Claims
- 1. A therapeutic method, comprising treating procedural vascular trauma by administering to a mammal a cytostatic dosage of a cytoskeletal inhibitor effective to inhibit the proliferation of vascular smooth muscle cells, wherein the vessel is traumatized by angioplasty, placement of a stent, or grafting.
- 2. The method of claim 1 wherein the cytoskeletal inhibitor is a cytochalasin or an analog thereof.
- 3. The method of claim 1 wherein the administration is local.
- 4. The method of claim 1 wherein the administration is systemic.
- 5. The method of claim 1 wherein the administration is before, during or after the trauma.
- 6. The method of claim 1 wherein the administration is during the trauma.
- 7. The method of claim 1 wherein the administration of the cytoskeletal inhibitor is accomplished by a device.
- 8. The method of claim 7 wherein the device is a catheter.
- 9. The method of claim 1 wherein the amount is effective to inhibit migration of vascular smooth muscle cells.
- 10. The method of claim 1 wherein the cytoskeletal inhibitor is in a sustained release dosage form.
- 11. The method of claim 10 wherein the cytoskeletal inhibitor is administered in a polymeric carrier.
- 12. The method of claim 10 wherein the sustained release dosage form is biodegradable.
- 13. The method of claim 1 or 10 wherein the cytoskeletal inhibitor is in a sustained release dosage form and is administered by a device.
- 14. The method of claim 10 wherein the sustained release form comprises a binding peptide or protein which specifically binds to smooth muscle cells, stromal cells or interstitial matrix surrounding smooth muscle cells.
- 15. The method of claim 10 wherein the sustained release dosage form comprises microparticles or nanoparticles.
- 16. The method of claim 15 wherein the sustained release dosage form comprises a polymer derived from the condensation of alpha-hydroxycarboxylic acids and related lactones.
- 17. The method of claim 16 wherein the polymer is selected from the group consisting of a polylactide, a polyglycolide, and a copolymer of lactide and glycolide subunits.
- 18. The method of claim 17 wherein the polymer is poly(lactide co-glycolide).
