Claims
- 1. A dual compartment system for transdermally delivering an active agent to the skin of a wearer comprising:
- (a) a solvent reservoir compartment defined by walls and one closed and one open end and containing ethanol as a solvent,
- (b) a drug compartment having walls and opposing open ends and containing said active agent,
- (c) an epoxidized silicone rubber containing membrane having opposing surfaces, one surface being attached to and overlying the open end of said solvent reservoir compartment and the other surface being attached to and overlying one open end of said drug compartment thereby separating said solvent reservoir from said drug compartment by said membrane, said membrane being permeable to ethanol from said solvent reservoir and substantially impermeable to drug and water from said drug compartment; and
- (d) a second membrane having opposing surfaces and being permeable to said ethanol and drug, one surface being attached to and overlying the other open end of said drug compartment said second membrane having adhesive means on the other surface thereof for attaching said delivery system to the skin of a wearer.
- 2. A delivery system according to claim 1 wherein said epoxidized silicone rubber containing membrane is an epoxidized silicon rubber consisting of a copolymer of about 90-98% w a polydimethyl siloxane and about 2-10% w of a polyglycerol-polyglycidyl ether.
- 3. A delivery system according to claim 2 wherein said polyglycerol-polyglycidyl ether has the formula ##STR3## wherein z is an integer of 2 to 3.
- 4. A delivery system according to claim 3 wherein said epoxidized silicone rubber containing membrane is a bilaminate consisting of a layer of said epoxidized silicone rubber and a layer of a dense hydrophobic copolymer of vinyl-terminated polydimethyl siloxane with methylhydro-dimethyl siloxane.
- 5. A delivery system according to claim 4 wherein said dense hydrophobic copolymer contains about 90 to 98% w. of the vinyl-terminated polydimethyl siloxane and about 2 to 10% w. of the methylhydro-dimethyl siloxane.
- 6. A delivery system according to claim 5 wherein said epoxidized silicone rubber containing membrane has a thickness of between about 0.4 and 1.0 mm and wherein said epoxidized silicone rubber portion of the membrane faces the solvent reservoir and the dense hydrophobic copolymer portion of the membrane faces the drug compartment.
- 7. A delivery system according to claim 6 wherein said epoxidized silicone rubber portion of said epoxidized silicone rubber containing membrane is thicker than said dense hydrophobic copolymer portion of said membrane.
- 8. A delivery system according to claim 7 wherein said active agent is soluble in said ethanol solvent.
- 9. A delivery system according to claim 1 wherein said drug compartment contains said active agent and aqueous ethanol.
- 10. A delivery system according to claim 3 wherein said epoxidized silicone rubber containing membrane is a trilaminate consisting of a layer of said epoxidized silicone rubber, an intermediate layer of cellulose acetate and a layer of a dense hydrophobic copolymer of vinyl-terminated polydimethyl siloxane with methylhydro-dimethyl siloxane.
- 11. A delivery system according to claim 10 wherein said dense hydrophobic copolymer contains about 90 to 98% w. of the vinyl-terminated polydimethyl siloxane and about 2 to 10% w. of the methylhydro-dimethyl siloxane.
- 12. A delivery system according to claim 11 wherein said epoxidized silicone rubber containing membrane has a thickness of between about 0.4 and 1.0 mm and wherein said epoxidized silicone rubber portion of the membrane faces the solvent reservoir and the dense hydrophobic copolymer portion of the membrane faces the drug compartment.
- 13. A delivery system according to claim 12 wherein said epoxidized silicone rubber portion of said epoxidized silicone rubber containing membrane is thicker than said dense hydrophobic copolymer portion of said membrane.
- 14. A delivery system according to claim 13 wherein said cellulose acetate layer has a thickness of about 10 to 50 microns.
- 15. A delivery system according to claim 14 wherein said active agent is soluble in said ethanol solvent.
- 16. A delivery system according to claim 1 wherein said drug compartment contains said active agent and aqueous ethanol.
- 17. A delivery system according to claim 16 wherein said active agent is a hydrophobic drug.
- 18. A silicone rubber containing membrane permeable to lower alkanol solvents and substantially impermeable to water and hydrophobic solutes comprising
- (a) a first layer comprising an epoxidized silicone rubber consisting of a copolymer of about 90-98% w a polydimethyl siloxane and about 2-10% w of a polyglycerol-polyglycidyl ether having the formula ##STR4## wherein z is an integer of 2 or 3, and (b) a second layer of a dense hydrophobic copolymer containing about 90 to 98% w. of the vinyl-terminated polydimethyl siloxane and about 2 to 10% w. of the methylhydro-dimethyl siloxane.
- 19. A silicone rubber containing membrane according to claim 1 wherein said epoxidized silicone rubber first layer portion of said silicone rubber containing membrane is thicker than said dense hydrophobic copolymer second layer portion of said membrane.
- 20. A silicone rubber containing membrane according to claim 19 wherein said membrane has a thickness of between about 0.4 and 1.0 mm and wherein the epoxidized silicone rubber first layer portion is about twice as thick as said dense hydrophobic copolymer second layer portion.
- 21. A silicone rubber containing membrane according to claim 19 containing a layer of hydrophilic cellulose acetate sandwiched between said epoxidized silicone rubber first layer portion and said dense hydrophobic copolymer second layer portion.
- 22. A silicone rubber containing membrane according to claim 21 wherein said cellulose layer has a thickness of between about 10 and 50 microns.
