Claims
- 1. A C5a anaphylotoxin activity modulating compound of the formula:
- A-B-D-E-G-J-L-M-Q-T
- or a pharmaceutically acceptable salt thereof wherein the groups A through T have the values:
- A is R.sub.1 -R.sub.2 -R.sub.3 ;
- B is selected from the group consisting of R.sub.4 -R.sub.5 -R.sub.6, R.sub.31, R.sub.32, R.sub.35 and R.sub.37 ;
- D is selected from the group consisting of R.sub.7 -R.sub.8 -R.sub.9, R.sub.31, R.sub.32, R.sub.35 and R.sub.37 ;
- E is selected from the group consisting of R.sub.10 -R.sub.11 -R.sub.12, R.sub.31, R.sub.32, R.sub.35 and R.sub.37 ;
- G is selected from the group consisting of R.sub.13 -R.sub.14 -R.sub.15, R.sub.31, R.sub.32, R.sub.35 and R.sub.37 ;

- D and E, taken together, optionally represent a group selected from the group consisting of R.sub.33, R.sub.34, R.sub.38, R.sub.39, R.sub.40, R.sub.41, R.sub.42, and R.sub.43 ;

- M and Q, taken together, optionally represent a group selected from the group consisting of R.sub.33, R.sub.34, R.sub.38, R.sub.39, R.sub.40, R.sub.41, R.sub.42, and R.sub.43, and one or more of the groups R.sub.5 -R.sub.6 -R.sub.7, R.sub.8 -R.sub.9 -R.sub.10, R.sub.11 -R.sub.12 -R.sub.13, R.sub.14 -R.sub.15 -R.sub.16, R.sub.17 -R.sub.18 -R.sub.19, R.sub.20 -R.sub.21 -R.sub.22, R.sub.23 -R.sub.24 -R.sub.25, or R.sub.26 -R.sub.27 -R.sub.28, independently may optionally represent R.sub.36 ; wherein
- (a) R.sub.1 is selected from the group consisting of amino,(lower alkyl)amino, dialkylamino, (arylalkyl)amino, hydroxy, alkoxy, arloxy, arylalkoxy, acetamido, thioalkoxy, halogen, aryl, lower alkyl, arylalkyl, (heterocyclic)alkyl, heterocyclic, arylamino, and hydrogen;
- (b) R.sub.2 is selected from the group consisting of >CR.sub.99 R.sub.100, >C.dbd.CR.sub.95 R.sub.96, existing in either the Z- or E-configuration, oxygen, amino, and alkylamino, with the proviso that when R.sub.2 is oxygen, amino or alkylamino, R.sub.1 is aryl, lower alkyl, arylalkyl or (heterocyclic)alkyl;
- (c) R.sub.3 is selected from the group consisting of >C.dbd.O,>CH.sub.2, >C.dbd.S, >SO.sub.2, with the proviso that when R.sub.3 is >CH.sub.2 or >SO.sub.2 then R.sub.2 cannot be oxygen, amino or alkylamino;
- (d) R.sub.4 is selected from the group consisting of >CH.sub.2,>O,>S, and >NR.sub.101 where R.sub.101 is hydrogen, lower alkyl, arylalkyl, alkenyl, hydroxy or alkoxy, with the proviso that when R.sub.4 is >O or >S then R.sub.1, R.sub.2 and R.sub.3 taken together represent a group selected from the group consisting of lower alkyl, arylalkyl, aryl or hydrogen;
- (e) R.sub.5 is selected from the group consisting of >CR.sub.201 R.sub.202, >NR.sub.203, >C.dbd.CR.sub.205 R.sub.206, existing in either the Z- or E-configuration, and substituted cyclopropyl of the formula ##STR27## (f) R.sub.6, R.sub.9, R.sub.12, R.sub.15, R.sub.18, R.sub.21, and R.sub.24 are independently selected from the group consisting of >C.dbd.O, >CH.sub.2, --CH.sub.2 C(O)--, --NHC(O), >C.dbd.S, >SO.sub.2, and >P(O)X where X is selected from the group consisting of hydroxy, alkoxy, amino, alkylamino and ialkylamino;
- (g) R.sub.7, R.sub.10, R.sub.13, R.sub.16, R.sub.19, R.sub.22, R.sub.25 are independently selected from the group consisting of >CH.sub.2 and >NR.sub.50 where R.sub.50 is selected from the group consisting of hydrogen, lower alkyl, arylalkyl, aryl, hydroxy and alkoxy;
- (h) R.sub.8 is selected from the group consisting of >CR.sub.210 R.sub.211, >NR.sub.213, >C.dbd.CR.sub.215 R.sub.216, existing in either the Z- or E-configuration, and substituted cyclopropyl of the formula ##STR28## (i) R.sub.11 is selected from the group consisting of >CR.sub.220 R.sub.221, >NR.sub.223, >C.dbd.CR.sub.225 R.sub.226, existing in either the Z- or E-configuration, and substituted cyclopropyl of the formula ##STR29## (j) R.sub.14 is selected from the group consisting of >CR.sub.230 R.sub.231, >NR.sub.233, >C.dbd.CR.sub.235 R.sub.236, existing in either the Z- or E-configuration, and substituted cyclopropyl of the formula ##STR30## (k) R.sub.17 is selected from the group consisting of >CR.sub.301 R.sub.302, >NR.sub.303, >C.dbd.CR.sub.305 R.sub.306, existing in either the Z- or E-configuration, and substituted cyclopropyl of the formula ##STR31## (l) R.sub.20 is selected from the group consisting of >CR.sub.310 R.sub.311, >NR.sub.313, >C.dbd.CR.sub.315 R.sub.316, existing in either the Z- or E-configuration, and substituted cyclopropyl of the formula ##STR32## (m) R.sub.23 is selected from the group consisting of >CR.sub.320 R.sub.321, >NR.sub.323, >C.dbd.CR.sub.325 R.sub.326, existing in either the Z- or E-configuration, and substituted cyclopropyl of the formula ##STR33## (n) R.sub.26 is selected from the group consisting of >CR.sub.330 R.sub.331, >C.dbd.CR.sub.335 R.sub.336, existing in either the Z- or E-configuration, and substituted cyclopropyl of the formual ##STR34## (o) R.sub.27 is selected from the group consisting of >C.dbd.O, >CH.sub.2, --CH.sub.2 C(O)--, >C.dbd.S, >SO.sub.2, and >P(O)X wherein X is selected from hydroxy, alkoxy, amino, alkylamino and dialkylamino;
- (p) R.sub.28 is selected from the group consisting of >O, >S, >CH.sub.2, and >NR.sub.109 where R.sub.109 is selected from hydrogen, lower alkyl, (heterocyclic)alkyl, and arylalkyl, with the proviso that when R.sub.27 is >SO.sub.2 or >P(O)X, then R.sub.28 is >O or >NR.sub.109 ;
- (q) R.sub.29 is selected from the group consisting of hydrogen, lower alkyl, arylalkyl, and >NR.sub.110 where R.sub.110 is selected from hydrogen, lower alkyl, aryl, and arylalkyl, with the provisos that
- (i) when R.sub.28 is >O, or >S then R.sub.29 is lower alkyl or arylalkyl, and
- (ii) when R.sub.29 is hydrogen, lower alkyl, or arylalkyl then R.sub.30 is absent;
- (r) R.sub.30 is selected from the group consisting of hydrogen, aryl, lower alkyl, and arylalkyl;
- (s) R.sub.31 is ##STR35## where m and n are integers independently selected from 0, 1 and 2; (t) R.sub.32 is ##STR36## where p and q are integers independently selected from 0, 1 and 2; (u) R.sub.33 is ##STR37## where t and v are integers independently selected from 0, 1, 2 and 3; (v) R.sub.34 is ##STR38## where r and s are integers independently selected from 0, 1, 2 and 3; (w) R.sub.35 is ##STR39## where f is and integer of 0 to 3, X is selected from the group consisting of >C.dbd.O and --CH.sub.2 -- and R is selected from the group consisting of hydrogen and lower alkyl, with the provisos that
- (i) when f is 0,X is at C-2 and R is at C-3 or C-4;
- (ii) when f is 1, X is at C-2 and R is at C-3, C-4 or C-5 and C-3,4 are saturated or unsaturated;
- (iii) when f is 2, X is at C-2, C-3 or C-4 and R is at C-2, C-3, C-4, C-5 or C-6 when the position is unoccupied by X and C-3,4 or C-4,5 are saturated or unsaturated; and

- (z) R.sub.38 is ##STR42## (aa) R.sub.39 is ##STR43## (ab) R.sub.40 is ##STR44## (ac) R.sub.41 is ##STR45## (ad) R.sub.42 is ##STR46## (ae) R.sub.43 is ##STR47## where k is an integer of from zero to two; (af) R.sub.1 is R.sub.2, taken together, optionally represent a group selected from the group consisting of aryl, heterocyclic, and hydrogen;

- (al) R.sub.27, R.sub.28, R.sub.29 and R.sub.30, taken together, optionally represent a group selected from the group consisting of hydrogen, lower alkyl, aryl, and arylalkyl;

- (au) R.sub.320 and R.sub.323 are selected from the group consisting of hydrogen, lower alkyl, alkenyl, aryl, benzyl, (cycloalkyl)alkyl, -(alkylene)-C(O)NR.sub.340 R.sub.341, -(alkylene)-NR.sub.342 R.sub.343, -(alkylene)-NR.sub.344 C(O)R.sub.345, hydroxyalkyl, guanidinoalkyl, carboxyalkyl, (carboxyhydrazino)alkyl, ureidoalkyl, heterocyclic substituted methyl, (thioalkoxy)alkyl, sulfhydraylalkyl, (aminothioalkoxy)alkyl, protected sulfhydrylalkyl, and halosubstituted alkyl, where R.sub.340, R.sub.341, R.sub.342, and R.sub.343 are independently selected from hydrogen and lower alkyl; and R.sub.344 and R.sub.345 are independently selected from the group consisting of hydrogen, lower alkyl, and halosubstituted lower alkyl, with the proviso that R.sub.323 may not be a vinyl group or have a heteroatom directly attached to the nitrogen or separated from it by one methylene unit;
- (av) R.sub.325 and R.sub.325 are independently selected from the group consisting of hydrogen, lower alkyl, aryl, (cycloalkyl)alkyl, -(alkylene)-NR.sub.344 C(O)R.sub.345, (carboxyamido)alkyl, ureidoalkyl, (heterocyclic)alkyl, and halosubstituted alkyl, where R.sub.344 and R.sub.345 are as defined above;
- (aw) R.sub.201 and R.sub.202, R.sub.210 and R.sub.211, R.sub.220 and R.sub.221, R.sub.230 and R.sub.231, R.sub.301 and R.sub.302, R.sub.310 and R.sub.311, R.sub.320 and R.sub.321, and R.sub.330 and R.sub.331, each pair taken together, independently may optionally represent --(CH.sub.2).sub.z -- where z is an integer of from 2 to 6;
- all of the foregoing with the provisos that
- (i) when more than one sulfhydrylalkyl is present in the compound, the compound exists in the oxidized disulfide form producing a cyclic molecule, or the two sulfhydryl moieties are connected by a C.sub.2 to C.sub.8 alkylene chain and
- (ii) when the compound contains a free amino group and carboxyl group, they can be cyclized to give the corresponding lactam.
- 2. A compound as defined by claim 1 wherein R.sub.4, R.sub.7, R.sub.10, R.sub.13, R.sub.16, R.sub.19, R.sub.22, and R.sub.25 are independently selected fromthe group consisting of >NH and >N-(lower alkyl).
- 3. A compound as defined by claim 1 wherein R.sub.6, R.sub.9, R.sub.12, R.sub.15, R.sub.18, R.sub.21, R.sub.24, and R.sub.27 are independently selected from the group consisting of >C.dbd.O and >CH.sub.2.
- 4. A compound as defined by claim 1 wherein R.sub.5 is selected from the group consisting of >CR.sub.201 R.sub.202 ;>NR.sub.203 ; >C.dbd.CR.sub.205 R.sub.206, existing in the Z- or E-configuration; and substituted cyclopropyl of the formula ##STR48## where R.sub.201 is selected from the group consisting of lower alkyl, alkenyl, aryl, arylalkyl, (cycloalkyl)alkyl, amidoalkyl, (carboxyamido)alkyl, (heterocyclic)alkyl, (thioalkoxy)alkyl, (thioarylalkoxy)alkyl, protected sulfhydrylalkyl, and halosubstituted alkyl;
- R.sub.202 and R.sub.205 are selected from the group consisting of hydrogen and lower alkyl;
- R.sub.203 is selected from the group consisting of lower alkyl, alkenyl, arylalyl, (cycloalkyl)alkyl, amidoalkyl, (carboxyamido)alkyl, (heterocyclic)alkyl, (thioalkoxy)alkyl, (thioarylalkoxy)alkyl, protected sulfhydrylalkyl, with the proviso that R.sub.203 may not be a vinyl group or have a heteroatom directly attached to the nitrogen or separated from it by one methylene group; and
- R.sub.206 is selected fromt he group consisting of lower alkyl; aryl; arylalkyl; (cycloalkyl)alkyl; amidoalkyl; (carboxyamido)alkyl; (heterocyclic)alkyl; and halosubstituted alkyl.
- 5. A compound as defined in claim 1 wherein R.sub.8 is selected from the group consisting of >CR.sub.210 R.sub.211 ; >NR.sub.213 ; >C.dbd.CR.sub.215 R.sub.216, existing in either the Z- of E configuration; and substituted cyclopropyl of the formula ##STR49## where R.sub.210 is selected from the group consisting of arylalkyl; aminoalkyl; guanidinoalkyl; (heterocyclic)alkyl; (aminothioalkoxy)alkyl;
- R.sub.211 and R.sub.215 selected from hydrogen and lower alkyl;
- R.sub.213 is selected from the group consisting of arylalkyl; aminoalkyl; guanidinoalkyl; (heterocyclic)alkyl; and (aminothioalkoxy)alkyl; with the proviso that R.sub.213 may not have a herteroatom directly attached to the nitrogen or separated from it by one methylene unit; and
- R.sub.216 is selected from the group consisting of arylalkyl and (heterocyclic)alkyl.
- 6. A compound as defined by claim 1 wherein R.sub.26 is selected from the group consisting of >CR.sub.330 R.sub.331 ; >C.dbd.CR.sub.335 R.sub.356, existing in either the Z- or E-configuration; and substituted cyclopropyl of the formula ##STR50## where R.sub.330 is selected from the group consisting of arylalkyl, aminoalkyl, guanidinoalkyl, (heterocyclic)alkyl and (aminothioalkoxy)alkyl;
- R.sub.331 and R.sub.335 are independently selected from the group consisting of hydrogen and lower alkyl; and
- R.sub.336 is selected from the group consisting of arylalkyl and (heterocyclic)alkyl.
