Claims
- 1. A compound of formula I whereinR1 is selected from the group consisting of: (a) —OSO3H, (b) —OCH(COOR5)2, (c) —OCH2(COOR5), (d) —OCH(COOR5)CH2COOR5, (e) —OC(COOR5)═CHCOOR5, (f) —CH2CH(COOR5)2, (g) —CH═C(COOR5)2, (h) —OCH2CONHOH, (i) —N(CH2COOR5)2, and (j) —OCHF(COOR5); G1 is selected from the group consisting of: (a) —C1-C10 alkyl, optionally substituted by 1 or 2 —COOR5 bonded to the same or different carbon atoms; or with —CONH2, (b) —C3-C8 cycloalkyl, optionally substituted by one —COOR5, (c) —C0-C6 alkyl-phenyl, optionally substituted by 1 or 2 —COOR5 bonded to the same or different carbon atom, or by —CH2CH(COOR5)2, (d) —CH(R7)—NH—X—R6, and G2 is CONHR3; R3 is selected from the group consisting of: (a) —C1-C12 alkyl, optionally substituted by 1-3 O, S, N, —O—C1-C4 alkyl, —S—C1-C4 alkyl, —O—G3, —S—G3, or OH, (b) —C1-C4 alkyl-C3-C6 cycloalkyl, (c) —C2-C12 alkenyl, (d) —C3-C12 alkynyl, (e) —C0-C10 alkyl (G3)n, wherein alkyl is optionally substituted by 1-3 of O, S, or N, and (f) —CH(CONH2)C1-C12 alkyl; R5 is selected from the group consisting of: (a) hydrogen, (b) —C1-C10 alkyl, and (c) —C1-C5 alkyl-phenyl; R6 is selected from the group consisting of: (a) —C1-C10 alkyl, (b) —C0-C6 alkyl-G3, (c) —C1-C6 alkyl-CONH2, (d) —C1-C6 alkyl-NH—COOR5, (e) —C1-C6 alkyl-OR5, (f) —C1-C6 alkyl-NHSO2Me, (g) —C1-C6 alkyl-O—G3, (h) —C1-C6 alkyl-S—G3, and (i) —C1-C6 alkyl-COOR5; R7 is selected from the group consisting of: (a) hydrogen, (b) —C1-C6 alkyl-G3, (c) —C1-C6 alkyl-COOR5, (d) —C1-C6 alkyl-CONH2, (e) —C1-C6 alkyl-NH—COOR5, (f) —C1-C10 alkyl, (g) —C1-C10 cycloalkyl, (h) —C1-C6 alkyl-S—R5, and (i) —C1-C6 alkyl-S(═O)R5; G3 is selected from the group consisting of: (a) phenyl substituted by 0-3 R9, (b) naphthyl substituted by 0-3 R9, and (c) het substituted by 0-3 R9; het is a 5- or 6-membered saturated or unsaturated ring containing from 1-4 heteroatoms selected from the group consisting of N, O and S, and including any bicyclic group in which any of the above heterocyclic rings is fused to a benzene ring, C3-C8 cycloalkyl, or another hetercycle, and if chemically feasible, the nitrogen and sulfur atoms may be in the oxidized forms; R9 is selected from the group consisting of: (a) C1-C8 alkyl substituted by 0-3 halo, (b) C2-C8 alkenyl, (c) OH, (d) —O—C1-C5 alkyl, (e) —O—C0-C5 alkyl-phenyl, (f) —(CH2)n—O—C1-C5 alkyl substituted by 0-3 hydroxy, (g) halo, (h) NH2, (i) amino-C1-C5 alkyl, (j) mono- or di-C1-C5 alkylamino, (k) —C(O)—C1-C5 alkyl, (l) —CHO, (m) —C(O)—C0-C5 alkyl-phenyl, (n) —COOR5, (o) —CON (R5)2 (p) —C3-C7 cycloalkyl, (q) —NO2 (r) —CN, (S) —SO3H, (t) —SO2N(R5)2, (u) —O((CH2)2—O)n—CH3, (v) —(CH2—O)n—C1-C3 alkyl, (w) —NR5(CO)—NR5, (x) —CF3, (y) —NR5(CO)C1-C5 alkyl, (z) —N(R5)—SO2—R5, (a1) —O—C(O)—R5, (b1) —S(O)—R5, (c1) —SR5, and (d1) —SO2—R5; R10 is selected from the group consisting of: (a) hydrogen, (b) CO2R57, (c) CONHOH, (d) 5-tetrazolyl, (e) F, and (f) —OCH2CO2R5; R11 is selected from the group consisting of: (a) hydrogen, and (b) methyl; X is CO, SO2, or CO2; and n is 0, 1, 2, or 3, or a pharmaceutically acceptable salt thereof, provided that when R10 is H, R1 is other than —OCH2(CO2R5).
- 2. The compound of claim 1, of formula III or IV wherein G1, R3 and R5 are as defined in claim 1 and the chiral center depicted in III is in the (S) configuration.
- 3. The compound of claim 2 wherein G1 isa) —CH(CH2phenyl)NHCO2t-Bu, b) —CH(CH2phenyl)NHCOC1-C3 alkyl-G3, c) —CH(CH2phenyl)NHCOC1-C3 alkyl-CO2R5 d) wherein R3 isa) —C5-C6 alkyl, or b) —C3-C6 alkyl-phenyl; wherein R5 is —H;wherein the configuration of the chiral center(s) is (S).

- 5. A method for treating noninsulin-dependent diabetes mellitus in a patient in need of such treatment, comprising:administering said compound according to claim 2 or a pharmaceutically acceptable salt thereof to said patient.
- 6. The method according to claim 1, wherein said compound or a pharmaceutically acceptable salt thereof is administered orally, intranasally, transdermally, subcutaneously or parenterally.
- 7. The compound according to claim 1, wherein R1 is —OCH(COOR5)2, and wherein R5 is the same as defined above.
- 8. The compound according to claim 1, said compound having the formula III or IV: wherein G1, R3 and R5 are the same as defined above.
- 9. The compound according to claim 7, wherein R1 is —OCH(COOH)2.
- 10. The compound according to claim 9, having the formula wherein R3 is the same as defined above.
- 11. The compound according to claim 9, having the formula B-5: wherein G1 is the same as defined above.
- 12. The compound according to claim 1, wherein said compound exists in (S)-configuration.
- 13. The compound according to claim 1, wherein the pharmaceutically acceptable salt of formula I is selected from the group consisting of lithium, sodium and potassium.
- 14. A pharmaceutical composition comprising:a compound according to claim 1 as an active ingredient; and a pharmaceutically and pharmacologically acceptable excipient.
- 15. A method for treating noninsulin-dependent diabetes mellitus in a patient in need of such treatment, comprising:administering a compound according to claim 1 to said patient.
- 16. The method according to claim 15, wherein said compound or pharmaceutically acceptable salt thereof is administered orally, intranasally, transdermally, subcutaneously or parenterally.
- 17. The method according to claim 15, wherein said compound or pharmaceutically acceptable salt thereof is administered in a dose of 0.1 to 1000 mg/kg body weight per day.
- 18. The method according to claim 17, wherein said compound or pharmaceutically acceptable salt thereof is administered in a dose of 0.1 to 100 mg/kg body weight per day.
- 19. The method according to claim 16, wherein said compound or pharmaceutically acceptable salt thereof is administered orally.
- 20. The method according to claim 16, wherein said compound or pharmaceutically acceptable salt thereof is administered parenterally.
- 21. A method for treating noninsulin-dependent diabetes mellitus in a patient in need of such treatment, comprising:administering a compound according to claim 8 or a pharmaceutically acceptable salt thereof to said patient.
- 22. The method according to claim 21, wherein said compound or a pharmaceutically acceptable salt thereof is administered orally, intranasally, transdermally, subcutaneously or parenterally.
- 23. The method according to claim 21, wherein said compound or pharmaceutically acceptable salt thereof is administered in a dose of 0.1 to 1000 mg/kg body weight per day.
- 24. The method according to claim 23, wherein said compound or pharmaceutically acceptable salt thereof is administered in a dose of 0.1 to 100 mg/kg body weight per day.
- 25. The method according to claim 22, wherein said compound or pharmaceutically acceptable salt thereof is administered orally.
- 26. The method according to claim 22, wherein said compound or pharmaceutically acceptable salt thereof is administered parenterally.
