Claims
- 1. A method of synthesizing diverse compounds on a plurality of substrates comprising the steps of:
- a) in each of a first plurality of reaction vessels, providing a plurality of substrates;
- b) delivering through a pressurized delivery means a first compound to each of said reaction vessels wherein said delivery means is coupled to each of said reaction vessels and to a mixing vessel and can deliver selected reagents to said mixing vessel or to at least one of said plurality of reaction vessels and wherein the first compound in one of said reaction vessels is different from the first compound in selected other reaction vessels;
- c) in each of said reaction vessels, coupling said first compound to said substrates to produce an immobilized compound;
- d) isolating said immobilized compounds in each of said reaction vessels by introducing a pressurized medium into each of said reaction vessels and allowing any fluids contained within the vessel to drain from each of said vessels; and
- e) transferring said immobilized compounds through a pressurized transfer means to said mixing vessel, said transfer means comprising a common manifold connected to said mixing vessel and a plurality of flow lines connecting said plurality of reaction vessels to said common manifold, wherein said transfer means is coupled to each of said reaction vessels and to said mixing vessel
- f) mixing said immobilized compounds;
- g) redistributing said immobilized compounds through said transfer means from said mixing vessel back to said plurality of reaction vessels;
- h) delivering through said pressurized delivery means a second compound to each of said reaction vessels; and
- i) coupling said second compound to said immobilized compounds.
- 2. The method of claim 1, wherein said mixing vessel, said reaction vessels, and said delivery means are sealed from the atmosphere.
- 3. The method of claim 1, wherein said transfer means further comprises a valve for controlling the transfer of said solid supports between said mixing vessel and said plurality of reaction vessels, said valve being the only valve through which solid supports traverse during the shift between said mixing vessel and said plurality of reaction vessels.
- 4. The method of claim 1, wherein after said first compound is coupled to said substrates, a first molecular tag is delivered through said delivery means to each of said reaction vessels, wherein said tag identifies the compound bound to said substrate or records a step in a synthesis of said compound.
- 5. The method of claim 4, wherein said tag has a recognizable feature due to a physical property of said tag, said physical property selected from the group consisting of size, shape, color, optical density, differently absorbing or emitting of light, magnetic or electronic properties; and chemical reactivity.
- 6. The method of claim 5, wherein said tag comprises a fluorophore.
- 7. The method of claim 5, wherein said tag is a nucleotide or an oligonucleotide.
- 8. The method of claim 4, wherein said first molecular tag is coupled to a preexisting tag.
- 9. The method of claim 4, wherein said preexisting tag is coupled to said substrate.
- 10. The method of claim 4, wherein said molecular tag is coupled to said solid support and not to another molecular tag.
- 11. The method of claim 4, wherein said molecular tag is coupled to a reactive group of said compound.
- 12. The method of claim 1, wherein said substrate is a bead or particle.
- 13. The method of claim 1, wherein said substrate is a nonporous bead.
- 14. The method of claim 1, wherein said substrate is a bead with a diameter in the range of 1 .mu.m to 1 mm.
