Claims
- 1. A purified protein having ELF3 protein biological activity, and comprising an amino acid sequence selected from the group consisting of:
(a) the amino acid sequence shown in SEQ ID NO: 2 or SEQ ID NO: 13; (b) amino acid sequences that differ from SEQ ID NO: 2 or SEQ ID NO: 13 by one or more conservative amino acid substitutions; and (c) amino acid sequences having at least 91% sequence identity to the sequences specified in (a) or (b).
- 2. A nucleic acid molecule encoding the protein according to claim 1.
- 3. The nucleic acid molecule according to claim 2 wherein the nucleic acid molecule comprises a nucleotide sequence selected from the group consisting of:
(a) SEQ ID NO: 1; (b) SEQ ID NO: 3; (c) SEQ ID NO: 4; and (d) SEQ ID NO: 12.
- 4. A recombinant nucleic acid molecule comprising a promoter sequence operably linked to the nucleic acid sequence according to claim 2.
- 5. A cell transformed with the recombinant nucleic acid molecule according to claim 4.
- 6. A transgenic plant comprising the recombinant nucleic acid molecule according to claim 4.
- 7. The transgenic plant according to claim 6 wherein the plant is selected from the group consisting of: Arabidopsis, Cardamine, Medicago, Mimulus, Xanthia, pepper, tomato, carrot, tobacco, broccoli, cauliflower, cabbage, canola, bean, pea, soybean, rice, corn, wheat, barley, flax, citrus, cotton, cassava, walnut, conifers, and ornamental plants.
- 8. An oligonucleotide comprising a sequence selected from the group consisting of:
(a) at least 15 contiguous nucleotides of nucleotides 1-1135 or 2502-2518 of the sequence shown in SEQ ID NO: 1; (b) at least 15 contiguous nucleotides of nucleotides 1-1639 of the sequence shown in SEQ ID NO: 3; and (c) at least 15 contiguous nucleotides of nucleotides 1-852 of the sequence shown in SEQ ID NO: 4.
- 9. An isolated nucleic acid molecule comprising a sequence selected from the group consisting of:
(a) at least 20 contiguous nucleotides of the sequence shown in SEQ ID NO: 1; (b) at least 20 contiguous nucleotides of the sequence shown in SEQ ID NO: 3; and (c) at least 20 contiguous nucleotides of the sequence shown in SEQ ID NO: 4.
- 10. A recombinant nucleic acid molecule comprising a promoter sequence operably linked to the nucleic acid sequence according to claim 9.
- 11. The recombinant nucleic acid molecule according to claim 10 wherein the nucleic acid sequence is in antisense orientation relative to the promoter sequence.
- 12. A cell transformed with the recombinant nucleic acid molecule according to claim 10.
- 13. A transgenic plant comprising the recombinant nucleic acid molecule according to claim 10.
- 14. The transgenic plant according to claim 13 wherein the plant is selected from the group consisting of: Arabidopsis, Cardamine, Medicago, Mimulus, Xanthia, pepper, tomato, carrot, tobacco, broccoli, cauliflower, cabbage, canola, bean, pea, soybean, rice, corn, wheat, barley, flax, citrus, cotton, cassava, walnut, conifers, and ornamental plants.
- 15. An isolated nucleic acid molecule that:
(a) hybridizes with a nucleic acid probe comprising the sequence shown in SEQ ID NO: 1 or SEQ ID NO: 12 under wash conditions of 55° C., 0.2× SSC and 0.1% SDS for 1 hour; and (b) encodes a protein having ELF3 protein biological activity.
- 16. A recombinant nucleic acid molecule comprising a promoter sequence operably linked to the nucleic acid sequence according to claim 15.
- 17. A cell transformed with the recombinant nucleic acid molecule according to claim 16.
- 18. A transgenic plant comprising the recombinant nucleic acid molecule according to claim 16.
- 19. The transgenic plant according to claim 18 wherein the plant is selected from the group consisting of: Arabidopsis, Cardamine, Medicago, Mimulus, Xanthia, pepper, tomato, carrot, tobacco, broccoli, cauliflower, cabbage, canola, bean, pea, soybean, rice, corn, wheat, barley, flax, citrus, cotton, cassava, walnut, conifers, and ornamental plants.
- 20. An isolated nucleic acid molecule that:
(a) hybridizes with a nucleic acid probe comprising the sequence shown in SEQ ID NO: 1 or SEQ ID NO: 12 under wash conditions of 50° C., 2× SSC, 0.1% for 3 hours; and (b) encodes a protein having ELF3 protein biological activity.
- 21. A recombinant nucleic acid molecule comprising a promoter sequence operably linked to the nucleic acid sequence according to claim 20.
- 22. A cell transformed with the recombinant nucleic acid molecule according to claim 21.
- 23. A transgenic plant comprising the recombinant nucleic acid molecule according to claim 21.
- 24. The transgenic plant according to claim 23 wherein the plant is selected from the group consisting of: Arabidopsis, Cardamine, Medicago, Mimulus, Xanthia, pepper, tomato, carrot, tobacco, broccoli, cauliflower, cabbage, canola, bean, pea, soybean, rice, corn, wheat, barley, flax, citrus, cotton, cassava, walnut, conifers, and ornamental plants.
- 25. An isolated nucleic acid molecule encoding a plant ELF3 protein.
- 26. A recombinant nucleic acid molecule comprising a promoter sequence operably linked to the nucleic acid sequence according to claim 25.
- 27. A cell transformed with the recombinant nucleic acid molecule according to claim 26.
- 28. A transgenic plant comprising the recombinant nucleic acid molecule according to claim 26.
- 29. The transgenic plant according to claim 27 wherein the plant is selected from the group consisting of: Arabidopsis, Cardamine, Medicago, Mimulus, Xanthia, pepper, tomato, carrot, tobacco, broccoli, cauliflower, cabbage, canola, bean, pea, soybean, rice, corn, wheat, barley, flax, citrus, cotton, cassava, walnut, conifers, and ornamental plants.
- 30. A recombinant nucleic acid molecule comprising a promoter operably linked to an open reading frame, wherein the promoter is an ELF3 promoter.
- 31. The nucleic acid molecule of claim 30 wherein the promoter comprises a sequence selected from the group consisting of:
(a) SEQ ID NO: 5; (b) nucleotides 500-4071 of SEQ ID NO: 5; (c) nucleotides 1000-4071 of SEQ ID NO: 5; (d) nucleotides 1500-4071 of SEQ ID NO: 5; (e) nucleotides 2000-4071 of SEQ ID NO: 5; (f) nucleotides 2500-4071 of SEQ ID NO: 5; (g) nucleotides 3500-4071 of SEQ ID NO: 5; and (h) nucleotides 4000-4071 of SEQ ID NO: 5.
- 32. A method of modifying the level of expression of an ELF3 protein in a plant, the method comprising expressing in the plant a recombinant genetic construct comprising a promoter operably linked to a nucleic acid molecule, wherein the nucleic acid molecule comprises at least 20 consecutive nucleotides of the sequence shown in SEQ ID NO: 4.
- 33. The method of claim 32 wherein the nucleic acid molecule is in antisense orientation relative to the promoter.
- 34. A method of modifying at least one circadian response of a plant, the method comprising expressing in the plant a recombinant genetic construct comprising a promoter operably linked to a nucleic acid molecule, wherein the nucleic acid molecule comprises at least 20 consecutive nucleotides of the sequence shown in SEQ ID NO: 4, SEQ ID NO: 12, SEQ ID NO: 15, SEQ ID NO: 18, SEQ ID NO: 20, SEQ ID NO: 21, SEQ ID NO: 22, SEQ ID NO: 24, SEQ ID NO: 26, SEQ ID NO: 28, SEQ ID NO: 30, SEQ ID NO: 32, SEQ ID NO:34, SEQ ID NO: 36, SEQ ID NO: 38, SEQ ID NO: 40, SEQ ID NO: 42, SEQ ID NO: 44, SEQ ID NO: 46, SEQ ID NO: 48, SEQ ID NO: 50, SEQ ID NO: 52, SEQ ID NO: 54, SEQ ID NO: 56, SEQ ID NO: 58, SEQ ID NO: 60, or SEQ ID NO: 62.
- 35. The method of claim 34 wherein the circadian response is selected from the group consisting of gene transcription, leaf movement, photosynthetic ability, stomatal opening, hypocotyl elongation, and photoperiodic control of flowering.
- 36. The method of claim 34, wherein the plant is selected from the group consisting of: Arabidopsis, Cardamine, Medicago, Mimulus, Xanthia, pepper, tomato, carrot, tobacco, broccoli, cauliflower, cabbage, canola, bean, pea, soybean, rice, corn, wheat, barley, flax, citrus, cotton, cassava, walnut, conifers, and ornamental plants.
- 37. A method of modifying at least one circadian response of a plant, the method comprising expressing in the plant a recombinant genetic construct encoding at least one polypeptide comprising an Essence of ELF3 Consensus (EEC) region.
- 38. The method of claim 35, wherein the EEC region is depicted in FIG. 2.
- 39. The method of claim 37 wherein the circadian response is selected from the group consisting of gene transcription, leaf movement, photosynthetic ability, stomatal opening, hypocotyl elongation, and photoperiodic control of flowering.
- 40. The method of claim 37, wherein the plant is selected from the group consisting of: Arabidopsis, Cardamine, Medicago, Mimulus, Xanthia, pepper, tomato, carrot, tobacco, broccoli, cauliflower, cabbage, canola, bean, pea, soybean, rice, corn, wheat, barley, flax, citrus, cotton, cassava, walnut, conifers, and ornamental plants.
- 41. A plant modified by the method of claim 37.
