Claims
- 1. An isolated nucleotide sequence encoding a mutant of a wild-type D-N-.alpha.-carbamoylase,
- wherein the wild-type D-N-.alpha.-carbamoylase comprises the amino acid sequence of SEQ ID NO: 1,
- wherein the amino acid at position 184 of the mutant is not methionine, the amino acid at position 212 of the mutant is not threonine, the amino acid at position 262 of the mutant is not threonine, or the amino acid at position 304 of the mutant is not phenylalanine,
- wherein the amino acid at position 184 of the mutant, the amino acid at position 212 of the mutant, the amino acid at position 262 of the mutant, and the amino acid at position 304 of the mutant are each a natural amino acid.
- 2. The isolated nucleotide sequence of claim 1, wherein the amino acid at position 184 of the mutant is L-leucine.
- 3. The isolated nucleotide sequence of claim 1, wherein the amino acid at position 304 of the mutant is L-isoleucine.
- 4. The isolated nucleotide sequence of claim 1, wherein the amino acid at position 212 of the mutant is L-alanine.
- 5. The isolated nucleotide sequence of claim 1, wherein the amino acid at position 262 of the mutant is L-alanine.
- 6. The isolated nucleotide sequence of claim 1, wherein the amino acid at position 184 of the mutant is not methionine, and the amino acid at position 304 of the mutant is not phenylalanine.
- 7. The isolated nucleotide sequence of claim 1, wherein the amino acid at position 184 of the mutant is not methionine, and the amino acid at position 212 of the mutant is not threonine.
- 8. An expression vector comprising the nucleotide sequence of claim 1.
- 9. The expression vector of claim 8, wherein the expression vector is selected from the group consisting of pSM754, pSM755, pSM756, pSM761, pSM762, pSM763, pSM764, pSM765, pSM770, and pSM771.
- 10. The expression vector of claim 8, wherein the expression vector is pSM754 having deposit number CBS665.95.
- 11. The expression vector of claim 8, wherein the expression vector is pSM755 having deposit number CBS666.95.
- 12. The expression vector of claim 8, wherein the expression vector is pSM756 having deposit number CBS667.95.
- 13. The expression vector of claim 8, wherein the expression vector is pSM769 having deposit number CBS758.95.
- 14. A microorganism transformed with the expression vector of claim 8, wherein the microorganism is Bacillus subtilis or Escherichia coli.
- 15. The microorganism of claim 14, wherein the microorganism is Escherichia coli SMC341 (pSM754) CBS 665.95.
- 16. The microorganism of claim 14, wherein the microorganism is Escherichia coli SMC342 (pSM755) CBS 666.95.
- 17. The microorganism of claim 14, wherein the microorganism is Escherichia coli SMC343 (pSM756) CBS 667.95.
- 18. The microorganism of claim 14, wherein the microorganism is Escherichia coli SMC355 (pSM769) CBS 758.95.
- 19. A method for the preparation of a mutant of a wild-type D-N-.alpha.-carbamoylase,
- wherein the wild-type D-N-.alpha.-carbamoylase comprises the amino acid sequence of SEQ ID NO: 1,
- wherein the amino acid at position 184 of the mutant is not methionine, the amino acid at position 212 of the mutant is not threonine, the amino acid at position 262 of the mutant is not threonine, or the amino acid at position 304 of the mutant is not phenylalanine,
- wherein the amino acid at position 184 of the mutant, the amino acid at position 212 of the mutant, the amino acid at position 262 of the mutant, and the amino acid at position 304 of the mutant are each a natural amino acid,
- wherein the method comprises:
- (a) culturing the microorganism of claim 14 in a medium, and
- (b) separating and purifying the mutant of the wild-type D-N-.alpha.-carbamoylase.
- 20. A process for the production of D-.alpha.-amino acids by the stereospecific conversion of D-N-.alpha.-carbamoyl amino acids or by the stereospecific conversion of racemic mixtures of 5-substituted hydantoins,
- a mutant of a wild-type D-N-.alpha.-carbamoylase or a mutant of a wild-type D-N-.alpha.-carbamoylase and a D-hydantoinase,
- wherein the wild-type D-N-.alpha.-carbamoylase comprises the amino acid sequence of SEQ ID NO: 1,
- wherein the amino acid at position 184 of the mutant is not methionine, the amino acid at position 212 of the mutant is not threonine, the amino acid at position 262 of the mutant is not threonine, or the amino acid at position 304 of the mutant is not phenylalanine,
- wherein the amino acid at position 184 of the mutant, the amino acid at position 212 of the mutant, the amino acid at position 262 of the mutant, and the amino acid at position 304 of the mutant are each a natural amino acid.
- 21. The method of claim 20, wherein the stereospecific conversion is carried out in the presence of a microorganism capable of producing the mutant of the wild-type D-N-.alpha.-carbamoylase or capable of producing the mutant of the wild-type D-N-.alpha.-carbamoylase and the D-hydantoinase.
- 22. The method of claim 20, wherein the mutant of the wild-type D-N-.alpha.-carbamoylase is immobilized on an insoluble solid carrier, or the mutant of the wild-type D-N-.alpha.-carbamoylase and the D-hydantoinase are immobilized on an insoluble solid carrier.
