Claims
- 1. Method for determining the presence of valproate in a physiological fluid which comprises combining in an aqueous assay medium a sample of said physiological fluid,
- antibodies prepared in response to an antigen of the formula: ##STR5## wherein: R is an alkylene group of from 1 to 6 carbon atoms;
- m.sup.2 is 0 or 1;
- Z.sup.2 is a poly(amino acid);
- n.sup.2 is at least 1 and not greater than the molecular weight of Z.sup.2 divided by 500; and
- when m.sup.2 is 0, W.sup.4 is methylene bonded to amino groups of Z.sup.2 and when m.sup.2 is 1, W.sup.4 is a bond to amino groups of Z.sup.2 to form amide linkages,
- an enzyme conjugate of the formula: ##STR6## wherein R is an alkylene group of from 1 to 6 carbon atoms;
- m.sup.2 is 0 or 1;
- Z.sup.2 is an NAD dependent enzyme;
- n.sup.2 is at least 1 and not greater than the molecular weight of Z.sup.2 divided by 500; and
- when m.sup.2 is 0, W.sup.4 is methylene bonded to amino groups of Z.sup.2 and when m.sup.2 is 1, W.sup.4 is a bond to amino groups of Z.sup.2 to form amide linkages, NAD and remaining substrate for said enzyme, and determining the rate of transformation from NAD to NADH as compared to the rate determined with an assay medium having a known amount of valproate.
- 2. Antibodies prepared in response to an antigen conjugate of the formula: ##STR7## wherein: R is an alkylene group of from 1 to 6 carbon atoms;
- m.sup.2 is 0 or 1;
- Z.sup.2 is a poly(amino acid);
- n.sup.2 is at least 1 and not greater than the molecular weight of Z.sup.2 divided by 500; and
- when m.sup.2 is 0, W.sup.4 is methylene bonded to amino groups of Z.sup.2 and when m.sup.2 is 1, W.sup.4 is a bond to amino groups of Z.sup.2 to form amide linkages, which antibodies will bind to valproate, antigen conjugate of the above formula, and enzyme conjugate of the formula ##STR8## wherein: R is an alkylene group of from 1 to 6 carbon atoms;
- m.sup.2 is 0 or 1;
- Z.sup.2 is an enzyme;
- n.sup.2 is at least 1 and not greater than the molecular weight of Z.sup.2 divided by 500; and
- when m.sup.2 is 0, W.sup.4 is methylene bonded to amino groups of Z.sup.2 and when m.sup.2 is 1, W is a bond to amino groups of Z.sup.2 to form amide linkages.
