Claims
- 1. A method for preparing a type II proteinaceous, antigenic factor derived form a group A streptococcus selected from the group consisting of (1) a factor which is a receptor for the Fc region of human IgG.sub.3 and which exhibits a major diffuse protein band on polyacrylamide gel electrophoresis and which has a molecular weight of approximately 38,000 daltons and (2) a factor which is a receptor for the Fc region of human IgG.sub.1, IgG.sub.2 and IgG.sub.4 and which exhibits a major diffuse protein band on polyacrylamide gel electrophoresis and which has a molecular weight of approximately 56,000 daltons comprising:
- 1) solubilizing said factor from a group A streptococcus,
- 2) purifying said factor by affinity chromatography on an immobilized IgG sub-class or sub-classes, and
- 3) separating said factors.
- 2. The method of claim 1 including the step of chromatographically separating said solubilized factors from said solution.
- 3. The method of claim 1, including the step of increasing the Fc-receptor expression in said group A streptococcus by serial passage thereof in a mouse.
- 4. The method of claim 1 wherein said affinity purification step 2) comprises:
- a) applying said factor to a column comprising said IgG sub-class or sub-classes covalently coupled to an inert substrate and eluting unbound material therefrom, and
- b) eluting bound factor from said column.
- 5. The method of claim 4 including the additional steps of concentrating said eluted solution of factor from said column of immobilized IgG sub-class or sub-classes to from about 40 .mu.g/ml to about 400 .mu.g/ml.
- 6. The method of claim 2 wherein said separation of factors is effected by polyacrylamide gel electrophoresis.
