Claims
- 1. A method of assaying for the presence of .beta.-lactamase in a sample comprising the steps of:
- a) contacting the sample with a fluorogenic cephalosporin derivative having a group --CH.sub.2 --X--Y at position 3 of the cephalosporin, wherein X is ##STR7## and Y is selected from the group consisting of: ##STR8## 1-naphthol, 2-naphthol, naphthylamine, fluorescein, 4-methyl umbelliferone, 1-methyl-7-hydroxyquinolinium iodide and rhodamine,
- whereby X--Y is eliminated from the fluorogenic cephalosporin derivative in the presence of .beta.-lactamase and X--Y breaks down to X and Y, and;
- b) exciting Y in the presence of light and detecting emitted fluorescence as an indicator of the presence of .beta.-lactamase.
- 2. The method according to claim 1 wherein the sample is contacted with a fluorogenic cephalosporin derivative selected from the group consisting of cephalothin aminomethylcoumarin carbamate and cefotaxime aminomethylcoumarin carbamate.
- 3. A method of assaying for the presence of .beta.-lactamase in a sample comprising the steps of:
- a) contacting the sample with a chromogenic cephalosporin derivative having a group --CH.sub.2 --X--Y at position 3 of the cephalosporin, wherein X is ##STR9## and Y is selected from the group consisting of: ##STR10## N-methyl indoxyl, p-nitrophenol and o-nitrophenol, whereby X--Y is eliminated from the chromogenic cephalosporin derivative in the presence of .beta.-Lactamase and X--Y breaks down to X and Y, and;
- reacting Y with a reagent to produce a colored reaction product and detecting the colored reaction product as an indicator of the presence of .beta.-lactamase.
- 4. The method according to claim 3 wherein the sample is contacted with a chromogenic cephalosporin derivative selected from the group consisting of cephalothin aminoindole carbamate and cefotaxime aminoindole carbamate.
- 5. The method according to claims 3 or 4 wherein the chromogenic cephalosporin derivative is affixed to a solid surface and the colored product is deposited on the solid surface.
- 6. The method according to claims 3 or 4 wherein a diazonium salt is added to produce a colored reaction product.
- 7. The method according to claims 3 or 4 wherein nitroblue tetrazolium is added to produce a colored reaction product.
- 8. A kit for determining the presence of .beta.-lactamase in a sample comprising, in a first containing means, a cephalosporin derivative having a group --CH.sub.2 --X--Y at position 3 of the cephalosporin, wherein X is ##STR11## and Y is selected from the group consisting of: ##STR12## 1-naphthol, 2-naphthol, naphthylamine, fluorescein, N-methyl indoxyl, 4-methyl umbelliferone, 1-methyl-7-hydroxyquinolinium iodide, rhodamine, p-nitrophenol and o-nitrophenol; and in a second containing means, a .beta.-lactamase reagent suitable for standardization of an assay for .beta.-lactamase.
- 9. The kit according to claim 8 wherein the cephalosporin derivative is chromogenic.
- 10. The kit according to claim 9 wherein the chromogenic cephalosporin derivative is selected from the group consisting of cephalothin aminoindole carbamate and cefotaxime aminoindole carbamate.
- 11. The kit according to claims 9 or 10 further comprising, in a third containing means, a reagent selected from the group consisting of a diazonium salt and nitroblue tetrazolium.
- 12. The kit according to claim 8 wherein the cephalosporin derivative is fluorogenic.
- 13. The kit according to claim 12 wherein the fluorogenic cephalosporin derivative is selected from the group consisting of cephalothin aminomethylcoumarin carbamate and cefotaxime aminomethylcoumarin carbamate.
