Claims
- 1. A transgenic non-human animal comprising a recombinant nucleic acid molecule stably integrated into the genome of said animal, said molecule encoding a light-generating fusion protein comprising a ligand binding site and a light-generating polypeptide moiety.
- 2. The animal of claim 1, whereupon ligand binding to said ligand binding site alters the luminescence of said light-generating polypeptide moiety.
- 3. The animal of claim 1, wherein said ligand binding site comprises a polypeptide selected from the group consisting of (a) a polypeptide comprising SEQ ID NO: 9; (b) a polypeptide comprising amino acids 556 to 575 of SEQ ID NO: 9; and (c) a polypeptide comprising amino acids 530-625 of SEQ ID NO: 9.
- 4. The animal of claim 1, wherein said recombinant nucleic acid molecule is operably linked to one or more regulatory sequences.
- 5. The animal of claim 4, wherein said further regulatory sequence is a promoter.
- 6. The animal of claim 1, wherein said recombinant nucleic acid molecule is of human or murine origin.
- 7. The animal of claim 1, wherein said light-generating polypeptide moiety is selected from the group consisting of ferredoxin IV, green fluorescent protein, red fluorescent protein, yellow fluorescent protein, blue fluorescent protein, the luciferase family and the aequorin family.
- 8. An isolated cell of the animal of claim 1.
- 9. The cell of claim 8, wherein said cell is a stem cell, a germ cell, a precursor cell or a progenitor cell.
- 10. A transgenic non-human animal comprising a recombinant nucleic acid molecule stably integrated into the genome of said animal, said recombinant nucleic acid molecule comprising SEQ ID NO:25.
- 11. The animal of claim 10, wherein said animal is a mouse.
- 12. A method for the production of a transgenic non-human animal, comprising introduction of a recombinant nucleic acid molecule into a germ cell, an embryonic cell, an egg cell or a cell derived therefrom, said recombinant nucleic acid molecule encoding a light-generating fusion protein comprising a ligand binding site and a light-generating polypeptide moiety
- 13. The method of claim 12, wherein said animal is a rodent.
- 14. The method of claim 13, wherein said rodent is a mouse.
- 15. A method for the identification of a compound capable of modifying an activity of HIF1α, comprising:
(a) contacting the transgenic non-human animal of claim 1 or a cell thereof with a test compound; and (b) measuring the effect of said test compound on said light-generating moiety; thereby identifying a compound that modifies an activity of HIF1α.
- 16. The method of claim 15, wherein said compound decreases HIF1α stability.
- 17. The method of claim 15, wherein said compound increases HIF1α stability.
- 18. A method of detecting hypoxic tissue, comprising:
(a) administering luciferin to the mouse of claim 11; and (b) measuring with a photodetector device, photon emission through opaque tissue, thereby detecting hypoxic tissue in said mouse.
