoring viability of said organ or tissue by near normothermic perfusion; and
wherein said base solution is an extracellular base solution, and the additive of said at least one said contacting step is a rescue additive.
- 13. The method of claim 1, wherein at least one said contacting step comprises flushing unwanted molecules away from an organ or tissue;
and wherein said base solution is an extracellular base solution, and the additive of said at least one said contacting step is a rinse additive.
- 14. The method of claim 13, wherein said unwanted molecules are preservation solution molecules, and said at least one said contacting step is followed by reimplantation of the organ or tissue.
- 15. The method of claim 13, wherein said unwanted molecules are erythrocytes and other blood components, and said at least one said contacting step is followed by a preservation process.
- 16. The method of claim 1, wherein at least one said stage of step (c) comprises cryopreservation of said organ or tissue by sub-zero cooling;
and wherein said base solution is an intracellular base solution, and said first additive is a cryoprotective additive.
- 17. The method of claim 1, wherein said base solution comprises:
Na+; K+; Ca++; Mg++; Cl−; H2PO4−; HCO3−; HEPES; Lactobionate; Sucrose; Mannitol; Glucose; Gluconate; Dextran 40; Adenosine; and optionally Glutathione.
- 18. The method of claim 17, wherein the base solution does not contain Glutathione; and further comprising the step of adding Glutathione to the base solution or to the mixed solution.
- 19. The method of claim 1, wherein said base solution comprises:
40-80 or 100-150 mM/l Na+; 15-40 or 50-90 mM/l K+; 0.01-0.1 mM/l Ca++; 5-25 mM/l Mg++; 20-40 mM/l Cl−; 1-5 mM/l H2PO4−; 3-7 mM/l HCO3−; 25-50 mM/l HEPES; 25-50 mM/l Lactobionate; 10 mM/l-1M/l Sucrose; 15-30 mM/l Mannitol; 1-10 mM/l Glucose; 50-100 mM/l Gluconate; 6% Dextran 40; 0.1-2 mM/l Adenosine; and optionally 1-5 mM/l Glutathione.
- 20. The method of claim 19, wherein the base solution does not contain Glutathione; and further comprising the step of adding said Glutathione to the base solution or to the mixed solution.
- 21. The method of claim 19, wherein the base solution comprises 40-80 mM/l Na+ and 50-90 mM/l K+.
- 22. The method of claim 19, wherein the base solution comprises 100-150 mM/l Na+ and 15-40 mM/l K+.
- 23. The method of claim 1, wherein said base solution comprises:
about 62.5 or about 125 mM/l Na+about 25 or about 70.0 mM/l K+; about 0.05 mM/l Ca++; about 15.0 mM/l Mg++; about 30.1 mM/l Cl−; about 2.5 mM/l H2PO4−; about 5.0 mM/l HCO3−; about 35.0 mM/l HEPES; about 30.0 mM/l Lactobionate; about 15.0 mM/l Sucrose; about 25.0 mM/l Mannitol; about 5.0 mM/l Glucose; about 70.0 mM/l Gluconate; about 6% Dextran 40; about 2.0 mM/l Adenosine; and optionally about 3.0 mM/l Glutathione.
- 24. The method of claim 23, wherein the base solution does not contain Glutathione; and further comprising the step of adding said Glutathione to the base solution or to the mixed solution.
- 25. The method of claim 23, wherein the base solution comprises about 62.5 mM/l Na+; and about 70 mM/l K+.
- 26. The method of claim 23, wherein the base solution comprises about 125 mM/l Na+ and about 25 mM/l K+.
- 27. The method of claim 1, wherein the base solution comprises:
Na+; K+; Ca++; Mg++; Cl−; SO4−; H2PO4−; HCO3−; HEPES; Glucose; Dextran 40; Adenosine; and optionally Glutathione.
- 28. The method of claim 27, wherein the base solution does not contain Glutathione; and further comprising the step of adding said Glutathione to the base solution or to the mixed solution.
- 29. The method of claim 1, wherein the base solution comprises:
120-160 mM/l Na+; 3-9 mM/l K+; 1-3 mM/l Ca++; 1-10 mM/l Mg++; 100-150 mM/l Cl−; 1-10 mM/l SO4−; 1-3 mM/l H2PO4−; 20-30 mM/l HCO3−; 5-15 mM/l HEPES; 5-10 mM/l Glucose; 6% Dextran 40; 0.1-2 mM/l Adenosine; and optionally 1-5 mM/l Glutathione.
- 30. The method of claim 29, wherein the base solution does not contain Glutathione; and further comprising the step of adding said Glutathione to the base solution or to the mixed solution.
- 31. The method of claim 1, wherein the base solution comprises:
about 141.2 mM/l Na+; about 6.0 mM/l K+; about 1.5 mM/l Ca++; about 5.0 mM/l Mg++; about 122.0 mM/l Cl−; about 1.0 mM/l SO4−; about 1.2 mM/l H2PO4−; about 25.0 mM/l HCO3−; about 25.0 mM/l HEPES; about 5.0 mM/l Glucose; about 6% Dextran 40; about 1.0 mM/l Adenosine; and optionally about 3.0 mM/l Glutathione.
- 32. The method of claim 31 wherein the base solution does not contain Glutathione; and further comprising the step of adding said Glutathione to the base solution or to the mixed solution.
- 33. An organ or tissue preservation solution, comprising:
40-80 or 100-150 mM/l Na+; 15-40 or 50-90 mM/l K+; 0.01-0.1 mM/l Ca++; 5-25 mM/l Mg++; 20-40 mM/l Cl−; 1-5 mM/l H2PO4−; 3-7 mM/l HCO3−; 25-50 mM/l HEPES; 25-50 mM/l Lactobionate; 10 mM/l-1M/l Sucrose; 15-30 mM/l Mannitol; 1-10 mM/l Glucose; 50-100 mM/l Gluconate; 6% Dextran 40; 0.1-2 mM/l Adenosine; and optionally 1-5 mM/l Glutathione.
- 34. The solution of claim 33, wherein said solution further comprises 1-5 mM/l Glutathione.
- 35. The solution of claim 33, wherein the solution comprises 40-80 mM/l Na+ and 50-90 mM/l K+.
- 36. The solution of claim 33, wherein the solution comprises 100-150 mM/l Na+ and 15-40 mM/l K+.
- 37. An organ or tissue preservation solution, comprising:
about 62.5 or 125 mM/l Na+; about 25 or 70.0 mM/l K+; about 0.05 mM/l Ca++; about 15.0 mM/l Mg++; about 30.1 mM/l Cl−; about 2.5 mM/l H2PO4−; about 5.0 mM/l HCO3−; about 35.0 mM/l HEPES; about 30.0 mM/l Lactobionate; about 15.0 mM/l Sucrose; about 25.0 mM/l Mannitol; about 5.0 mM/l Glucose; about 70.0 mM/l Gluconate; about 6% Dextran 40; and about 2.0 mM/l Adenosine.
- 38. The solution of claim 37, wherein said solution further comprises about 3.0 mM/l Glutathione.
- 39. The solution of claim 37, wherein the solution comprises about 52.5 mM/l Na+; and about 70 mM/l K+.
- 40. The solution of claim 37, wherein the solution comprises about 125 mM/l Na+ and about 25 mM/l K+.
- 41. An organ or tissue preservation solution, comprising:
120-160 mM/l Na+; 3-9 mM/l K+; 1-3 mM/l Ca++; 1-10 mM/l Mg++; 100-150 mM/l Cl−; 1-10 mM/l SO4−; 1-3 mM/l H2PO4−; 20-30 mM/l HCO3−; 5-15 mM/l HEPES; 5-10 mM/l Glucose; 6% Dextran 40; 0.1-2 mM/l Adenosine; and optionally 1-5 mM/l Glutathione.
- 42. The solution of claim 41, wherein said solution further comprises 1-5 mM/l Glutathione.
- 43. An organ or tissue preservation solution, comprising:
about 141.2 mM/l Na+; about 6.0 mM/l K+; about 1.5 mM/l Ca++; about 5.0 mM/l Mg++; about 122.0 mM/l Cl−; about 1.0 mM/l SO4−; about 1.2 mM/l H2PO4−; about 25.0 mM/l HCO3−; about 25.0 mM/l HEPES; about 5.0 mM/l Glucose; about 6% mM/l Dextran 40; and about 1.0 mM/l Adenosine.
- 44. The solution of claim 43, wherein said solution further comprises about 3.0 mM/l Glutathione.
- 45. The solution of claim 43, wherein said solution has an osmolality (mOsm/Kg) of about 350, a pH of about 7.6, and a [K+][Cl−] of about 2100.
- 46. The solution of claim 43, wherein said solution has an osmolality (mOsm/Kg) of about 315, a pH of about 7.5, and a [K+][Cl−] of about 732.
- 47. A solution kit comprising a saleable package containing:
at least one first container containing at least one base electrolyte solution; and at least two second containers respectively containing different additives for converting said base solution to different mixed solutions for use in different stages of procurement, preservation or transplantation of an organ or tissue.
- 48. The kit of claim 47, wherein said at least one first container comprises at least one container containing an intracellular base solution, and at least one container containing an extracellular base solution.
- 49. The kit of claim 47, wherein at least one said additive is a purge additive for purging an organ or tissue of blood.
- 50. The kit of claim 47, wherein at least one said additive is a cryoprotection additive.
- 51. The kit of claim 50, wherein the base solution is an intracellular base solution.
- 52. The kit of claim 47, wherein at least one said additive is a rescue additive.
- 53. The kit of claim 52, wherein the base solution is an extracellular base solution.
- 54. The kit of claim 47, wherein at least one said additive is a rinse additive for flushing unwanted preservation molecules away from an organ or tissue.
- 55. The kit of claim 54, wherein the base solution is an extracellular base solution.
- 56. The kit of claim 47, wherein said base solution comprises:
Na+; K+; Ca++; Mg++; Cl−; H2PO4−; HCO3−; HEPES; Lactobionate; Sucrose; Mannitol; Glucose; Gluconate; Dextran 40; and Adenosine.
- 57. The kit of claim 56, wherein said base solution further comprises Glutathione or wherein said kit further comprises a container containing Glutathione.
- 58. The kit of claim 47, wherein said base solution comprises:
40-80 or 100-150 mM/l Na+; 15-40 or 50-90 mM/l K+; 0.01-0.1 mM/l Ca++; 5-25 mM/l Mg++; 20-40 mM/l Cl−; 1-5 mM/l H2PO4−; 3-7 mM/l HCO3−; 25-50 mM/l HEPES; 25-50 mM/l Lactobionate; 10 mM/l-1M/l Sucrose; 15-30 mM/l Mannitol; 1-10 mM/l Glucose; 50-100 mM/l Gluconate; 6% Dextran 40; 0.1-2 mM/l Adenosine; and optionally 1-5 mM/l Glutathione.
- 59. The organ or tissue preservation kit of claim 58, wherein said base solution further comprises 1-5 mM/l Glutathione or wherein said kit further comprises a container containing an amount of Glutathione that, when added to the intracellular base solution, produces an intracellular base solution that comprises 1-5 mM/l Glutathione.
- 60. The kit of claim 58, wherein the base solution comprises 40-80 mM/l Na+; and 50-90 mM/l K+.
- 61. The kit of claim 58, wherein the base solution comprises 100-150 mM/l Na+ and 15-40 mM/l K+.
- 62. The kit of claim 47, wherein said base solution comprises:
about 62.5 or 125 mM/l Na+; about 25 or 70.0 mM/l K+; about 0.05 mM/l Ca++; about 15.0 mM/l Mg++; about 30.1 mM/l Cl−; about 2.5 mM/l H2PO4−; about 5.0 mM/l HCO3−; about 35.0 mM/l HEPES; about 30.0 mM/l Lactobionate; about 15.0 mM/l Sucrose; about 25.0 mM/l Mannitol; about 5.0 mM/l Glucose; about 70.0 mM/l Gluconate; about 6% Dextran 40; and about 2.0 mM/l Adenosine.
- 63. The organ or tissue preservation kit of claim 62, wherein said base solution further comprises about 3.0 mM/l Glutathione or wherein said kit further comprises a container containing an amount of Glutathione that, when added to the intracellular base solution, produces an intracellular base solution that comprises about 3.0 mM/l Glutathione.
- 64. The kit of claim 62, wherein the base solution comprises about 62.5 mM/l Na+; and about 70 mM/l K+.
- 65. The kit of claim 62, wherein the base solution comprises about 125 mM/l Na+ and about 25 mM/l K+.
- 66. The kit of claim 47, wherein said base solution comprises:
Na+; K+; Ca++; Mg++; Cl−; SO4−; H2PO4−; HCO3−; HEPES; Glucose; Dextran 40; and Adenosine.
- 67. The kit of claim 66, wherein said base solution further comprises Glutathione or wherein said kit further comprises a container containing Glutathione.
- 68. The kit of claim 47, wherein said base solution comprises:
120-160 mM/l Na+; 3-9 mM/l K+; 1-3 mM/l Ca++; 1-10 mM/l Mg++; 100-150 mM/l Cl−; 1-10 mM/l SO4−; 1-3 mM/l H2PO4−; 20-30 mM/l HCO3−; 5-15 mM/l HEPES; 5-10 mM/l Glucose; 6% Dextran 40; and 0.1-2 mM/l Adenosine.
- 69. The kit of claim 68, wherein said base solution further comprises 1-5 mM/l Glutathione or wherein said kit further comprises a container containing an amount of Glutathione that, when added to the base solution, produces a base solution that comprises 1-5 mM/l Glutathione.
- 70. The kit of claim 47, wherein said base solution comprises:
about 141.2 mM/l Na+; about 6.0 mM/l K+; about 1.5 mM/l Ca++; about 5.0 mM/l Mg++; about 122.0 mM/l Cl−; about 1.0 mM/l SO4−; about 1.2 mM/l H2PO4−; about 25.0 mM/l HCO3−; about 25.0 mM/l HEPES; about 5.0 mM/l Glucose; about 6% mM/l Dextran 40; and about 1.0 mM/l Adenosine.
- 71. The kit of claim 70, wherein said base solution further comprises about 3.0 mM/l Glutathione or wherein said kit further comprises a container containing an amount of Glutathione that, when added to the base solution, produces a base solution that comprises about 3.0 mM/l Glutathione.
- 72. The kit of claim 70, wherein said base solution has an osmolality (mOsm/Kg) of about 350, a pH of about 7.6, and a [K+][Cl−] of about 2100.
- 73. The kit of claim 70, wherein said base solution has an osmolality (mOsm/Kg) of about 315, a pH of about 7.5, and a [K+][Cl−] of about 732.
