The present invention relates to a T-type calcium channel inhibitor. More specifically, the present invention relates to a novel compound having 1,3-hydro-2H-benzimidazol-2-one structure with the effect of inhibiting T-type calcium channels. Also, the present invention can be used as a medicament for treating a disease associated with the activation of T-type calcium channels such as pain and pruritus which can inhibit the risk of side effects by decreasing the effect of inhibiting dopamine D2 receptor (hereinafter referred to as D2 receptor, as necessary).
Ion channels are channels extending through the cell membrane and are classified broadly into two types: ligand-dependent channels and potential-dependent channels. In recent years, as a type of the mechanism of neuropathic pain, a potential-dependent sodium channel and a potential-dependent calcium channel have been found as a target. As drugs targeting a potential-dependent sodium channel, for example, lidocaine, carbamazepine, lamotrigine, mexiletine and the like are known. As drugs targeting a potential-dependent calcium channel, for example, gabapentin, pregabalin, ziconotide and the like are known.
The neuropathic pain is refractory and has the problem of inadequate response to existing analgesic drugs. The efficacy of even an established drug therapy is difficult to predict accurately, and other drugs must often be used in combination. In the Guidelines for the Pharmacologic Management of Neuropathic Pain from Japan Society of Pain Clinicians, high-potential-activated calcium channel inhibitors are listed as a primary choice, but in many cases, an adequate therapeutic effect is not obtained. These inhibitors have the problem of causing side effects such as lightheadedness.
Potential-dependent calcium channels are classified into two types: high-potential-activated types and low-potential-activated types based on a difference in activation and deactivation potentials. L-type, N-type, P/Q-type and R-type calcium channels are activated by large depolarization, and therefore classified as high-potential-activated calcium channels. On the other hand, T-type calcium channels are activated by small depolarization, and therefore classified as low-potential-activated calcium channels.
Recently, T-type calcium channels have been reported to be involved in the onset and development of neuropathic pain, and the potential of T-type calcium channel inhibitors as therapeutic agents for neuropathic pain has been suggested. Examples of the T-type calcium channel inhibitor include mibefradil (for example, Non-Patent Document 1), ethosuximide, and (1S,2S)-2-[2-[[3-(1H-benzimidazol-2-yl)propyl]methyamino]ethyl]-6-fluoro-1,2,3,4-tetrahydro-1-(1-methylethyl)-2-naphthalenyl-cyclopropane carboxylate hydrochloride (NNC55-0396) (for example, Non-Patent Document 2).
Also, it has been known that T-type calcium channels are associated with the pathology of various diseases and disorders including epilepsy, essential tremor, schizophrenia, Parkinson's disease, depression, anxiety, sleep disorder, mental illness, schizophrenia, cardiac arrhythmia, hypertension, cancer, diabetes, overactive bladder, chronic kidney disease, sterility and sexual dysfunction, besides pains such as neuropathic pain, inflammatory pain and cancer pain (Patent Document 1).
1-{1-[4,4-Bis(4-fluorophenyl)butyl]piperidin-4-yl}-1,3-dihydro-2H-benzimidazol-2-one (hereinafter also referred to as pimozide, as necessary) is currently known as a compound having 1,3-dihydro-2H-benzimidazol-2-one structure with the effect of inhibiting T-type calcium channels. However, the compound has the effect of inhibiting D2 receptor stronger than the effect of inhibiting T-type calcium channels. Pimozide has the problem of causing undesired effects such as extrapyramidal symptoms due to the stronger effect of inhibiting D2 receptor. As a result, pimozide could not lead to treat a disease by the effect of inhibiting T-type calcium channels. No compound having 1,3-dihydro-2H-benzimidazol-2-one structure, which can reduce the effect of inhibiting D2 receptors and effectively inhibit the risk of side effects, has been known yet.
As described above, some compounds, which can be used as a T-type calcium channel inhibitor, have been found. However, there is a need to further find a novel compound which can be used as the T-type calcium channel inhibitor in view of various factors such as side effects.
An object of the present invention is to find a novel compound with the effect of inhibiting T-type calcium channels and to provide a medicament useful for the treatment of a disease associated with the activation of T-type calcium channels (for example, pain, pruritus) (for example, T-type calcium channel inhibitor) which can inhibit the risk of side effects by decreasing the effect of inhibiting D2 receptor.
The present inventors have extensively studied to reach the above object, and then have found that a compound of the following formula (I) or a pharmaceutically acceptable salt thereof (hereinafter referred to as “the present compound”, as necessary) has the effect of inhibiting T-type calcium channels. In addition, the present inventors have found that the risk of side effects can be inhibited by decreasing the effect of inhibiting D2 receptor. Based on the new findings, the present invention has been completed.

According to the present invention, a novel benzimidazolone compound with an excellent effect of inhibiting T-type calcium channels can be provided. Also, the present compound can be used as a new medicament for treating a disease associated with the activation of T-type calcium channels, for example, pain and pruritus, which can reduce the effect of inhibiting D2 receptor and inhibit the risk of side effects.
Each term as used herein is explained below.
As used herein, the term “halogen” means fluorine, chlorine, bromine or iodine.
As used herein, the term “C1-10 alkyl” means a linear or branched-chain saturated hydrocarbon group having 1 to 10 carbon atoms. The terms “C1-6 alkyl” and “C1-4 alkyl” mean alkyl having 1 to 6 carbon atoms and alkyl having 1 to 4 carbon atoms, respectively.
Examples of the “C1-10 alkyl” include methyl, ethyl, propyl, isopropyl, butyl, isobutyl, sec-butyl, tert-butyl, pentyl, isopentyl, neopenthyl, hexyl, heptyl, octyl, nonyl, decyl, 1,1-dimethylbutyl, 2,2-dimethylbutyl, 3,3-dimethylbutyl, 2-ethylbutyl, and the like. They may be optionally substituted with one or more substituents of the present invention at any replaceable positions.

Examples of the “4- to 6-membered nitrogen-containing heterocycle” include azetidine, pyrrolidine, pyrroline, piperidine and piperideine. They may be optionally substituted with one or more substituents of the present invention at any replaceable positions.
As used herein, the term “optionally substituted” means a case in which the position is not substituted (non-substituted) and a case in which it is substituted at a position which can be substituted. The term “non-substituted” means that all of the replaceable positions in a group are hydrogen atoms. When a group is substituted, the group may be substituted with two or more substituents, if possible, and the substituents may be same or different.

Examples of the salt with an inorganic acid include inorganic acid salts such as hydrochloride, hydrobromide, hydroiodide, nitrate, sulfate and phosphate. Examples of the salt with an organic acid include organic acid salts such as acetate, oxalate, fumarate, maleate, succinate, malate, citrate, tartrate, adipate, gluconate, glucoheptonate, glucuronate, terephthalate, methanesulfonate, alanine, lactate, hippurate, 1,2-ethanedisulfonate, isethionate, lactobionate, oleate, gallate, pamoate, polygalacturonate, stearate, tannate, trifluoromethanesulfonate, benzenesulfonate, p-toluenesulfonate, naphthalenesulfonate and sulfosalicylate.
The compound of the present invention may be present in the form of a hydrate and/or solvate. The hydrate or the solvate such as ethanol solvate thereof is also included in the compound of the present invention. In addition, the compound of the present invention encompasses all embodiments of crystalline forms.
The compound of the present invention may be present in the form of a tautomer. The tautomer thereof is also included in the compound of the present invention.
The compound of the present invention may contain one or more asymmetric carbon atoms. The compound of the present invention encompasses not only racemic forms of the compounds of formulae (I), (Ia) and (Ib) but also optically-active forms of the compounds. Hereinafter, the processes of preparing the compound of the present invention are exemplified along with examples, but the processes of the present invention should not be limited to the examples.
Each compound used as a starting compound may be used in the salt form. The shown processes are just examples to prepare the compounds and may be optionally modified by those skilled in the organic synthesis field.
In each Preparation Process described below, protecting groups can be used as necessary, even if the use of protecting groups is not explicitly stated. And, the protecting groups can be deprotected after a reaction is completed or a series of reactions have been carried out to obtain the desired compound.
As such protecting groups, for example, general protecting groups described in T. W. Greene, and P. G. M. As appropriate, the solvent, acid and base to be used in the above preparation processes should be selected according to various factors such as the type of starting compound. Also, the conditions such as reaction temperature in the above preparation processes should be selected as appropriate.
The compound of the present invention or an intermediate thereof can be isolated and purified by the methods well known in the art. Examples of the methods include extraction, distribution, reprecipitation, column chromatography (for example, silica gel column chromatography, ion-exchange column chromatography, or preparative liquid chromatography), and recrystallization. The solvents used for recrystallization may include, for example, alcohol-based solvents such as methanol, ethanol and 2-propanol; ether-based solvents such as diethyl ether; ester-based solvents such as ethyl acetate; aromatic hydrocarbon-based solvents such as benzene and toluene; ketone-based solvents such as acetone; halogen-based solvents such as dichloromethane and chloroform; hydrocarbon-based solvents such as hexane; non-protic solvents such as dimethylformamide and acetonitrile; water; or a mixed solvent selected from two or more of the above solvents. Other purification methods, for example, those disclosed in Experimental Chemistry Textbook Vol. 1 (the Chemical Society of Japan, ed., Maruzen) can also be used herein.
As used herein, the term “disease associated with the activation of T-type calcium channels” means a disease caused due to the activation of T-type calcium channels or the associated symptom. Examples thereof include pain, epilepsy, essential tremor, schizophrenia, Parkinson's disease, manic depression, bipolar disorder, depression, anxiety, dementia, Huntington's disease, sleep disorder, stroke, pruritus, atopic dermatitis, hyperaldosteronism, edema, ischemic heart disease, age-related macular degeneration, cancer, diabetes, sterility, sexual dysfunction, arrhythmia, hypertension, renal disease and overactive bladder, but are not limited thereto. In the present invention, the disease is preferably pain and pruritus, more preferably visceral pain, somatic pain and pruritus, furthermore preferably visceral pain, acute pain, chronic pain, neuropathic pain. The pain is particularly preferably neuropathic pain.
As used herein, the term “treatment (treating)” means all types of treatment (for example, improvement, alleviation, suppression of progression) of a disease and the associated symptom in mammals, particularly in humans. Also, the term includes inhibiting the development and/or progression of a disease.
As used herein, the term “patient” means humans and animals such as dogs, cats and horses. Among them, human is preferred.
As used herein, the term “therapeutically effective amount” means an amount for treating a disease and the associated symptom or an amount for slowing progression of a disease and the associated symptom, relative to untreated subjects. The term also means an effective amount for promoting a normal physiological function.
The effective amount encompasses an amount of the present compound alone, an amount of a combination of the present compounds and/or an amount of the present compound in combination with another active ingredient useful for the treatment of a disease.
The compound of the present invention can be used in the form of pharmaceutical composition. The pharmaceutical composition of the present invention may comprise a pharmaceutically acceptable carrier.
The compound of the present invention has the effect of inhibiting T-type calcium channels. Thus, the pharmaceutical composition of the present invention can be used as a novel medicament of treating a disease such as pain (for example, visceral pain, somatic pain), epilepsy, essential tremor, schizophrenia, Parkinson's disease, manic depression, bipolar disorder, depression, anxiety, dementia, Huntington's disease, sleep disorder, stroke, pruritus (for example, intractable pruritus caused by chronic kidney disease, chronic liver disease), atopic dermatitis, hyperaldosteronism, edema, ischemic heart disease, age-related macular degeneration, cancer, diabetes, sterility, sexual dysfunction, arrhythmia, hypertension, renal disease, overactive bladder. The pharmaceutical composition of the present invention can be used as preferably a medicament for treating visceral pain, somatic pain and pruritus, more preferably a medicament for treating visceral pain, acute pain, chronic pain, neuropathic pain and pruritus, furthermore preferably a medicament for treating neuropathic pain and a medicament for treating pruritus.
The compound and pharmaceutical composition of the present invention may be administered as a preparation in an appropriate dosage form by oral administration or parenteral administration.

Examples of the pharmaceutically acceptable additive (carrier) include a stabilizing agent, a surfactant, a solubilizer, a buffer, a suspending agent, an antioxidant agent, a coating agent, a moistening agent, a moisture regulator, an algefacient, a coloring agent, a flavor, an isotonicity agent, an emulsifier, a pH adjuster, a skin protectant, a dispersant, a propellant, a fragrance, an antiseptic agent, and a solvent. The additive (carrier) can be used for the intended use.
The compound of the present invention may be used in combination with another drug as long as the drug does not prevent the therapeutic effect of the compound of the present invention against a disease, for example, pain and pruritus.
The dosage varies according to the type of compounds or various factors such as disease, age, body weight, sex, symptom of patients and administration route. The compound of the present invention is typically administrated to an adult (body weight: 60 kg) at a dose of 0.1 to 1000 mg/day, preferably 0.1 to 300 mg/day, which is administered once a day or in 2 or 3 divided doses. Also, the compound of the present invention may be administered once every few days to every few weeks.
Hereinafter, the present invention is illustrated in more detail with Reference Examples, Examples and Test Examples, but the present invention should not be limited thereto. The names of compounds used in the following Reference Examples and Examples do not necessarily follow the IUPAC nomenclature, and the identification of each compound was performed by NMR spectroscopy.
The following abbreviations may be used in Examples for the sake of simplicity. The signs used in NMR are follows: s means singlet, d means is doublet, dd is doublet of doublet, ddd means doublet of doublet of doublet, t means triplet, td means triplet of doublet, tt means triplet of triplet, q means quartet, qd means quartet of doublet, quin means quintet, sext means sextet, m means multiplet, br means broad, brs means broad singlet, and J means coupling constant.
To a solution of o-phenylenediamine (200 mg, 1.85 mmol) in N,N-dimethylformamide (5 mL) were added 4-phenylbutyl bromide (432 mg, 2.04 mmol) and then potassium carbonate (141 mg, 2.04 mmol) under Ar atmosphere, and the mixture was stirred at room temperature for 16 hours. After the reaction was completed, the reaction solution was diluted with diethyl ether (1.5 mL) and water (1.5 mL) was added thereto. The organic layer was separated, and then the aqueous layer was extracted with diethyl ether (1.5 mL×3) and combined with the separated organic layer. The organic layer was dried over anhydrous sodium sulfate, the solvent therein was removed using a rotary evaporator, and then N,N-dimethylformamide (5 mL) was added to the resulting residue. To the mixture was then added carbonyldiimidazole (600 mg, 0.37 mmol) at room temperature and the mixture was stirred at room temperature for 15 hours. After the reaction is completed, the solvent in the mixture was removed using a rotary evaporator, and then the resulting residue was purified by silica gel column chromatography (n-hexane: ethyl acetate=1:1) to give the title compound (163 mg, 33% 2 steps, 0.61 mmol).

To a solution of tert-butyl 4-(2-oxo-2,3-dihydro-1H-benzo[d]imidazol-1-yl)piperidine-1-carboxylate (1) (98 mg, 0.31 mmol) in N,N-dimethylformamide (1.5 mL) was added sodium hydride (11 mg, 60%, 0.46 mmol) under Ar atmosphere and ice temperature, and the mixture was stirred at room temperature for 30 minutes. To the mixture was then added 4-phenylbutyl bromide (78 mg, 0.37 mmol) under ice temperature, and the mixture was stirred at room temperature for 1 hour. After the reaction was completed, the reaction solution was diluted with diethyl ether (1.5 mL) and water (1.5 mL) was added thereto. The organic layer was separated, and then the aqueous layer was extracted with diethyl ether (1.5 mL×3) and combined with the separated organic layer. The organic layer was dried over anhydrous sodium sulfate, the solvent therein was removed using a rotary evaporator, and then the resulting residue was purified by silica gel column chromatography (n-hexane:ethyl acetate=2:1) to give the title compound (113 mg, 82%, 0.25 mmol).

According to the process in Reference Example 3, the compounds of Reference Examples 4 to 13 were prepared by using the corresponding alkyl bromide instead of 3-phenylpropyl bromide.
To a solution of tert-butyl 4-(6-fluoro-2-oxo-2,3-dihydro-1H-benzo[d]imidazol-1-yl)piperidine-1-carboxylate (15) (0.31 mmol) in N,N-dimethylformamide (1.5 mL) was added sodium hydride (11 mg, 60%, 0.46 mmol) under Ar atmosphere and ice temperature, and the mixture was stirred at room temperature for 30 minutes. To the mixture was then added 4-phenylbutyl bromide (79 mg, 0.37 mmol) at ice temperature and the mixture was stirred at room temperature for 1 hour.
After the reaction was completed, the reaction solution was diluted with diethyl ether (1.5 mL) and water (1.5 mL) was further added thereto. The organic layer was separated, and then the aqueous layer was extracted with diethyl ether (1.5 mL×3) and combined with the separated organic layer. The organic layer was dried over anhydrous sodium sulfate, the solvent therein was removed using a rotary evaporator, and then the resulting residue was purified by silica gel column chromatography (n-hexane:ethyl acetate=5:1 to 1:1) to give the title compound (Yield: 45%).

The compound of Reference Example 15 (Yield: 58%) was prepared according to the process in Reference Example 14 by using tert-butyl 4-(5-chloro-2-oxo-2,3-dihydro-1H-benzo[d]imidazol-1-yl)piperidine-1-carboxylate (16) instead of tert-butyl 4-(6-fluoro-2-oxo-2,3-dihydro-1H-benzo[d]imidazol-1-yl)piperidine-1-carboxylate (15).

The compound of Reference Example 17 (Yield: 69%) was prepared according to the process in Reference Example 14 by using tert-butyl 4-(7-chloro-2-oxo-2,3-dihydro-1H-benzo[d]imidazol-1-yl)piperidine-1-carboxylate (18) instead of tert-butyl 4-(6-fluoro-2-oxo-2,3-dihydro-1H-benzo[d]imidazol-1-yl)piperidine-1-carboxylate (15).

To a solution of tert-butyl 4-(2-oxo-2,3-dihydro-1H-benzo[d]imidazol-1-yl)piperidine-1-carboxylate (1) (100 mg, 0.32 mmol) in dichloromethane (1.5 mL) was added trifluoroacetic acid (0.24 mL, 3.15 mmol) under Ar atmosphere and ice temperature, and the mixture was stirred at room temperature for 3 hours. After the reaction was completed, the solvent in the mixture was removed using a rotary evaporator, and N,N-dimethylformamide (1.5 mL) was added to the resulting residue.
To the mixture were then added benzyl bromide (0.35 mmol) and then potassium carbonate (221 mg, 1.60 mmol) at room temperature, and the mixture was stirred at room temperature for 22 hours. After the reaction was completed, the reaction solution was diluted with diethyl ether (1.5 mL) and water (1.5 mL) was further added thereto. The organic layer was separated, and then the aqueous layer was extracted with diethyl ether (1.5 mL×3) and combined with the separated organic layer. The organic layer was dried over anhydrous sodium sulfate, the solvent therein was removed using a rotary evaporator, and then the resulting residue was purified by silica gel column chromatography (n-hexane:ethyl acetate=1:1 to 1:5) to give the title compound (Yield: 81%, 2 steps).
The product was analyzed by NMR. The result of NMR was completely identical to that of J. Med. Chem., 2018, 61, 8895-8907.
The title compound (Yield: 53%, 2 steps) was prepared according to the process in Example 1 by using 2-phenylethyl bromide instead of benzyl bromide.
The product was analyzed by NMR. The result of NMR was completely identical to that of the commercial compound of Aquila Pharmatech LLC (No. AQ303831).
The title compound (Yield: 78%, 2 steps) was prepared according to the process in Example 1 by using 3-phenylpropyl bromide instead of benzyl bromide.
The product was analyzed by NMR. The result of NMR was completely identical to that of J. Med. Chem., 2018, 61, 8895-8907.
The title compounds (compounds 4 and 5) were prepared according to the process in Example 1 by using 4-phenylbutyl bromide instead of benzyl bromide.
Compound 4 (Yield: 25%, 2 steps) was analyzed by NMR. The result of NMR was completely identical to that of the commercial compound of Chemieliva Pharmaceutical Co., Ltd (No. CB0902185).

To a solution of compound 4 (29 mg, 0.07 mmol) in N,N-dimethylformamide (1.5 mL) was added sodium hydride (4 mg, 60%, 0.10 mmol) under Ar atmosphere and ice temperature, and the mixture was stirred at room temperature for 30 minutes. To a solution of the compound of Reference Example 14 (0.21 mmol) in dichloromethane (1 mL) was added trifluoroacetic acid (0.16 mL, 2.14 mmol) under Ar atmosphere and ice temperature, and the mixture was stirred at room temperature for 3 hours. After the reaction was completed, the solvent in the mixture was removed using a rotary evaporator, and N,N-dimethylamide (1 mL) was added to the resulting residue. To the mixture were then added 4-phenylbutyl bromide (92 mg, 0.43 mmol) and then potassium carbonate (147 mg, 1.07 mmol) at room temperature, and the mixture was stirred at room temperature for 22 hours. After the reaction was completed, the reaction solution was diluted with diethyl ether (1.5 mL) and water (1.5 mL) was further added thereto. The organic layer was separated, and then the aqueous layer was extracted with diethyl ether (1.5 mL×3) and combined with the separated organic layer. The organic layer was dried over anhydrous sodium sulfate, the solvent therein was removed using a rotary evaporator, and then the resulting residue was purified by silica gel column chromatography (n-hexane:ethyl acetate=5:1 to 1:2) to give the title compound (Yield: 42%, 2 steps).

To a solution of the compound of Reference Example 2 (63 mg, 0.14 mmol) in dichloromethane (1.5 mL) was trifluoroacetic acid (0.11 mL, 1.40 mmol) under Ar atmosphere and ice temperature, and the mixture was stirred at room temperature for 3 hours. After the reaction was completed, the solvent in the mixture was removed using a rotary evaporator, and N,N-dimethylamide (1.5 mL) was added to the resulting residue. The organic layer was separated, and then the aqueous layer was extracted with diethyl ether (1.5 mL×3) and combined with the separated organic layer. The organic layer was dried over anhydrous sodium sulfate, the solvent therein was removed using a rotary evaporator, and then the resulting residue was purified by silica gel column chromatography (n-hexane:ethyl acetate=5:1 to 1:2) to give the title compound (Yield: 100%, 2 steps).

The effect of the compound of the present invention on human Cav3.2 T-type calcium channels was evaluated by whole-cell patch clamp method. As a control (vehicle), DMSO solution was used.
Human kidney cell-derived cell line, HEK 293 cells, in which human Cav3.2 T-type calcium channels were forcibly expressed, were seeded into cell culture dishes using DMEM medium containing 10% fetal bovine serum and cultured at 37° C. under 5% CO2 conditions. In the whole-cell patch clamp method, the following solutions were used as extracellular and intracellular solutions, the holding potential is set at −80 mV, and the barium current flowing as an inward current in a potential jump from −80 mV to −20 mV was measured with a patch clamp amplifier and AD converter (Axon Instruments). In order to eliminate the influence of high-threshold activated calcium channels, the value obtained by subtracting the current at 150 ms after the start of stimulation from the peak current was used for data analysis as the T channel current (T-current). The data analysis was performed using pClamp8 (Axon Instruments).
As for test compounds, each compound dissolved in extracellular solution was added directly to the dishes (direct addition method) or added to the dishes by perfusion (perfusion addition method).

According to a similar method to the perfusion addition method of Test Example 1, the effect of each compound of the present invention on human Cav3.2 T-type channels was evaluated. As the test compounds, pimozide at each concentration of 0.01 μM, 0.1 μM, 0.3 μM, 1.0 μM and 3.0 μM, compound 5, compound 14 and compound 15 were used and the test compounds were added by perfusion.
The test results are shown in Table 2. Table 2 shows the IC50 values (μM) analyzed and calculated with Prism(GraphPad Software Inc.) from the percentage change in the T-channel current values before and after the addition of the test compound (n=3 to 17).
The above test results showed that the compound of the present invention had the excellent effect of inhibiting T-type calcium channels.
In this test, the binding ability of the present compound to dopamine D2 receptor was studied to evaluate the dopamine D2 receptor binding inhibitory effect of each compound. Compounds 4 to 7, 25, 26, 28, 30, 36 and 38 were used as the present compound, and pimozide and sulpiride were used as the comparative control.
The following reaction solution containing rat brain striatum homogenate was used, 5 μL of the present compound, sulpiride or pimozide was added to adjust the final concentration of each compound to 1 or 10 μM, and they were reacted for 90 minutes at room temperature. After the reaction was completed, the reaction solution was filtered through glass fiber filter, and the filter was washed and dried to measure the radioactivity thereof. The binding ability of each compound to dopamine D2 receptor was evaluated by the percentage inhibition of the binding between dopamine D2 receptor and [3H]-spiperone and the amount of [3H]-spiperone bond.

As shown in the test results, the present compound did not inhibit the binding between dopamine D2 receptor and [3H]-spiperone as compared to pimozide and sulpiride. Hence, it was shown that the present compound did not have any dopamine D2 receptor binding inhibitory effect, that is, the present compound could reduce the dopamine D2 receptor binding effect.
In this test, the catalepsy-inducing effect of the present compound was studied to evaluate whether the present compound can inhibit side effects caused by the dopamine D2 receptor blockade. Compound 5 and compound 12 were used as the present compound and 2% DMSO solution was used as vehicle. Also, pimozide was used as the control drug.
16 male ddY mice were randomly divided into compound 5 administration group, compound 12 administration group, pimozide administration group and vehicle administration group (4 mice per group). The mice in each group were received the administration of 8 nmol (0.8 mM/10 μL) of compound 5, compound 12 and pimozide, and 10 μL of vehicle in the lateral ventricle. both forepaws of the mice were forced to place themselves on a horizontal iron bar (Height: 5 cm) and the time of keeping the posture (catalepsy) was measured to evaluate the presence or absence of catalepsy. The measurement was terminated when the forepaws touched the floor or when the mouse was on the bar.

In this test, the antipruritic and analgesic effects of the present compound was studied using mouse Na2S intrabuccal injection pruritus models. Compound 5 was used as the present invention and saline comprising 1% DMSO and 5% Tween 80 was used as vehicle.
Male ICR mice were received the intrabuccal injection of 3.8 μmol (300 μg/10 μL) of Na2S and T-type calcium channel-independent pain was induced to prepare mouse Na2S intrabuccal injection pruritus models. 18 mice were randomly divided into vehicle+saline administration group (control group), vehicle+Na2S administration group and compound 5 (10 mg/kg)+Na2S administration group (6 mice per group). The mice in each group were anesthetized with isoflurane and their right cheek hairs were shaved the day or a few days before the experiment. On the day of the experiment, the mice were placed in an acrylic observation cage with mirrors on all four sides and allowed to acclimate the environment for at least 30 minutes. After the acclimatization, Na2S was intradermally administered to the shaved cheeks of the mice without anesthesia, and their subsequent behavior was recorded on video. Compound 5 (10 mg/kg) or vehicle (V, 10 mL/kg) was intraperitoneally administered 30 minutes before Na2S was administered. At the later date, the video was replayed and the number of occurrences of the scratching bouts by the hind paws to the administration site (as pruritus response) and the wipes (wiping) by the forepaws (as pain response) was counted for 60 minutes to evaluate the antipruritic and analgesic effects of the present compound.

Male ICR mice were received the intraplantar injection of 10 μmol/paw/10 μL of Na2S and T-type calcium channel-dependent pain was induced to prepare mouse Na2S intraplantar injection pain models. The mice were randomly divided into the vehicle+vehicle administration group (control group), vehicle+Na2S administration group, compound 5 (1 mg/kg)+Na2S administration group, compound 5 (3 mg/kg)+Na2S administration group and compound 5 (10 mg/kg)+Na2S administration group (n=7 to 11). Vehicle (V, 10 mL/kg) or 1, 3 or 10 mg/kg of compound 5 was intraperitoneally administered 30 minutes before Na2S was injected intraplantarlly in the hind paws of the mice.
Pain thresholds were measured every 15 minutes until 60 minutes after the administration of Na2S by the von Frey method to evaluate the analgesic effect of the present compound.

In this test, the antipruritic effect of the present compound was studied using mouse intrabuccal injection pruritus models in which histamine or chloroquine is intra-buccally administered as a pruritogen. Compound 5 and compound 38 were used as the present compound, and saline comprising 1% DMSO and 5% Tween 80 was used as control. Compound 5 and compound 38 were dissolved into saline comprising 1% DMSO and 5% Tween 80 to adjust the concentration of each compound. According to the method in Shimada SG & LaMotte R H (2008) Behavioral differentiation between itch and pain in mouse. Pain 139 (3): 681-687, histamine or chloroquine, which is a pruritogen, was dissolved into saline and the solution was administered into the buccal skin of each male ICR mouse and pruritus was induced to prepare mouse intrabuccal injection pruritus models. The mice were randomly divided into saline comprising 1% DMSO and 5% Tween 80 (Solvent 1)+saline (Solvent 2) administration group (control group), Solvent 1+histamine administration group, compound 5 (10 mg/kg)+histamine administration group, Solvent 1+chloroquine administration group, compound 5 (10 mg/kg)+chloroquine administration group, compound 38 (1 mg/kg)+chloroquine administration group, compound 38 (3 mg/kg)+chloroquine administration group and compound 38 (10 mg/kg)+chloroquine administration group (n=6 to 10). The mice in each group were anesthetized with isoflurane and their right cheek hairs were shaved a few days before the experiment. On the day of the experiment, the mice were placed in an acrylic observation cage (10×14×30 cm) with mirrors on all four sides and allowed to acclimate the environment for at least 30 minutes. After the acclimatization, saline in a volume of 10 μL/site, histamine in a volume of 300 μL/site or chloroquine in a volume of 100 μL/site was intradermally administered to the shaved cheeks of the mice without anesthesia, and their subsequent behavior was recorded on video for 60 minutes. Compound 5 (10 mg/kg), compound 38 (1 mg/kg, 3 mg/kg or 10 mg/kg) or Solvent 1 (10 mL/kg) was intraperitoneally administered 30 minutes before saline or the pruritogen was administered. At the later date, the video was replayed and the number of occurrences of the scratching bouts by the hind paws to the administration site (as pruritus response) was counted for 60 minutes to evaluate the antipruritic effect of the present compound.

The present compound strongly inhibited both histamine-dependent pruritus and histamine-independent pruritus. As a result, it was suggested that the present compound could be used as a medicament for treating intractable pruritus.
According to the present invention, a new benzimidazolone compound with the excellent effect of inhibiting T-type calcium channels can be provided. Also, the present compound can be used as a new medicament for treating a disease associated with the activation of T-type calcium channels, for example, pain and pruritus, which can reduce the effect of inhibiting D2 receptor and inhibit the risk of side effects.
