Claims
- 1. An isolated polynucleotide encoding a substantially purified transmembrane segment-free polypeptide having α1,3-fucosyltransferase activity, or enzymatically active portions thereof, and its complement.
- 2. The polynucleotide of claim 1, wherein the polynucleotide encodes a polypeptide having α1,2-fucosyltransferase activity or α1,4-fucosyltransferase activity.
- 3. A polynucleotide selected from the group consisting of:
a) SEQ ID NO:4; b) SEQ ID NO:4, wherein T is U: c) nucleic acid sequences complementary to a) or b); and d) fragments of a), b), or c) that are at least 15 nucleotide bases in length and that hybridize to DNA which encodes any one of the polypeptide set forth in SEQ ID NO:1, SEQ ID NO:2 and SEQ ID NO:3.
- 4. A vector containing the polynucleotide of claim 1.
- 5. A host cell containing the vector of claim 4.
- 6. A recombinant method for producing a transmembrane segment-free α1,3-fucosyltransferase polypeptide, comprising:
inserting a nucleic acid encoding the polynucleotide of claim 1 adjacent to a selectable marker, such that the resulting polynucleotide encodes a recombinant α1,3-fucosyltransferase polypeptide fused to the selectable marker.
- 7. A polynucleotide produced by the method of claim 6.
- 8. A host cell containing the polynucleotide of claim 6.
- 9. A recombinant method for producing a transmembrane segment-free α1,3-fucosyltransferase polypeptide, comprising:
a) growing a recombinant host cell containing a polynucleotide of claim 1 encoding a polypeptide having α1,3-fucosyltransferase activity under conditions which allow expression and secretion of α1,3-fucosyltransferase polypeptide; and b) isolating the polypeptide.
- 10. A method of producing a transmembrane segment-free α1,3-fucosyltransferase-fusion protein comprising:
a) growing a host cell containing a polynucleotide of claim 1 encoding a polypeptide having α1,3-fucosyltransferase activity operably linked to a polynucleotide encoding a polypeptide or peptide of interest under conditions which allow expression and secretion of the fusion protein; and b) isolating the fusion protein.
- 11. A gene expression system for producing a transmembrane segment-free α1,3-fucosyltransferase comprising a host cell modified with a polynucleotide of claim 1 encoding a polypeptide having α1,3-fucosyltransferase activity or an enzymatically active portion thereof.
- 12. The gene expression system of claim 11, wherein the polynucleotide is DNA, cDNA, or RNA.
- 13. The gene expression system of claim 11, wherein the host cell is selected from the group consisting of a bacterial cell, a yeast cell, a fungal cell, a plant cell, an insect cell, or an animal cell.
- 14. The gene expression system of claim 11, wherein the host cell is recombinantly modified by transfection with a plasmid.
- 15. The gene expression system of claim 14, wherein the plasmid comprises a selectable marker.
- 16. The gene expression system of claim 15, wherein the selectable marker is glutamine synthetase.
- 17. A method for producing a transmembrane segment-free α1,3-fucosyltransferase polypeptide, comprising the steps of:
a) culturing a gene expression system comprising a host cell modified with a polynucleotide of claim 1 encoding a polypeptide having α1,3-fucosyltransferase activity or an enzymatically active portion thereof; and b) harvesting the α1,3-fucosyltransferase.
- 18. The method of claim 17, further comprising substantially purifying the harvested α1,3-fucosyltransferase polypeptide.
- 19. The method of claim 17, wherein the host cell is recombinantly modified by transfection with a plasmid.
- 20. The method of claim 17, wherein the host cell is selected form the group consisting of a bacterial cell, a yeast cell, a fungal cell, a plant cell, an insect cell, or an animal cell.
