Claims
- 1. An immunogenic composition, comprising an immunoeffective amount of an isolated and purified nucleic acid molecule comprising a mutant Haemophilus hin47 gene encoding an analog of Haemophilus influenzae Hin47 protein having a decreased protease activity which is less than about 10% of that of natural Hin47 protein and having substantially the immunogenic properties of natural Hin47 protein.
- 2. The immunogenic composition of claim 1 formulated as a vaccine for in vivo administration to a host to confer protection against diseases caused by a bacterial pathogen that produces Hin47 protein or a protein capable of inducing antibodies in the host specifically reactive with Hin47 protein.
- 3. The immunogenic composition of clam 1 wherein the bacterial pathogen is a Haemophilus species.
- 4. The immunogenic composition of claim 3 wherein the Haemophilus species is Haemophilus influenzae.
- 5. The immunogenic composition of claim 2 further comprising at least one other immunogenic or immunostimulating material.
- 6. A method of generating an immune response in a host comprising administering thereto an immuno-effective amount of the immunogenic composition of claim 1.
- 7. The composition of claim 1 wherein at least one codon of a wild-type hin47 gene encoding an amino acid contributing to protease activity and which is the codon encoding an amino acid selected from the group consisting of amino acids 91, 121 and 195 to 201, has been deleted or replaced by a codon encoding different amino acid to provide the reduced protease activity.
- 8. The composition of claim 1 wherein the at least one codon is that encoding Serine-197.
- 9. The composition of claim 8 wherein the codon encoding Serine-197 is replaced by a codon encoding alanine.
- 10. The composition of claim 1 wherein the codon encoding His-91 is replaced by a codon encoding alanine, lysine or arginine.
- 11. The composition of claim 1 wherein the codon encoding Asp-121 is replaced by a codon encoding alanine.
- 12. The composition of claim 1 wherein said mutant gene is formed by site-directed mutagenesis of a wild-type hin47 gene.

No. 5,869,302) filed on Jun. 7, 1995 which itself is a continuation-in-part of copending patent application Ser. No. 08/296,149 filed Aug. 26, 1994, (now U.S. Pat. No. 5,939,297) which itself is a continuation-in-part of U.S. patent application Ser. No. 08/278,091 filed on Jul. 21, 1994 (now U.S. Pat. No. 5,506,139).
